To clarify whether flavonoids and phenols in Allium mongolicum Regel have the effect of improving gastrointestinal function and analyze its quality, this study was designed to isolate and identify them from the aerial parts of A. mongolicum by using various chromatographic and spectrophotometric methods, a bioassay on motility of mouse isolated intestine tissue, as well as qualitative analysis using liquid chromatography/mass spectrometry (LC-MS) analysis. As a result, 31 flavonoids and phenolic acids were obtained and identified, including six new flavonoid glycosides, mongoflavonosides A1 (1), A2 (2), A3 (3), A4 (4), B1 (5), B2 (6), and four new phenolic acid glycosides, mongophenosides A1 (7), A2 (8), A3 (9), B (10). Among them, eleven flavonoids and three phenolic acids showed significant increase in the height of mouse small intestinal muscle. It was a first systematic bioactive constituents’ study for A. mongolicum on gastrointestinal tract. Furthermore, according to the retention time (tR) and the exact mass-to-charge ratio (m/z), thirty-one compounds were unambiguously identified by comparing to the standard references by using LC-MS. Then, on the basis of generalized rules of MS/MS fragmentation pattern, chromatographic behaviors, as well as biosynthetic laws of the 31 isolates, five flavonoid glycosides and one phenolic acid glycoside were tentatively speculated. On the basis of the study, a fast analysis method for flavonoids and phenolic acids in A. mongolicum was established.

Papaver nudicaule L. (Iceland poppy) is widely used for ornamental purposes. A previous study demonstrated the alleviation of lipopolysaccharide-induced inflammation mediated by P. nudicaule extract through nuclear factor-kappa B and signal transducer and activator of transcription 3 inactivation. As isoquinoline alkaloids are chemical markers and bioactive constituents of Papaver species, the present study investigated the alkaloid profile of aerial parts of five P. nudicaule cultivars with different flower colors and a P. rhoeas cropped for two years. A combination of liquid chromatography high-resolution mass spectrometry and molecular networking was used to cluster isoquinoline alkaloids in the species and highlight the possible metabolites. Aside from the 12 compounds, including rotundine, muramine, and allocryptopine, identified from Global Natural Products Social library and reported information, 46 structurally related metabolites were quantitatively investigated. Forty-two and 16 compounds were proposed for chemical profiles of P. nudicaule and P. rhoeas, respectively. Some species-specific metabolites showed similar fragmentation patterns. The alkaloid abundance of P. nudicaule differed depending on the flower color, and the possible chemical markers were proposed. These results show that molecular networking-guided dereplication allows investigation of unidentified metabolites. The derived chemical profile may facilitate evaluation of P. nudicaule quality for pharmacological applications.

AbstractSterols, triterpenes, volatiles, polar and other constituents in aerial parts of Carthamus lanatus were analyzed by gas chromatography-mass spectrometry. Over 90 compounds were identified most of them new for the species. Sitosterol and stigmasterol were the most abundant of 10 sterols identified in the sterol fraction. Taraxasterol, α- and β-amyrine prevailed in the triterpene fraction. Volatiles, sterols and a fraction of the dichloromethane extract showed strong cytotoxicity (Artemia salina assay).

Objective:  The present study was carried out to characterize the bioactive constituents present in aerial parts of ethanolic extract of Sarcostemma brevistigma using UV-VIS and FTIR.Methods:  FTIR spectroscopy is an established time‐saving method to characterize and identify functional groups.Results:  The UV-VIS profile showed different peaks ranging from 200‐1044 nm with different absorption respectively. UV-VIS profile showed the peaks at 254.00 and 680.00 nm for flavonoid. FTIR spectra showed the peak at 3418.85 cm-1 for N-H group. FTIR analysis and UV-VIS analysis showed the presence of flavonoids and a phenolic compound. The FTIR spectra had amply evidenced the occurrence of OH group together with the Terpenoids, and Phenol. The FT-IR spectrum showed the presence of an amine (N-H), alkyl (C-H), nitrile (C≡N), halo formyl (C=O), alkenyl (C=C), hydroxyl (O-H), haloalkane (C-F), nitrates and carbonate compounds.Conclusion:  The results confirm the fact that this plant posse’s important bioactive constituent, so further scientific investigation is needed.

Hypericum triquetrifolium and H. neurocalycinum were evaluated for their phytochemical content and in vitro bioactivity. NMR analyses were performed on the methanol extract of the aerial parts of H. triquetrifolium to establish the main classes of phytoconstituents. Then, LC-DAD-MSn analyses were performed in order to compare the composition of aerial parts and roots extracts of both Hypericum species, obtained using either methanol or water as solvents. Results, processed using multivariate data analysis, showed a significantly higher phenolic content of methanol extracts compared to water extracts, while minor qualitative differences were observed between the two. Distinctive flavonoid and PAC patterns were observed for H. triquetrifolium and H. neurocalycinum, and specific compounds were exclusively detected in one or the other species. Specifically, the phloroglucinols 7-epiclusianone, hyperfirin and hyperforin were present only in H. neurocalycinum, while hyperforin was detected only in H. triquetrifolium. Extracts were assayed using different in vitro tests to evaluate their antioxidant properties and their inhibitory activity against several enzymes, showing significant antioxidant and metal chelating activities. Furthermore, inhibitory properties against acetylcholinesterase, butyrylcholinesterase and tyrosinase were observed. Multivariate approaches were used to correlate biological data with the phytochemical composition of the different extracts. The results, showing positive correlations between specific chemical constituents and the measured bioactivities, represent preliminary data that could guide future studies aimed at isolating bioactive constituents from H. neurocalycinum and H. triquetrifolium for further pharmacological evaluations.

Objective: This study focused on the extraction of bioactive constituents in Iraqi cultivated Artemisia dracunculus L dried aerial parts and phytochemical screening with qualitative estimation of three important flavonols (kampferol, quercetin and rutin). The study also included evaluating the antitumor effects (skin cancer) of methanolic extract for this plant.Methods: Extraction of bioactive constituents was carried out according to standard protocols using 85% methanol. The phytochemical screening has been done using chemical tests and GC/MS with qualitative estimation of three flavonols (kampferol, quercetin and rutin) utilized preparative high-performance liquid chromatography (PHPLC). Skin cancer in mice was induced by using 7, 12-dimethylbenze (a) anthracene to evaluate the antitumor effect of methanolic extract of Artemisia dracunculus when administered orally via intragastric feeding tube. Body weight, tumor volume, tumor burden, percent of tumor incidence and histopathological examination were measured in all tested animals.Results: Phytochemical screening revealed the presence of glycosides, volatile oils, alkaloids, tannin, terpenoids, phenolic compounds and flavonoids. The preparative high-performance liquid chromatography (PHPLC) analysis indicated that rutin had the highest concentration followed by quercetin while kampferol had the lowest concentration. The methanolic extract of the Artemisia dracunculus significantly reduced the percent of tumor incidence, tumor volume and tumor burden of skin cancer in mice (p<0.05).Conclusion: Dried aerial parts of Iraqi Artemisia dracunculus contain many bioactive constituents including three flavonoid compounds (quercetin, kampferol and rutin) that may be responsible for chemo preventive effect exerting by methanolic extract of Artemisia dracunculus against 7, 12-dimethylbenzeanthracine-induced skin cancer in mice.
