The red macroalga Gelidium latifolium is widely distributed in the coastal areas of Indonesia. However, current knowledge on its potential biological activities is still limited. In this study, we investigated the potential bioactive compounds in Gelidium latifolium ethanol extract (GLE), and its cytotoxic effects against the murine B16-F10 melanoma cell line. GLE shows high total phenolic content (107.06 ± 17.42 mg GAE/g) and total flavonoid content (151.77 ± 3.45 mg QE/g), which potentially contribute to its potential antioxidant activity (DPPH = 650.42 ± 2.01 µg/mL; ABTS = 557.01 ± 1.94 µg/mL). ESI-HR-TOF-MS analysis revealed large absorption in the [M-H]- of 327.2339 m/z, corresponding to the monoisotopic molecular mass of brassicolene. The presence of this compound potentially contributes to GLE’s cytotoxic activity (IC50 = 84.29 ± 1.93 µg/mL). Furthermore, GLE significantly increased the number of apoptotic cells (66.83 ± 3.06%) compared to controls (18.83 ± 3.76%). Apoptosis was also confirmed by changes in the expression levels of apoptosis-related genes (i.e., p53, Bax, Bak, and Bcl2). Downregulated expression of Bcl2 indicates an intrinsic apoptotic pathway. Current results suggest that components of Gelidium latifolium should be further investigated as possible sources of novel antitumor drugs.

Abstract In this study, total phenolic, flavonoid and protein contents, antiradical capacities, antibacterial and cytotoxic properties of five different Salvia species (Salvia macrochlamys Boiss., Salvia kronenburgii Rech.f., Salvia euphratica Montbret. ex Aucher var. euphratica, Salvia huberi Hedge and Salvia kurdica Benth) were investigated. The total phenolic content of the species was in the range of 59.6 ± 20 to 150.1 ± 1.1 mg/g, total flavonoid content in the range of 20.4 ± 2.2 to 38.9 ± 4.1 mg/g, and total protein content in the range of 14.0 ± 0.5 to 22.0 ± 1.1 mg/g. Radical scavenging capacities of the Salvia species were determined in the range of 84.1 ± 4.5 to 96.8 ± 0.1%. Moreover, Salvia extracts showed powerful antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria. Cytotoxic effects of the samples on human lymphocytes were determined by MTS assay and S. kronenburgii and S. euphratica var. euphratica decreased the harmful effects of 2-hydroperoxy-2-methylpropane (tert-butyl hydroper-oxide, t-BOOH) at all time points tested. The highest amounts of rutin hydrate belong to S. kronenburgii and S. euphratica var. euphratica, and the highest amounts of luteolin-7-glucoside belong to S. huberi and S. kronenburgii. Vanillic acid was present only in S. huberi and S. kurdica.

The present study aimed to study the anti-oxidant potential of isolated flavonoids from the ethanol extract (70%v/v) of Cassia tora leaves. Three flavonoids were isolated viz Luteolin-7-O-β-glucopyranoside (I), quercetin-3-O-β-d-glucuronide (II) and Formononetin-7-O-β-D-Glucoside (III) from the ethanol extract (70%v/v) of Cassia tora leaves in the previous study. A comprehensive study on the phytochemical contents - total phenolic content and flavonoid content was accessed using Folin-Denis and AlCl3 method respectively. The antioxidant potential of the samples was evaluated using inhibition of hydroxyl radical, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and nitric oxide scavenging methods. The total phenolic content and flavonoid content was found to be 18.60 % w/w and 9.5% w/w respectively. The IC50 values of ethanol extract against hydroxyl, DPPH and nitric oxide radical were found to be 270 µg/mL, 190 µg/mL and 130 µg/mL respectively. The radical scavenging activity of the isolated flavonoids decreased in the following order: quercetin (IC50 values 15, 14, 18 µg/mL) >formononetin (IC50 values 19, 21, 14 µg/mL) > luteolin (IC50 values 20, 23, 18 µg/mL) respectively.

Phenolic and flavonoid content in ethanol extract and agro-morphological diversity of Curcuma aeruginosa accessions growing in West Java, Indonesia. Biodiversitas 20: 656-663. Curcuma aeruginosa is a rhizomatous medicinal plant with beneficial pharmacological activities. The aim of this work was to analyze the agro-morphological, extract yield, and phenolic content of ten C. aeruginosa accessions which were collected from different locations in Indonesia. Cultivation was carried out in the open field in West Java of Indonesia using a completely randomized design. Qualitative and quantitative parameters were used to investigate agro-morphological traits. Total phenolic and total flavonoids contents were determined in ethanol extracts of samples. The plants were phenotypically diverse, in which there were significant variations among the ten C. aeruginosa accessions in number of leaves, plant height, number of shoots, fresh weight of rhizome, and dry weight of rhizome characters. Variability in the total phenolic and total flavonoid contents ranged from 29.08-46.92 mg GAE/g, and 21.31-33.81 mg QE/g, respectively. Six accessions had high phenolic content and extract yield. Therefore, these accessions could be utilized for commercial scale and also showed a high potency for medicinal plant breeding programs.

The aim of this study was to elucidate the correlation between total flavonoid content and total phenolic content on antioxidants activity of ethanol extracts from three cultivars of papaya leaves: 'Holland,' 'Khak Dam' and 'Red Lady.' All crude extracts were investigated to find antioxidant capacity in DPPH radical scavenging. The result indicated that the ethanol extract of ‘Red Lady’ papaya leaves exhibited the highest level of DPPH radical scavenging activity with the IC50 of 0.18 mg/mL, followed by the ‘Khak Dam’ and ‘Holland’ papaya leaves having an IC50 value of 0.24 and 0.44 mg/mL, respectively. The ethanol extract from 'Red Lady' papaya leaves showed the highest level of total flavonoid content (TFC) of 276.72 ± 1.04 µgQE/g DW and total phenolic content (TPC) of 169.85 ± 6.54 mgGAE/g DW. All three cultivars showed a distinctive correlation between IC50 and the content of both total flavonoid and total phenolic with a negative relationship of Pearson’s correlation of -0.922 and -0.940, respectively.

Antioxidant activity, total phenolic and flavonoid content of several indigenous species of ferns in East Kalimantan, Indonesia. Biodiversitas 20: 576-580. This study aimed to determine the total phenolic and flavonoids content and antioxidant activity of ethanol extract of several indigenous species of ferns in East Kalimantan. Total phenolic content was determined by Folin-Ciocalteau method and flavonoid content was measured by colorimetric method. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. The result of photochemical screening indicated that the leaves of Plagiogyria pycnophylla, Plagiogyria glauca, and Stenochlaena palustris contained alkaloid, flavonoid, tannin, saponin, and steroid while Acrostichum aureum contained flavonoid, tannin, saponin and steroid. The result showed extract Acrostichum aureum had the highest total phenolic content (366.4573 ± 2.2117 mg GAE.g-1), flavonoid content (228.6087 ± 2.2548 mg QE.g-1), and very strong antioxidant activity with IC50 value 29.5303 ppm. There is positive correlation between total phenolic content, flavonoid with antioxidant activity.

The study investigated the effect of Musanga cecropioides (MC) water-ethanol stem bark extract on blood glucose level in both hyperglycemic loaded glucose rats and streptozotocin (STZ)-induced diabetic rats, and evaluated its antioxidant capacity. The Wistar rats were induced diabetes after fasting. Oral Glucose Tolerance Test (OGTT) was conducted on normoglycemic rats, and anti-hyperglycemic test on diabetic rats; five groups with five rats each were constituted. Group 1: negative control was treated with vehicle; Group 2, Group 3, and Group 4 were treated with increasing water-ethanol extract (200, 300 and 400 mg/kg b.w); Group 5 was the positive control, treated with glibenclamide. The antioxidant capacity of the extract was also evaluated by measuring the Ferric Reducing Antioxidant Power, Total Phenolic Content, Total Flavonoid Content, and radical scavenging activity of water-ethanol stem bark extract. In OGTT the water-ethanol extract of MC, at the dose of 300 mg/kg, significantly lowered the Area under Curve (AUC) induced by glucose. In STZ diabetic rats, the extract significantly lowered the AUC of blood glucose, at all doses. Glibenclamide was more efficient in both OGTT and anti-hyperglycemic test. The MC extract presented relevant antioxidant activity with IC50 = 6.23 mg/mL. Both the Total Phenolic Content and the Total Flavonoid Content increased in a dose-dependent manner. The correlation of DPPH % free radical scavenged and Total Flavonoid Content was positive and statistically significant. MC water-ethanol extract possesses a good antioxidant potential, and could be helpful to lower hyperglycemic state associated with diabetes.

Abstract Marine sponge Stylissa flabelliformis is a marine sponge species that is widely found in Indonesia. Marine sponges have been proven to have pharmacological activities including wound healing, cytotoxicity and antimicrobial activity. All these activities are related to its antioxidant potential. Antioxidants are important for human health due to its ability to protect cell damage caused by free radicals. The aim of this research was to study the antioxidant activity of marine sponge (Stylissa flabelliformis) and to observe the phytochemical compound of the marine sponge such as the phenolic and flavonoid content. Extraction was performed using cold maceration with 96% ethanol solvent and continued by rotary evaporator to get the concentrated extract. Antioxidants activity was analysed using DPPH (2,2-diphenyl-1-picrylhydrazyl) method, which examines the inhibitory concentration (IC50) of the ethanol extract. Total phenolic content (TPC) was examined using Gallic acid as the standard, while total flavonoid content (TFC) was examined using quercetin as the standard. The result showed a concentration-dependent antiradical activity by reduction of DPPH with an IC50 value 2000 µg/mL. Total phenolic content was 152,67 mg GAE/g, while the total flavonoid content was 155,79 mg QE/g.

This is the first study to investigate the effect of ethanol concentrations on the phenolic content and biological activities of Sripanya Punica granatum peel extract. Various concentrations of ethanol, (20 to 80% v/v), were used in the maceration process. Extracts were investigated for total phenolic content by the Folin–Ciocalteu method, antioxidant activities by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay, and anti-tyrosinase activity by a spectrophotometric method. The chemical composition was investigated using high-performance liquid chromatography. The irritation of the extracts was determined by Hen’s egg test–chorioallantoic membrane assay. The results demonstrated that E60, which was extracted using 60% v/v ethanol, had the highest yield (66.23 ± 0.38% w/w), the highest total phenolic content (490.42 ± 7.20 μg GAE/g), and the highest antioxidant and anti-tyrosinase activities. IC50 values on DPPH• and ABTS•+ inhibition were 10.97 ± 0.97 and 6.48 ± 0.06 µg/mL, respectively. The 60% v/v ethanol extract (E60) possessed strong tyrosinase inhibition with an IC50 value of 0.10 ± 0.13 µg/mL, which is more potent than kojic acid. The major constituents were punicalagin (4.05 ± 0.26% w/w) and ellagic acid (0.63 ± 0.04% w/w). E60 was safe for topical application since it did not induce irritation. Therefore, we suggest that E60 is a promising agent for further product development.

Objective: The present study qualitative phytoconstituents examine the total phenol, total flavonoid content (TFC), and antioxidant efficiencies traditionally used plant, Meyna spinosa. Methods: Chemical profiling, estimation of total phenolic content (TPC), TFC, and antioxidant activity of ethanol extracts of M. spinosa have performed by applying standard protocols. Antioxidant activity of leaf and stem was evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay. TPC and TFC of the plant were assessed using Folin–Ciocalteu colorimetric and aluminum colorimetric assay, respectively. Results: The findings of the study exhibit that ethanol extract of M. spinosa is proved to be the presence of phytoconstituents (7/9) such as alkaloids, terpenoids, saponins, tannins, phytosterols, flavonoids, and phenolic compounds, while carbohydrate, fixed oils, and fats are unavailable. In addition, phenolic compositions of ethanol extract of leaf and stem; 93.21±2.93 and 54.33±0.69 mg gallic acid equivalents/g extract, respectively; TFCs of leaf and stem have recorded as 61.55±1.21 and 37.55±1.28 mg quercetin equivalents/g extract, respectively. Antioxidant efficiency of both leaf and stem is tested using DPPH radical scavenging assay as IC50 20.68±0.32 and 50.99±0.56 μg/ml, respectively. Conclusion: From the above results, it has concluded that the ethanol extract of the M. spinosa leaves and stems seizes rich phytoconstituents which can be applied in food technology, drug industries, ethnopharmacological fields, etc., for the development of healthiness and to battle against negative health consequences.
