Fifteen soil samples (0-15cm depth) and fifteen betel leaf samples from Bara Moheshkhali, Choto Moheshkhali, Hoanak, Kalmarchora, Shaplapur union of Moheshkhali upazila, Cox’s Bazar district were collected on December-January (2020-2021). It was collected for the purpose of evaluating the color, particle density, moisture content, texture, pH, EC and organic matter content of soils and total nitrogen, phosphorous, potassium and sulfur of the soils and total protein content, nitrogen, phosphorus, potassium and sulfur of leaves. Maximum sampled soil was containing yellowish red color when moist and showed brown color when dried. Some of them were flood plain soil and that’s why showed grey color during dry and moist condition. Mean particle density was 2.49 g/cm3 and mean moisture content was 13.95%. Average sand, silt and clay were 65.08%, 18.56% and 19.26%, respectively. The texture of the soils was sandy loam to clay loam. The sand was by far the dominant fraction in the soils. Mean pH value was 5.63, organic matter was 0.30% and EC was 99.38μS/cm. Mean total nitrogen, phosphorus, potassium and sulfur of the soils were 0.04, 0.46, 0.25 and 0.26%, respectively. The mean concentration of protein, nitrogen, phosphorus, potassium and sulfur in the betel leaf samples were 20.56, 3.29, 0.39, 0.79 and 0.20%, respectively. Purpose: to study the effect of ethanol on the parameters of THC, the percentage of granulocytes and total protein in the hemolymph of the Red claw crayfish (Cherax quadricarinatus).Materials and methods. The object of this experiment was 26 males of the Australian red-clawed crayfish (Cherax quadricarinatus) weighing from 23 to 83 g. The individuals were evenly divided into two experimental groups - with an injection of ethanol and a control group without an injection of 13 crayfish for each group. The injection dose was 2515 mg per 100 g of body weight. A day after the introduction of ethanol, hemolymph was taken with a syringe from the ventral sinus, the syringe was pre-washed with a 4% EDTA-Na2 solution. Three parameters were determined: the total hemocyte count (THC), percent granulocytes and percent total protein content. Counting of hemocytes and determination of granulocytes were performed in a Goryaev chamber under a light microscope. The total protein was determined by the refractometric method.Results. Differences in THC and total protein between the groups were statistically unreliable (p>0,05). THC in the experimental group is 36% more than in the control group. The total protein after the introduction of ethanol actually increased by 0,7%, and relatively by 14%. There were statistically different indicators of the proportion of granulocytes (p<0,05) - the average value of 33,1% in the experimental group versus 24,5% in the control group. A reliable (p<0,05) strong feedback was revealed between the total protein and the mass of individuals in both experimental groups, while in the experimental group there is a visible shift in the values of dependent hemolymph indicators towards an increase in smaller individuals.Conclusion. A single injection of ethyl alcohol with a dosage of 2515 mg per 100 g of body weight into the hemolymph of C. quadricarinatus does not cause significant changes in the THC and total protein after 24 hours. At the same time, the proportion of granulocytes actually increases by 9%, relative to 37%. This may indicate that granulocytes are involved in the formation of cancer defense mechanisms when exposed to toxic substances. The effect of different dosages of ethanol injections and the duration of its effect on hematological parameters requires additional consideration. It is necessary to investigate its effect on other indicators, such as the pH and buffer capacity of the hemolymph, the concentration of hemocyanin, glucose, lactates and calcium.

Abstract BackgroundThe pathological characteristics of acute lung injury (ALI) or acute respiratory distress syndrome (ARDS) are pulmonary edema resulting from pulmonary permeability increasing. The main cause is uncontrolled inflammatory response leading to the damage of pulmonary vascular endothelial and alveolar epithelial barriers. However, there has not been effective drugs against ALI. In this study, we investigated the function of Isopropyl 3-(3, 4-dihydroxypheny l)-2-hydroxypropanoate (IDHP), a novel metabolite of Danshen dripping pill having anti-inflammatory effect, in lipopolysaccharide (LPS) induced ALI in mice, and its underlying mechanisms.MethodsPretreatment of IDHP in LPS-induced acute lung injury in mice were observed on survival rate, pulmonary morphologic changes, total protein content in bronchoalveolar lavage fluid (BALF), and inflammatory cytokines in lung tissue and BALF. To further explore its mechanism on ALI, THP-1 macrophages was studied to analyse pyroptosis related proteins and co-culture with epithelial or endothelial cells to assess protection function of IDHP in vitro.ResultsAs revealed by survival study, pretreatment with high dose of IDHP reduced the mortality of mice from ALI. IDHP pretreatment significantly improved LPS-induced lung pathological changes, reduced protein leakage and lung myeloperoxidase activity. IDHP also inhibited the release of inflammatory mediators TNFα, IL-1β, IL-6 and IL-18 in BALF and lung tissue. Meanwhile, IDHP decreased the expression of active-caspase1 (in canonical pyroptosis pathway), caspase4/5 (non-canonical pyroptosis pathway), Nrlp3, mature IL-1β, mature IL-18, Asc speck formation, and cleaved Gsdmd, all these are required for pyroptosis, in LPS stimulated THP-1 macrophages. Moreover, IDHP also decreased ROS production in LPS-stimulated THP-1 macrophages, inhibited the expression of tight junction proteins (Occludin, Zo-1) in endothelial cells, and decreased lactate dehydrogenase activity in supernatants of epithelial or endothelial cells, co-cultured with LPS-stimulated THP-1 macrophages. ConclusionsPretreatment of IDHP improves survival rate and ameliorates LPS-induced ALI in mice possibly via inhibiting canonical and non-canonical pyroptosis pathways.

Total protein describes the liver's ability to synthesize proteins and metabolize substances in the blood. Bovine serum can be used as a control material because the total protein analyte in bovine blood serum is almost identical to the analyte in human serum. The researchers intend to determine the homogeneity and stability of bovine serum with the addition of 7,5% ethylene glycol preservative after being stored for 12 weeks at -20°C as an alternative to control serum for total protein levels. This research used survey analytics using a one-group pretest-posttest research design.  Data analysis used ISO 13528 2005 calculations. From the examination of the total protein level in the serum, the value of Xr – Yr = 0.23817, which is where the value meets the test criteria according to |  Xr - Yr |  <0.36, that is 0.23817 <0.91585. The total protein content of bovine serum was homogeneous and stable with the addition of 7.5% ethylene glycol stored at -20oC for 12 weeks.

Background Any dairy herd that continually has a somatic cell count (SCC) above 200,000 cells/ml has an indication of mammary gland inflammation (mastitis). Routine use of antibiotics to prevent mastitis is prohibited by organic farming regulations. This limitation has lead researchers to focus on cows natural defense mechanisms [1]. Calcarea carbonica derivative complex (M8) is a complex high diluted medication comprised of comprised of Calcarea carbonica 16x, Aconitum napellus 20x, Arsenicum album 18x, Asa foetida 20x, Conium maculatum 17x, Ipecacuanha 13x, Phosphorus 20x, Rhus toxicodendron 17x, Silicea 20x, Sulphur 24x, and Thuya occidentalis 19x. Dilution procedures have followed standard methodology described at the Brazilian Homeopathic Pharmacopoeia. This medication has enhanced immune system responses both in vitro and in vivo in a murine model [2]. Aims In the present study, we investigate the response of dairy cows after M8 treatment. Methodology The study was performed as a randomized, observer double-blinded and placebo-controlled trial, with a stratified design, using lactation number and SCC as stratification factors. The study sample consisted of 42 lactating dairy cows (Holstein) in one high producing dairy herd with 52 cows in milk in southern Brazil, divided into two experimental groups (n=21). Exclusion criteria were cows with clinical mastitis or receiving any other medical treatment. Pre- and post-milking teat disinfection was practiced in the herd. All cows were clinically examined, with udder and milk samples being appraised according to Rosenberger (1990) [3]. During 3 months one group received daily M8 treatment, the other placebo. Oral administration of 5 ml/day/cow was performed using an automatic dosage dispenser. Monthly, milk production, SCC, fat and total protein content were carefully recorded for each animal by an official milk recording program. SCC were log transformed for analysis. ANOVA and Tukey test were used to compare the averages. The Bartlett´s test was used for homogeneity of variance evaluation. Results There were no significant differences (P=0.435) among the groups in the initial evaluation (Values of SCC x103 : Placebo 67,37±80,48; Treatment 359,39±677,02). After 3 months, the M8 treated group showed a decrease (134,00±178,76 P= 0.047) in SCC when compared with control group (391,71±686,60). Fat and protein did not differ between groups and time analysed. Milk production decreased in the placebo group during time (Before: 34,97±6,69kg; After:28,69±4,33kg), whereas the treatment group did not change total amount (Before: 28,7±6,54kg; After: 26,39±5,73kg; P > 0.05). Conclusions These results indicate that the M8 influenced positively SCC and suggest that it may be considered as a possible tool to promote bovine mastitis prophylaxis. Background: High level of salinity deteriorates seed germination, growth and yield of crops in cultivated lands all over the world. There is no effective remedy to mitigate this global problem. Homeopathy offers a remedy like Natrum mur which at ultra high dilution ameliorates diseases of patients having strong desire for salt consumption. The purpose of the present study is to see whether potentized Natrum mur could alleviate salt stress in germinating seeds of cowpea Vigna unguiculata (L) Walp. Methods: Water-soaked seeds were kept over moist filter paper in covered petridishes which were divided into 5 groups: (1) unstressed and untreated control in sterile distilled water, (2) in 100mM sodium chloride solution, (3) seeds pretreated with Natrum mur 200c and then kept in sterile distilled water, (4) seeds pretreated with Natrum mur 200c and then transferred to 100mM NaCl solution and (5) seeds pretreated with 90% ethanol. Both Natrum mur 200 c and its diluent medium 90% ethanol were diluted with distilled water 1:100 before use for treatment. Results: Natrum mur 200c increased the rate of seed germination, seed water content and growth of seedlings. The drug also enhanced chlorophyll, soluble and insoluble sugar, rubisco and total protein content as compared to the untreated salt stressed group. Treatment with Natrum mur 200c increased salt tolerance in the seedlings as compared to the untreated salt stressed group. All the data were analyzed by ANOVA and the significance level was not less than 1%. Conclusion: Natrum mur 200c reversed the effects of salt stress in germinated seeds thereby providing evidence for Hahnemann’s similia principle in plants. Potentized Natrum mur could be safely used with profit on plants grown on brackish soil.

Arsenic (As3+) is a toxic metalloid found in the earth’s crust, its elevated concentration is a concern for human health because rice is the staple grain in eastern part of India and the waterlogged rice field environment provides opportunity for more As3+ uptake. Magnesium (Mg2+) is an important plant nutrient. Present work is a search for reducing As3+ toxicity in plants through Mg2+ application. The findings are quite impressive, the root to shoot biomass ratio showed more than 1.5 times increase compared to the control. Total protein content increased 2 folds. Carbohydrate and chlorophyll content increased two to three times compared to control. On the other hand, Malondialdehyde content showed a decline with the application of increased Mg2+ dose. The in-silico study shows a better interaction with As3+ in presence of Mg2+ but interestingly without stress symptoms. These findings from the research indicate that Mg2+ application can be effective in reducing As3+ induced stress in plants.

Background: Linen dressings were invented a few years ago but are still being worked on. Methods: The obtained fabrics from the traditional variety of flax (Nike), two transgenic types of flax (M50 and B14) and the combination of these two flax fibers (M50 + B14) were tested in direct contact in cell cultures. Cell viability tests were performed, and the proliferation potential of cells on Balb3T3 and NHEK cell lines was checked using the Sulforhodamine-B (SRB) test. Moreover, the effect of new linen fabrics on apoptosis of THP-1 cells, as well as on the cell cycle of NHEK, HMCEV and THP-1, cells after 24 h of incubation was assessed. Results: All tested linen fabrics did not raise the number of necrotic cells. The tested fabrics caused a statistically significant decrease in the total protein content in skin cancer (except for 0.5 cm of Nike-type fabrics). The smallest cells in the apoptotic phase were in cultures treated with M50 fiber on an area of 0.5 cm. After 48 h of incubation of HEMVEC, NHEK and THP-1 cells with the tested fabrics, the growth of S-phase cells was noticed in all cases. At the same time, the greatest increase was observed with the use of B14 fabric. Necrosis is not statistically significant. Conclusions: All the obtained flax fibers in the form of flax dressings did not lose their wound-healing properties under the influence of the technological process. New dressings made of genetically modified flax are a chance to increase the effectiveness of treatment of difficult healing wounds.

Methylorubrum extorquens AM1 is an efficient platform strain possessing biotechnological potential in formate- and methanol-based single carbon (C1) bioeconomy. Constitutive expression or costly chemical-inducible expression systems are not always desirable. Here, several glucose-, xylose-, and levulinic acid (LA)-inducible promoter systems were assessed for the induction of green fluorescent protein (GFP) as a reporter protein. Among them, the LA-inducible gene expression system (HpdR/PhpdH) showed a strong expression of GFP (51-fold) compared to the control. The system was induced even at a low concentration of LA (0.1 mM). The fluorescence intensity increased with increasing concentrations of LA up to 20 mM. The system was tunable and tightly controlled with meager basal expression. The maximum GFP yield obtained using the system was 42 mg/g biomass, representing 10% of the total protein content. The efficiency of the proposed system was nearly equivalent (90%–100%) to that of the widely used strong promoters such as PmxaF and PL/O4. The HpdR/PhpdH system worked equally efficiently in five different strains of M. extorquens. LA is a low-cost, renewable, and sustainable platform chemical that can be used to generate a wide range of products. Hence, the reported system in potent strains of M. extorquens is highly beneficial in the C1-biorefinery industry to produce value-added products and bulk chemicals.
