Eight selected wild vegetables from Nepal (Alternanthera sessilis, Basella alba, Cassia tora, Digera muricata, Ipomoea aquatica, Leucas cephalotes, Portulaca oleracea and Solanum nigrum) were investigated for their antioxidative potential using 2,2-dyphenyl-1-picrylhydrazyl (DPPH) scavenging, hydrogen peroxide (H2O2), ferric reducing antioxidant power (FRAP), and ferric thiocyanate (FTC) methods. Among the selected plant extracts C. tora displayed the highest DPPH radical scavenging activity with an IC50 value 9.898 μg/mL, whereas A. sessilis had the maximum H2O2 scavenging activity with an IC50 value 16.25 μg/mL—very close to that of ascorbic acid (16.26 μg/mL). C. tora showed the highest absorbance in the FRAP assay and the lowest lipid peroxidation in the FTC assay. A methanol extract of A. sessilis resulted in the greatest phenolic content (292.65 ± 0.42 mg gallic acid equivalent (GAE)/g) measured by the Folin–Ciocalteu reagent method, while the smallest content was recorded for B. alba (72.66 ± 0.46 GAE/g). The greatest flavonoid content was observed with extracts of P. oleracea (39.38 ± 0.57 mg quercetin equivalents (QE)/g) as measured by an aluminium chloride colorimetric method, while the least was recorded for I. aquatica (6.61 ± 0.42 QE/g). There was a strong correlation between antioxidant activity with total phenolic (DPPH, R2 = 0.75; H2O2, R2 = 0.71) and total flavonoid content (DPPH, R2 = 0.84; H2O2, R2 = 0.66). This study demonstrates that these wild edible leafy plants could be a potential source of natural antioxidants.

Objective: The present study evaluates the antioxidant and α-amylase inhibitory activity of leaf and bark extracts of Phyllanthus indofischeri with methanol and water as solvents. In addition to this, the total phenolic content and total flavonoid content was determined.Methods: The total phenolic and total flavonoid content of the extracts was determined by folin ciocaletus reagent method and aluminium chloride colorimetric method respectively. The antioxidant and α-amylase inhibitory activity were measured by various assays, including α, α-diphenyl-ẞ-dipicryl-hydrazyl (DPPH) free radical scavenging, 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radical scavenging, superoxide radical scavenging, total antioxidant capacity by phosphomolybdate method and porcine pancreatic α-amylase inhibitory assay. The IC50 values were calculated and compared with standards such as gallic acid, ascorbic acid and α-acarbose.Results: The results illustrated that all the extracts of Phyllanthus indofischeri exhibit significant antioxidant and α-amylase inhibitory activity. Among the extracts, methanolic leaf extract showed high levels of activity followed by bark water extract.Conclusion: Phyllanthus indofischeri extracts had shown antioxidant and α-amylase inhibitory activity. On the basis of these results, Phyllanthus indofischeri can be used as a natural antioxidant and hypoglycemic agent against various disorders related to oxidative stress; and the isolation of bioactive compounds was warranted.

The methanolic extract of Artemisia incisa Pamp. was dissolved in distilled water and successively partitioned with n-hexane, chloroform, ethyl acetate and n-butanol. The antioxidant potential of all these fractions and remaining aqueous fraction was evaluated by four methods, i.e., the scavenging activity of the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), the total antioxidant activity, the ferric reducing antioxidant power (FRAP) assay and the ferric thiocyanate assay. In addition, the total phenolics was determined. The obtained results revealed that among the studied fractions the ethyl acetate soluble fraction showed the most potent DPPH-radical scavenging activity with an IC50 value of 5.3 ? 0.71 ?g mL-1, which is even more effective than the standard antioxidant butylated hydroxytoluene (BHT) (IC50 value 0f 12.1? 0.92 ?g mL-1). The ethyl acetate fraction also showed the highest FRAP value (3677.13 ? 27.1 ?g TE mL-1), inhibition of lipid peroxidation (60.93 ? 0.84 % at 500 ?g mL-1) and total phenolic content (95.5 ? 0.05 ?g GAE g-1) as compared to other fractions. However, the remaining aqueous fraction was found to posses the highest antioxidant activity of all the fractions.

Abstract Seaweed is one of the marine-biota that is widely known for its uses, one of them is in the food functional. Functional food is a program that promotes a healthy diet so that it can prevent a disease when consuming food. With functional food, daily eating patterns can be maintained and also the content of the food consumed daily has a good effect on the body. It is known that in seaweed, especially brown algae (S. crassifolium), it contains antioxidants and other compounds that are good for the health. In this study, phytochemical content and proximate composition was evaluated in S. crassifolium. Including total flavonoid content and total phenolic content, antioxidant capacity both of DPPH and ABTS assay used three type of solvents such as Ethanol, n-Hexana, and ethyl acetate for comparable the best solvents. Also, proximate analysis to determine the content of moisture content, crude fat content, crude protein content, and carbohydrates content. The results showed that n-Hexana and ethyl acetate solvents were significantly than ethanol. Which mean both of them had highest values of total flavonoid content (TFC), total phenolic content (TPC), and antioxidant activity of DPPH radical scavenging activity and ABTS radical scavenging activity. The results of chemical composition in proximate compounds showed that S. crassifolium was suitable to be candidate of functional food.

Plant based remedies are of much importance in healthcare due to low side effects. The effective constituents contained in seeds of a papaya have not been utilized efficiently in the production of medicines. The purpose of this study was driven towards determining the total phenolic content, total flavonoid content, antioxidant capacity, anti-inflammatory activity and antibacterial properties of the aqueous seeds extract of a Sri Lankan variety (Red Lady) of papaya (AESP). The AESP prepared according to the method of “Kasaya” in Ayurvedic medicine was used for the investigations according to standard procedures. Total phenolic content and the total flavonoid content of the AESP were 13.5±2.2 mg (pyrogallol equivalence)/g and 315.9±104.6 mg (quercetin equivalence)/g respectively. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of AESP ranged from 12.4% to 81.2% for concentrations of 1.0-20.0 mg/ml; ascorbic acid gave 31.7% - 91.9% activity. The AESP displayed strong reducing power compared to ascorbic acid in the reducing power assay. The hydroxyl radical scavenging activity of the AESP ranged from 6.6% to 69.1% for concentrations ranging from 20.0-95.0 mg/ml; ascorbic acid activity was 37.7%-74.9%. The nitric oxide radical scavenging activity of AESP was 23.9%-62.7% for concentrations of 2.0-20.0 µg/ml; ascorbic acid gave 26.8%-63.5% activity. AESP concentrations ranging from 75.0-150.0 µg/ml exhibited human red blood cell (HRBC) membrane stabilization protection of 15.5%-22.7% compared to 50.8%-58.4% for aspirin. The AESP showed antibacterial activity against Bacillus subtilis and Staphylococcus aureus bacterial species. AESP possess promising antioxidant, anti-inflammatory and antibacterial activity. The aqueous decoction of the discarded seeds of papaya would facilitate remedies for many diseases in which radicals are implicated as well as assist against certain bacterial infections and also has an anti-inflammatory potential.

Ocimum sanctum, commonly known as Tulasi in Nepal, is a pharmacologically important plant due to its active constituents. In this work, extraction was carried out in hexane, chloroform and methanol solvents and their phytochemical screening was performed. Total phenolic and flavonoid contents in the plant were measured by Folin-Ciocalteu colorimetric method and aluminum chloride colorimetric method respectively. Antioxidant activity of the extracts was evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Alkaloids, flavonoids, tannins, glycosides, polyphenols, terpenoids, tannins and steroids are mainly found in the extracts. Based on the result obtained, the plant posses a significant amount of total phenolic and total flavonoid content. Both phenolic and flavonoid contents were highest in methanol extract, followed by chloroform and hexane extract. Antioxidant activity of the extracts as ascorbic acid standard (IC50 value = 41.34 µg/mL) was in the order of methanol extract (IC50 value = 47.73 µg/mL) > chloroform extract (IC50 value = 79.46 µg/mL) > hexane extract (IC50 value = 94.68 µg/mL). The extent of the antioxidant activity of the plant is following the number of total phenolics and flavonoids present in it.

Citrus fruits exhibit a high level of different phytoconstituents, of which the changes in the different parts of the fruit during ripening have not been thoroughly studied yet. Thus, in this study, we have investigated how different parts of pomelo fruit (Citrus grandis L.) are modified throughout the development of two consecutive growing seasons. In detail, the main phytochemical compounds, such as total phenolic content, total flavonoid content, antioxidant capacity, DPPH free radical scavenging activity, Ferric reducing antioxidant power (FRAP), and naringin and tannin content, were analyzed. A systematic metabolism of these compounds was found during the development of the fruit, but some pomelo tissues showed a fluctuating trend, suggesting a dependence on the different growing season. Focusing on the tissue distribution of these compounds, the fruit membrane contained the highest level of total phenolic and flavonoid content; fruit flavedo displayed the highest antioxidant capacities and FRAP activities, whereas maximum accumulation of naringin was noticed in fruit albedo. Instead, the highest DPPH free radical scavenging activity and tannin contents were found in the pomelo juice. Regarding the distribution of compounds, a possible bias pattern for the accumulation of those compounds has been noticed throughout the fruit development. From the GC-MS analysis, a total of 111 compounds were identified, where 91 compounds were common in both seasons. Overall, these results could be useful for the food processing industry as guidelines for excellent quality foods and for introducing health-beneficial products and components into our daily diets.

Byproducts obtained after wine production constitute an important source for the extraction of antioxidant compounds. Phenolic compounds showed a wide spectrum of biological actions for the human health, often associated with neuroprotective, cardioprotective, antimicrobial and other health-related effects. The aim of the present study was to determine the phenolic compounds content and antioxidant activity of grape marc from selected grapes from Iaşi vineyard. The total phenolic content of the extracts was measured using the Folin-Ciocâlteu method, with some modifications. The DPPH (2,2-diphenyl-1-picrylhydrazyl) assay was used to evaluate radical scavenging activity, by bleaching of purple methanolic solution of the stable radical. All extracts were able to reduce DPPH radical with different degrees of scavenging activity. The highest radical scavenging activity was determined for the extract obtained from Merlot and Fetească Neagră (98.142 μg/mL, and 115.78 μg/mL respectively), with positive correlation between scavenging activity on DPPH and total phenolic content. The total phenolic content varied among the analysed extracts, with a higher concentration in extracts obtained from Merlot, Fetească Neagră and Traminer rosé (2.42 GAE/mL, 1.45 GAE/mL and 1.24 GAE/mL respectively), followed by Sauvignon Blanc and Muscat Ottonel extracts with comparable amounts. The FRAP (ferric reducing antioxidant power) method relies on the color change of a complex with Fe+3 ion of the 2,4,6-tri(2-pyridyl) – 1,3,5-triazine (TPTZ) radical due to the reduction of the ferric ion to the ferrous iron (Fe+2) in this complex. The highest radical scavenging activity was determined in the extract obtained from Merlot and Fetească Neagră (1164.22 μM Trolox/100 mL and 782.09 μM Trolox/100 mL respectively), a positive correlation between scavenging activity determined by FRAP method and total phenolic content being observed.Statistical analysis showed a close relationship between content of phenolic compounds and antioxidant capacity, the marc of Merlot and Fetească Neagră had the highest contents of anthocyanins. The results suggested that grape pomace may be exploited as antioxindant source.

Several wild honey samples collected from seven different regions in Indonesia were investigated to determine their total phenolic content, flavonoid content, and free radical scavenging activity by analyzing the 1,1-diphenyl-2-picrylhydrazyl (DDPH) radical and phenolic profile. Rutin, (+)-catechin, ferulic acid, and galangin were found to be the major phenolic compounds of Indonesia wild honey. The total phenolic content significantly correlated with the total flavonoid content (p=0.000) and the percentage of DPPH radical scavenging activity (p=0.000). Results indicated that there are different polyphenol profiles among the different regions.
