The aim of our investigation was to make a comparative assessment of the biological effects of silver nanoparticles encapsulated in a natural and synthetic polymer matrix. We carried out a comparative assessment of the biological effect of silver nanocomposites on natural (arabinogalactan) and synthetic (poly-1-vinyl-1,2,4-triazole) matrices. We used 144 three-month-old white outbred male rats, which were divided into six groups. Substances were administered orally for 9 days at a dose 500 μg/kg. Twelve rats from each group were withdrawn from the experiment immediately after nine days of exposure (early period), and the remaining 12 rats were withdrawn from the experiment 6 months after the end of the nine-day exposure (long-term period). We investigated the parietal–temporal area of the cerebral cortex using histological (morphological assessments of nervous tissue), electron microscopic (calculation of mitochondrial areas and assessment of the quality of the cell nucleus), and immunohistochemical methods (study of the expression of proteins regulating apoptosis bcl-2 and caspase 3). We found that the effect of the nanocomposite on the arabinogalactan matrix causes a disturbance in the nervous tissue structure, an increase in the area of mitochondria, a disturbance of the structure of nerve cells, and activation of the process of apoptosis.

The aim was to assess the effect of silver nanoparticles on the expression of proand anti-apoptotic proteins caspase-3 and bcl-2 by discriminant analysis.Materials and methods. 120 sexually mature outbred male rats were divided into 8 groups (pure arabinogalactan (AG), pure poly-1-vinyl-1,2,4-triazol (PVT), nanobiocomposites on the AG and PVT matrix at a dose of 100 and 500 μg/kg. The administration was done orally for 9 days. The experimental study was carried out in two stages. The first stage included the examination of animals immediately after the end of the exposure of the studied substances (early period), the second stage – a survey 6 months after the end of the exposure (the long-term period).Results. The most distinguished groups were the groups that were administered silver nanoparticles on the AG matrix at a dose of 100 and 500 μg/kg.Conclusion. Comparison of the results of the discriminant analysis allowed to fully determine the effect of silver nanoparticles on the expression of proand anti-apoptotic proteins caspase-3 and bcl-2 when they were introduced on the AG and PVT matrix. In the AG groups and nanobiocomposites based on it, in the feature space, it was shown that the most remote by differentiating characteristics are the groups nAG100 and nAG500. In nanocomposites on the PVT matrix, a group of animals differing in differentiating features has not been identified.

Renal hypertrophy was induced in 100 g male rats by the injection of 250 mg folic acid (FA) dissolved in 0.3 M NaHCO3/kg body weight (i.v.). Preliminary studies of the biochemical alterations in ribonucleic acid (RNA) metabolism of the renal tissue have been reported recently (1). They are: RNA content and concentration, orotic acid-c14 incorporation into RNA and acid soluble nucleotide pool, intracellular localization of the newly synthesized RNA, and the specific activity of enzymes of the de novo pyrimidine biosynthesis pathway. The present report describes the light and electron microscopic observations in these animals. For light microscopy, kidney slices were fixed in formalin, embedded, sectioned, and stained with H & E and PAS.

In 1930, Moore, Hughes and Gallager reported that after castration seminal vesicle epithelial cell atrophy occurred and that cell regeneration could be achieved with daily injections of testis extract. Electron microscopic studies have confirmed those observations and have shown that testosterone injections restore the epithelium of the seminal vesicle in adult castrated male rats. Studies concerned with the metabolism of androgens point out that dihydrotestosterone stimulates cell proliferation and that other metabolites of testosterone probably influence secretory function in certain target cells.Although the influence of androgens on adult seminal vesicle epithelial cytology is well documented, little is known of the effect of androgen depletion and replacement on those cells in aging animals. The present study is concerned with the effect of castration and testosterone injection on the epithelium of the seminal vesicle of aging rats.

Dimethylmethylphosphonate (DMMP) is a simple organophosphate used industrially as a flame retardant and to lower viscosity in polyester and epoxy resins. The military considered the use of DMMP as a nerve gas simulant. Since military use of DMMP involved exposure by inhalation, there was a need for a subchronic inhalation exposure to DMMP to fully investigate its toxic potential.Male Fischer-344 rats were exposed to 25 ppm or 250 ppm DMMP vapor on a continuous basis for 90 days. An equal number of control rats were sham-exposed. Following the 90-day continuous exposure period, 15 male rats were sacrificed from each group. Two rats from each group had the left kidney perfused for electron microscopic examination. The kidneys were perfused from a height of 150 cm water with 1% glutaraldehyde in Sorensen's 0.1M phosphate buffer pH 7.2. An additional kidney was taken from a rat in each group and fixed by immersion in 2.5% glutaraldehyde and 2% paraformaldehyde in 0.1M cacodylate buffer pH 7.4. A portion of the 9 kidneys collected for electron microscopy were processed into Epon 812. Thin sections, stained with uranyl acetate and lead citrate, were examined with a JEOL 100B Transmission Electron Microscope. Microvilli height was measured on photographs of the cells of proximal tubules. This data, along with morphologic features of the cells, allows the proximal convoluted tubules (PCT) to be identified as being S1, S2, or S3 segment PCT.

Previous work has shown that mirex undergoes photolytic dechlorination to chlordecone (CD) (KeponeR) in the environment. Much work has shown that prior exposure to nontoxic levels of CD causes potentiation of hepatotoxicity and lethality of CCl4, BrCCl3 and other halomethane compounds. Potentiation of bromotrichloromethane hepatotoxicity has been associated with compounds that stimulate the activity of hepatic mixed-function oxidase (MFO). An increase in the metabolism of halomethane by the MFO to a free radical initiates peroxidative decomposition of membranal lipids ending in massive cellular injury. However, not all MFO inducers potentiate BrCCl3 hepatotoxicity. Potentiation by much larger doses of phenobarbital is minimal and th at by a more potent inducer of MFO, mirex, is negligible at low doses. We suggest that the CD and bromotrichloromethane interaction results in a depletion of cellular energy and thereby reducing the cellular ability to undergo mitosis.

Disturbances in early pregnancy immunity affect embryo development, endometrial receptivity, placental development, fetal growth and lead to subfertility, dexamethasone is a synthetic glucocorticoid used for treatment of various complications. Immune cells and cytokines were examined during the early pregnancy in twenty-four female rats and six male rats for mating. Rats were grouped into two group control and dexamethasone treated by a dose of 50µgm/kgm body weight daily starting from one week before mating and persisted for one week after pregnancy. Blood samples were collected from each rat at 5hrs and at 1,3,7 day of pregnancy. Extracted RNA was subjected to real time PCR to determine mRNA levels for immune related genes interleukin1a(IL1A) and interleukin 10(IL10). Histopathological examination was done to uterus in order to detect leukocyte infiltration in uterine tissue. Results showed that significant increase in white blood cell count mainly eosinophil at 5hrs and lymphocyte at three and seven day of pregnancy of dexamethasone treated group. Moreover, TNF, C-reactive protein and progesterone were increased mainly at seven day of pregnancy of dexamethasone treated group. Similarly, interleukin 1alpha and interleukin 10 significantly increased at 5hrs and one day of pregnancy of dexamethasone treated group. In contrast, serum levels of total antioxidant capacity and estrogen were decreased significantly at 5hrs and seven day in dexamethasone treated group. Histopathological examination of uterus revealed leukocytic infiltration especially neutrophil and few eosinophils at five hours and one day of gestation then eosinophil become absent at 3day and seven day of dexamethasone group. Epithelial height and uterine gland diameter significantly increased at 5hrs, three day and seven days of gestation of dexamethasone treated group. The present investigation demonstrated that using of dexamethasone by dose of 50µgm/kgm during early pregnancy had a conflicting impact on some immune cytokines and parameters and may reflect a harmful response of immune system toward early period of pregnancy

Статья посвящена построению прогноза эффективности реконструктивных вмешательств на магистральных нервах предплечья в зависимости от протяженности дефекта нервной ткани и особенностей последующего периода реабилитации пациентов. Данная тематика является весьма актуальной, так как с каждым годом во всем мире наблюдается определенный рост случаев травм различного генеза магистральных нервных стволов, которые затем нередко приводят к временной нетрудоспособности и даже инвалидности пациентов. Реконструктивная микрохирургия многие десятилетия пытается решить целый ряд проблем аутотрансплантации нервных стволов и повысить ее общую эффективность. Но из-за определенных проблем связанных с финансированием, некоторые вопросы трансплантологии и реабилитации остаются нерешенными и в настоящий момент. Все это придает представленной работе важное значение не только медицинского, но и социально - экономического плана. Целью работы стала попытка построения прогноза восстановительных операций на нервной ткани, с учетом объема пораженных структур и периода реабилитации. Объектами исследования стало 180 больных, которым по той или иной причине, осуществлялась реконструктивная операция на одном из магистральных нервов предплечья. Все пациенты были разделены на 3 группы по 60 человек, в зависимости от протяженности дефекта магистрального нерва: до 4 см, от 4 до 8 см и от 8 до 12 см. Последующее разделение внутри каждой группы на подгруппы производилось в зависимости от определенного поврежденного нерва (лучевой, локтевой, срединный). В работе изучалось течение раннего послеоперационного воспалительного процесса, с определением бактериальной микрофлоры в ране. Изучались и отдаленные последствия оперативного вмешательства. Своеобразной новизной для данной тематики в целом, стало выявление последующего установления инвалидности пациентов. Кроме того, важные данные были получены и по срокам реабилитации и частичного или полного восстановления утраченных функций по срокам в зависимости от размеров восстанавливаемого дефекта и от наличия или отсутствия необходимой реабилитации. Полученные в работе данные могут представлять интерес не только для врачей хирургов и травматологов, но и для организаторов здравоохранения, позволяя производить прогнозы по выздоровлению пациентов в каждой определенной клинической ситуации The article is devoted to the construction of a forecast of the effectiveness of reconstructive interventions on the main nerves of the forearm, depending on the length of the defect in the nervous tissue and the characteristics of the subsequent period of rehabilitation of patients. This topic is very relevant, since every year all over the world there is a certain increase in cases of injuries of various origins of the main nerve trunks, which then often lead to temporary disability and even disability of patients. For many decades, reconstructive microsurgery has been trying to solve a number of problems of autotransplantation of nerve trunks and improve its overall efficiency. But due to certain problems associated with funding, some issues of transplantation and rehabilitation remain unresolved at the moment. All this gives the presented work important not only medical, but also socio - economic importance. The aim of this work was to attempt to predict restorative operations on the nervous tissue, taking into account the volume of the affected structures and the period of rehabilitation. The objects of the study were 180 patients who, for one reason or another, underwent a reconstructive operation on one of the main nerves of the forearm. All patients were divided into 3 groups of 60 people, depending on the length of the main nerve defect: up to 4 cm, from 4 to 8 cm, and from 8 to 12 cm. Subsequent division within each group into subgroups was performed depending on the specific damaged nerve ( radial, ulnar, median). The work studied the course of the early postoperative inflammatory process, with the determination of bacterial microflora in the wound. The long-term consequences of surgery were also studied. A peculiar novelty for this topic as a whole was the identification of the subsequent establishment of disability in patients. In addition, important data were obtained on the timing of rehabilitation and partial or complete restoration of lost functions in terms of timing, depending on the size of the restored defect and on the presence or absence of the necessary rehabilitation. The data obtained in this work may be of interest not only for surgeons and traumatologists, but also for healthcare organizers, allowing them to make predictions about the recovery of patients in each specific clinical situation

: Thymoquinone (TQ) is one of the leading phytochemicals, which is abundantly found in Nigella sativa L. seeds. TQ exhibited various biological effects such as antioxidant, anti-inflammatory, antimicrobial, and anti-tumoral in several pre-clinical studies. Parkinson's disease (PD) is a long-term neurodegenerative disease with movement difficulties, and the common feature of neurodegeneration in PD patients is caused by dopaminergic neural damage in the substantia nigra pars compacta. The neuroprotective activity of TQ has been studied in various neurological disorders. TQ-mediated neuroprotection against PD yet to be reported in a single frame; therefore, this review is intended to narrate the potentiality of TQ in the therapy of PD. TQ has been shown to protect against neurotoxins via amelioration of neuroinflammation, oxidative stress, apoptosis, thereby protects neurodegeneration in PD models. TQ could be an emerging therapeutic intervention in PD management, but mechanistic studies have been remained to be investigated to clarify its neuroprotective role.
