Dextranase, a hydrolase that specifically hydrolyzes α-1,6-glucosidic bonds, has been used in the pharmaceutical, food, and biotechnology industries. In this study, the strain of Catenovulum agarivorans MNH15 was screened from marine samples. When the temperature, initial pH, NaCl concentration, and inducer concentration were 30 °C, 8.0, 5 g/L, and 8 g/L, respectively, it yielded more dextranase. The molecular weight of the dextranase was approximately 110 kDa. The maximum enzyme activity was achieved at 40 °C and a pH of 8.0. The enzyme was stable at 30 °C and a pH of 5–9. The metal ion Sr2+ enhanced its activity, whereas NH4+, Co2+, Cu2+, and Li+ had the opposite effect. The dextranase effectively inhibited the formation of biofilm by Streptococcus mutans. Moreover, sodium fluoride, xylitol, and sodium benzoate, all used in dental care products, had no significant effect on dextranase activity. In addition, high-performance liquid chromatography (HPLC) showed that dextran was mainly hydrolyzed to glucose, maltose, and maltoheptaose. The results indicated that dextranase has high application potential in dental products such as toothpaste and mouthwash.

Condensed tannins are formed by monomers of procyanidins and prodelfinidins, where the proportion and concentration of their monomers varies according to the plant species and environmental conditions. In Lotus spp., condensed tannins prevent tympanism in ruminants that feed on them. This study aimed to evaluate the concentration of procyanidins and their monomers, catechin and epicatechin in the genotypes of Lotus L. grown in soil with different saturations of aluminum. A two-factor (genotype × Al saturation) assay was performed, where the genotypes São Gabriel, Ganador, and UFRGS (Lotus corniculatus L.); Serrano (Lotus uliginosus); and El Rincón (Lotus subbiflorus) were cultivated in soil with an Al saturation of 0-20%. The procyanidins were evaluated using high-performance liquid chromatography, which was previously validated for catechin and epicatechin. The concentration of procyanidins and the proportion of epicatechin:catechin were affected by the genotype × environment interaction. In L. corniculatus and L. subbiflorus, the concentration of procyanidin was significantly higher when they were grown in the soil with an Al saturation of 20% compared to that when they were grown in the soil with 0% Al saturation, but the opposite effect was observed in L. uliginosus. The proportion of epicatechin:catechin decreased in plants grown in soil without Al, and only the UFRGS genotype maintained a similar proportion under both the soil acidity conditions. The predominant monomer was epicatechin, which varied from 57% to 75% according to the soil in which the plants were grown.

α2-Antiplasmin (α2AP), the main physiological inhibitor of plasmin in human plasma, is a single–chain glycoprotein with a molecular weight of 67,000 consisting of about 510 amino acids and containing 13 percent carbohydrate.A tryptic digest on 400 mg of reduced, carboxymethylated and citraconylated purified α2AP was performed. Peptides were separated by combinations of ion exchange chromatography, gel filtration and high performance liquid chromatography, and sequenced using the manual Edman degradation. Some peptides were further digested in order to establish overlaps. At the time of submission of this abstract we have sequenced 7 out of the approximately 21 arginyl peptides completely (each between 3 and 21 residues) and are working on the others. At present we have about 200 residues of sequence. Here we only report the stretches of 10 amino acids or more, which may be useful to compare the structure of α2AP with that of other serine protease inhibitors.

The heterogeneity of Newfoundland winter flounder antifreeze polypeptides was analyzed by reverse phase high performance liquid chromatography and by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Seven antifreeze polypeptide components could be readily demonstrated. Five of the components were similar in molecular weight (3300) and amino acid composition. Two of the antifreeze polypeptide components were larger (4500) and contained valine. The two major components (components 6 and 8) were identical to those reported earlier from our laboratories.

Galactomannan is a biopolymer polysaccharides mostly obtained from Leguminosae plant have shown an interesting capability as drug carrier. In this study, galactomannan from the seeds ofLeucaenaleucocephalaknown locally as ‘petai belalang’ was obtained by water extraction. The galactomannan was characterized structurally by nuclear magnetic resonance (NMR) and fourier transform infrared spectroscopy (FTIR). Highly molecular weight of galactomannan, > 2.56 x 106Da, was successfully extracted from the plant. Total carbohydrate content of this polysaccharides is 74.84% and composition of galactose and mannose have been confirm by high performance liquid chromatography (HPLC) which is 36.67% and 38.17% respectively. M/G ratio was determined as 1:1. Hence, galactomannan has been successfully extracted from the seeds ofLeucaenaleucocephala.
