(1) Background: Soil degradation is an increasingly important problem in many parts of the world, particularly in arid and semiarid areas. Arbuscular mycorrhizal fungi (AMF) isolated from arid soils are recognized to be better adapted to these edaphoclimatic conditions than exogenous ones. Nevertheless, little is known about the importance of AMF inoculum sources on Tamarix articulata development in natural saline soils. Therefore, the current study aims at investigating the efficiency of two AMF-mixed inoculums on T. articulata growth, with consideration of its rhizosphere microbiota. (2) Methods: indigenous inoculum made of strains originating from saline soils and a commercial one were used to inoculate T. articulata in four saline soils with different salinity levels under microcosm conditions with evaluation of rhizosphere microbial biomasses. (3) Results: Our findings showed that indigenous inoculum outperforms the commercial one by 80% for the mycorrhizal rate and 40% for plant biomasses, which are correlated with increasing shoot phosphorus content. Soil microbial biomasses increased significantly with indigenous mycorrhizal inoculum in the most saline soil with 46% for AMF, 25% for saprotrophic fungi and 15% for bacterial biomasses. (4) Conclusion: Present results open the way towards the preferential use of mycorrhizal inoculum, based on native AMF, to perform revegetation and to restore the saline soil microbiota.

This study investigated arbuscular mycorrhizal fungi (AMF) inoculation and phosphorus fertilizer on growth, productivity and quality of onion grown under saline soil conditions. The experiment was conducted in the Experimental Farm of Desert Research Center, Ras Sudr, South Sinai Governorate, Egypt. Egyptian local onion cultivar “Giza 20” was selected. Treatments were: two arbuscular mycorrhizal fungi (AMF) inoculation treatments [without inoculation (–AMF) and with mycorrhizal inoculation (+AMF)] and four phosphorus supplied treatments (0, 48, 96 and 144 kg P2O5 ha–1). Mycorrhizal inoculation and phosphorus addition affected the plant growth, bulb yield and quality. Morphological traits of plant (height, leaf number, fresh and dry weight), neck diameter, bulb characters (diameter and weight), total soluble sugars (TSS), protein, P content increased, while proline content decreased due to the inoculation of AMF and phosphorus application. Onion inoculated by AMF combined with 96 or 144 kg P2O5 ha–1 gave the highest productivity under saline conditions.

Tomato (Lycopersicon esculentum Mill. `Heinz 1350 VF 402') seedlings were inoculated with populations of vesicular–arbuscular mycorrhizal (VAM) fungi collected from saline or nonsaline soil or remained nonmycorrhizal as a control. Plants then were salinated for 8 weeks at 1.0, 2.0, 5.0, or 10.0 dS·m–1 produced by dilutions of 1 m NaCl: 1 m CaCl2 in deionized water. Inoculation with VAM fungi from nonsaline soil enhanced shoot growth, while VAM fungi from saline soil suppressed shoot and root growth. Plants inoculated with VAM fungi from nonsaline soil and non-VAM control plants showed a quadratic increase in leaf Cl– concentration in response to an increased salinity level, whereas plants inoculated with VAM fungi from saline soil showed a linear increase in leaf Cl– concentration. Mycorrhizal-induced growth responses and changes in leaf Cl– concentration were not associated with any apparent alterations in tomato plant P status. Although VAM fungi originating from saline soil did not promote plant growth, reduction in leaf Cl– concentration mediated by these VAM fungi at moderate salinity levels may have beneficial implications for plant survival in saline soil.

Abstract Background Increasing the presence of beneficial soil microorganisms is a promising sustainable alternative to support conventional and organic fertilization and may help to improve crop health and productivity. If the application of single bioeffectors has shown satisfactory results, further improvements may arise by combining multiple beneficial soil microorganisms with natural bioactive molecules. Methods In the present work, we investigated in a pot experiment under greenhouse conditions whether inoculation of two phosphate-solubilizing bacteria, Pseudomonas spp. (B2) and Bacillus amyloliquefaciens (B3), alone or in combination with a humic acids (HA) extracted from green compost and/or a commercial inoculum (M) of arbuscular mycorrhizal fungi (AMF), may affect maize growth and soil microbial community. Phospholipid fatty acid (PLFA) and denaturing gradient gel electrophoresis (DGGE) fingerprinting analysis were performed to detect changes in the microbial community composition. Results Plant growth, N and P uptake, and mycorrhizal root colonization were found to be larger in all inoculated treatments than in the uninoculated control. The greatest P uptake was found when B. amyloliquefaciens was applied in combination with both HA and arbuscular mycorrhizal fungi (B3HAM), and when Pseudomonas was combined with HA (B2HA). The PLFA-based community profile revealed that inoculation changed the microbial community composition. Gram+/Gram− bacteria, AMF/saprotrophic fungi and bacteria/fungi ratios increased in all inoculated treatments. The greatest values for the AMF PLFA marker (C16:1ω5) and AMF/saprotrophic fungi ratio were found for the B3HAM treatment. Permutation test based on DGGE data confirmed a similar trend, with most significant variations in both bacterial and fungal community structures induced by inoculation of B2 or B3 in combination with HA and M, especially in B3HAM. Conclusions The two community-based datasets indicated changes in the soil microbiome of maize induced by inoculation of B2 or B3 alone or when combined with humic acids and mycorrhizal inoculum, leading to positive effects on plant growth and improved nutrient uptake. Our study implies that appropriate and innovative agricultural management, enhancing the potential contribution of beneficial soil microorganisms as AMF, may result in an improved nutrient use efficiency in plants.

Grapevine (Vitis vinifera L.), widely used for berry and wine production, is highly susceptible to the pathogenic oomycete Plasmopara viticola, the etiological agent of grapevine downy mildew disease. The method commonly used to prevent and control P. viticola infection relies on multiple applications of chemical fungicides. However, with European Union goals to lower the usage of such chemicals in viticulture there is a need to develop new and more sustainable strategies. The use of beneficial microorganisms with biocontrol capabilities, such as the arbuscular mycorrhizal fungi (AMF), has been pointed out as a viable alternative. With this study, we intended to investigate the effect of AMF colonization on the expression of P. viticola effectors during infection of grapevine. Grapevine plants were inoculated with the AMF Rhizophagus irregularis and, after mycorrhizae development, plants were infected with P. viticola. The expression of P. viticola RxLR effectors was analyzed by real-time PCR (qPCR) during the first hours of interaction. Results show that pre-mycorrhizal inoculation of grapevine alters the expression of several P. viticola effectors; namely, PvRxLR28, which presented decreased expression in mycorrhizal plants at the two time points post-infection tested. These results suggest that the pre-inoculation of grapevine with AMF could interfere with the pathogen’s ability to infect grapevine by modulation of pathogenicity effectors expression, supporting the hypothesis that AMF can be used to increase plant resistance to pathogens and promote more sustainable agriculture practices, particularly in viticulture.

A vast majority of terrestrial plants are dependent on arbuscular mycorrhizal fungi (AMF) for their nutrient acquisition. AMF act as an extension of the root system helping phosphate uptake. In agriculture, harnessing the symbiosis can potentially increase plant growth. Application of the AMF Rhizophagus irregularis has been demonstrated to increase the yields of various crops. However, there is a paradigm that AMF colonization of roots, as well as the plant benefits afforded by inoculation with AMF, decreases with increasing phosphorus (P) supply in the soil. The paradigm suggests that when fertilized with sufficient P, inoculation of crops would not be beneficial. However, the majority of experiments demonstrating the paradigm were conducted in sterile conditions without a background AMF or soil microbial community. Interestingly, intraspecific variation in R. irregularis can greatly alter the yield of cassava even at a full application of the recommended P dose. Cassava is a globally important crop, feeding 800 million people worldwide, and a crop that is highly dependent on AMF for P uptake. In this study, field trials were conducted at three locations in Kenya and Tanzania using different AMF and cassava varieties under different P fertilization levels to test if the paradigm occurs in tropical field conditions. We found that AMF colonization and inoculation responsiveness of cassava does not always decrease with an increased P supply as expected by the paradigm. The obtained results demonstrate that maximizing the inoculation responsiveness of cassava is not necessarily only in conditions of low P availability, but that this is dependent on cassava and fungal genotypes. Thus, the modeling of plant symbiosis with AMF under different P levels in nature should be considered with caution.

A broad diversity of microorganisms can be found in soil, where they are essential for nutrient cycling and energy transfer. Recent high-throughput sequencing methods have greatly advanced our knowledge about how soil, climate and vegetation variables structure the composition of microbial communities in many world regions. However, we are lacking information from several regions in the world, e.g. Middle-East. We have collected soil from 19 different habitat types for studying the diversity and composition of soil microbial communities (both fungi and bacteria) in Qatar and determining which edaphic parameters exert the strongest influences on these communities. Preliminary results indicate that in overall bacteria are more abundant in soil than fungi and few sites have notably higher abundance of these microbes. In addition, we have detected some soil patameters, which tend to have reduced the overall fungal abundance and enhanced the presence of arbuscular mycorrhizal fungi and N-fixing bacteria. More detailed information on the diversity and composition of soil microbial communities is expected from the high-throughput sequenced data.

<p>Copper-based fungicides are commonly applied in vineyards to control fungal diseases that can severely affect grapevine productivity. Continuous application of this type of fungicides contributes to Cu accumulation in surface horizons of the soil, which can generate toxicity problems in plants, regardless of being an essential nutrient. Several strategies have been proposed to immobilize or counteract the effect of soil contaminants, such as plant inoculation with arbuscular mycorrhizal fungi (AMF). However, depending on the element concentration, this may not be sufficient to avoid its excessive accumulation in belowground and/or aboveground organs. Since Fe is known to have an antagonistic interaction with Cu in plants, Fe application, as an amendment, in vineyard soils, could be a good strategy to avoid excessive Cu uptake by grapevines growing in Cu-contaminated soils. However, little information is available on the combined effects of both strategies.</p><p>In order to reveal the possible beneficial effects of plant mycorrhization and Fe application in Cu-contaminated soils on grapevine growth and nutrition, a mesocosm experiment was established under controlled conditions. Two-year-old plants, previously inoculated or not with two different AMF, were grown in pots filled with 6.5 kg of an Arenosol collected from a wine-growing region. These plants were subjected to three soil treatments: 1) soil contamination with Cu, where the grapevines were watered with a solution containing 5.89 mg/L CuSO<sub>4</sub> to ensure that the soil in each container reached 300 mg Cu/kg; 2) soil contamination with Cu + Fe addition, where the plants were watered with a solution that contained the same amount of CuSO<sub>4</sub> plus 0.38 mg/L of FeNaEDTA·3H<sub>2</sub>O to achieve 100 mg of Fe/kg soil; and 3) non-contaminated soil watered with deionized water. Four months later, at the end of the growing season, plant vegetative growth as well as leaf and root nutrient contents were analyzed.</p><p>Grapevines inoculated with AMF demonstrated a good level of tolerance to high Cu concentrations in soil, as they presented significantly higher root biomass than non-inoculated plants and Cu was mainly accumulated in the roots avoiding its translocation to the aerial part. However, when the Cu-contaminated soil was amended with Fe, a significant decrease was observed in root biomass in all mycorrhizal inoculation treatments and Cu was accumulated in grapevine leaves. Contrastingly, Fe application helped to avoid the excessive increase of Mn concentrations in leaf and roots that is commonly induced in Cu contaminated soils, which can be detrimental for grapevine growth.</p><p>These results demonstrated that mycorrhizal inoculation is a suitable strategy to promote grapevine growth in Cu-contaminated soils. However, special attention needs to be taken when applying amendments to correct Cu contamination, as the mycorrhizal status of plants may alter the expected outcome.</p><p> </p><div> <div> </div> </div>
