The consumption of prebiotics is known to affect the balance of gut microbiota. The purpose of this study was to explore how a galactomannan-rich effervescent drink can affect the population of Lactobacillus, Bifidobacterium, E. coli, and the concentration of short-chain fatty acids in the cecum of rats. Twenty-eight male Wistar rats (aged 2 months) were divided equally into 7 groups and treated orally each day for 15 days with 2 mL effervescent drinks with increasing levels of prebiotic galactomannan. The dosage of 500 mg galactomannan increased the growth of Lactobacillus spp. and Bifidobacterium spp. with inhibition of the growth of E.coli with increased formation of short-chain fatty acids such as acetate, propionate, and butyrate in the cecum of rats.

Abstract The optimal vitamin A (VA) status of lactating women is important for mothers and their breastfed infants, especially in protecting against infectious diseases. Vitamin A fortified rice is one of the food-base intervention strategy which has the potential to improve VA status. Vitamin A and gut microbiota are interrelated in their effect on human health and immunity however no specific relationship has been proved in these groups of population. This study aimed to determine the effect of VA fortified rice on the gut microbiota changes of lactating woman-exclusively breastfed infant pairs. A double-blind, randomized controlled trial (RCT) of VA fortified rice was conducted in 70 lactating women-infants pairs for 14 weeks. Gut microbiota was measured using the fluorescent in situ hybridization (FISH) and next generation sequencing (NGS) technique. Based on the FISH technique, the numbers of Clostridium spp. /Enterobacter spp. were significantly lower (P < 0.05) in mothers fed VA-fortified rice at the end of the study. In contrast, the abundance of Bifidobacterium spp. and Lactobacillus spp. of infants whose mothers fed with VA-fortified rice was significantly higher (P < 0.05) than the control group. The NGS technique confirmed that results with the increasing of Lactobacillus, B. longum and B. Choerinum in the infant of intervention group. In conclusion, VA-fortified rice was efficacious in decreasing Clostridium spp. /Enterobacter spp. in lactating women and raising the number of Bifidobacterium spp. and Lactobacillus spp. in their breastfed infants. AbstractRecently, Akkermansia muciniphila an anaerobic member of the gut microbiota, has been proposed as a next-generation probiotic. The aim of this study was evaluation of the effect of alive and pasteurized A. muciniphila on health status, intestinal integrity, immune response, lipid metabolism, and gut microbial composition in normal-diet fed mice as well as direct effects of the bacterium on Caco-2 cell line. A total of 30 mice were distributed into three different groups, control, alive, and pasteurized A. muciniphila-treated group. After acclimation, control and treatment groups were administrated with PBS and 109 CFU/200µL of bacterial suspension for 5 weeks, respectively. Besides, Caco-2 separately exposed to alive, pasteurized A. muciniphila and PBS for 24 h. The results showed that administration of A. muciniphila leads to reduction in body, liver, and white adipose weight. Histology data revealed both treatments had no adverse effects in colon, liver, and adipose tissues as well as induced better gut structure. Moreover, biochemical parameters and inflammatory biomarkers in plasma demonstrated that pasteurized A. muciniphila had more pronounce effect. Furthermore, alive A. muciniphia had better effects on the modulation of gene expression related to fatty acid synthesis, energy homeostasis, and immune response in the liver; meanwhile, these effects in the adipose was more in the pasteurized A. muciniphila administration. More importantly, the improvement of gut health by enhancing strengthen intestinal integrity and maintaining immune homeostasis was seen in both treatments; notably, pasteurized A. muciniphila had more effective. Similarly, treatment with the pasteurized form more effectively upregulated tight junction and regulated immune response-related genes in Caco-2 cell line. Both treatments triggered the improvement of microbiota communities, particularly the alive form. Therefore, both forms of A. muciniphila could modulate lipid and immune homeostasis, improved some gut microbiota, and promoted the overall health, while all these effects were dominantly observed in pasteurized form. In conclusion, pasteurized A. muciniphila can be considered as new medical supplement to maintain health state and prevent diseases in normal mice through different mechanisms.

Abstract Objectives Converging evidence suggests probiotic supplementation can reduce cognitive and metabolic concerns for persons with gastrointestinal disorders. However, in healthy populations the impact of probiotics on these outcomes is lacking. Thus, we aimed to determine whether symptoms of depression and anxiety, memory function, cortisol concentrations, and gut microbiota composition are altered by consumption of a fermented dairy beverage containing probiotic microorganisms in healthy adults. Methods Adults (25–45 yrs, N = 26) free of physician diagnosed gastrointestinal and mental illness were enrolled in a single-blind, randomized crossover trial. Participants completed testing prior to and after 4-week consumption, with a 2–4 week washout between treatments of kefir, a dairy-based fermented beverage containing 25–30 billion colony forming units of live and active kefir cultures (including 3 Bifidobacterium spp., 6 Lactobacillus spp., Streptococcus Diacetylacti., Luconostoc Cremoris, and Staccharomyces Florentinus) or isocaloric, non-fermented dairy-based control beverage. Hippocampal-dependent relational memory was assessed using a spatial reconstruction task. Symptoms of depression and anxiety were assessed using the depression anxiety and stress subscale (DASS). Pooled 24-hour urine samples were analyzed using an enzyme-linked immunosorbent assay to determine urinary-free cortisol (UFC) concentrations. Fecal microbiota composition was assessed using 16 s rDNA sequencing. Results Post-test logistic regression analysis revealed an increase in the number of participants with Lactobacillus spp. present (b = 3.00, P &lt; 0.01) after consumption of kefir. Two-by-two repeated measure ANOVA displayed that the treatment improved performance on two metrics of relational memory (F[1, 25] = 4.54, P = 0.043, F[1, 25] = 5.50, P = 0.027). UFC and DASS scores were not significantly changed by either arm of the intervention. Conclusions These results reveal that kefir consumption increases the presence of probiotic microorganisms in the gut and improves relational memory in healthy adults. The impact of fermented dairy beverages containing probiotic microorganisms in healthy populations should be studied at a larger scale to better elucidate these outcomes.
