In this study, the Pb(II) adsorption capabilities of the heavy metal tolerant strain of fungus, Aspergillus piperis, were studied. This study involved finding optimal growth conditions using a plating technique, and optimal adsorption conditions using submerged fermentation and fractional factorial experimental design. The adsorption behaviour was then elucidated using isotherm and kinetic models, of which the one surface Langmuir isotherm provided the best fit, with a maximum predicted adsorption capacity of 275.82 mg g−1. The kinetic models suggested that internal mass transfer is the driving force behind the reaction rate. After adsorption, biomass surface characterisation was undertaken using FESEM, EDS, and ATR-FTIR to explain observations. The system was characterised by a cation exchange mechanism with strong carboxyl and organophosphorus group interactions. This study demonstrates that due to the ease of propagation and high adsorption capacity, this locally sourced fungal strain is an ideal adsorbent for industrial Pb(II) bioremediation.

In this paper, a study on the feasibility of the treatment of raw cheese whey by anaerobic co-digestion using coffee pulp residues as a co-substrate is presented. It considers raw whey generated in artisanal cheese markers, which is generally not treated, thus causing environmental pollution problems. An experimental design was carried out evaluating the effect of pH and the substrate ratio on methane production at 35 °C (i.e., mesophilic conditions). The interaction of the parameters on the co-substrate degradation and the methane production was analyzed using a response surface analysis. Furthermore, two kinetic models were proposed (first order and modified Gompertz models) to determine the dynamic profiles of methane yield. The results show that co-digestion of the raw whey is favored at pH = 6, reaching a maximum yield of 71.54 mLCH4 gVSrem−1 (31.5% VS removed) for raw cheese whey and coffee pulp ratio of 1 gVSwhey gVSCoffe−1. The proposed kinetic models successfully fit the experimental methane production data, the Gompertz model being the one that showed the best fit. Then, the results show that anaerobic co-digestion can be used to reduce the environmental impact of raw whey. Likewise, the methane obtained can be integrated into the cheese production process, which could contribute to reducing the cost per energy consumption.

AbstractThe comparison between bentonite-alginate beads and bentonite-alginate films in Cd(II) removal from aqueous solution was investigated. The experimental Cd(II) adsorption data were fitted to adsorption isotherm and kinetic models. Results showed that both bentonite-alginate film and beads were fitted better to the Langmuir isotherm model, the calculated saturation adsorption capacity for bentonite-alginate film was 59.9 mg/g as compared to 17.9 mg/g for the beads. The kinetic studies indicated that both samples followed the pseudo-second-order kinetic. The bentonite- alginate film showed higher adsorption capacity, rate constant as well as initial adsorption rate. This investigation showed that the adsorption behavior of both samples, in terms of adsorption isotherm and kinetic models, did not deviate significantly. The bentonite-alginate film has better performance in Cd(II) removal, attributed to the larger surface area which is exposed to the Cd(II) solution.

Background: Modified bio-based adsorbents from plant sources can be used for pollution remediation by adsorption due to their low cost and availability in large quantities. Objective: In this study, the competitive biosorption of Pb(II) and Cu(II) by Micropogonias undulates functionalised fish scales (FFS) was conducted. The functionalisation was done by wet impregnation with Fe2+. Method: The biosorbent was characterised by Fourier Transform Infrared Spectroscopy (FT-IR), Scanning Electron Microscopy with Energy-Dispersive X-ray Spectroscopy (SEM-EDS) and Branueur–Emmett–Teller (BET) analyses. Results: The major constituents in the FFS were calcium and phosphorus from the collagen and apatite on the scales. Optimum removal efficiency for both metals was >99% at 10 g/l dosage. It was observed that the Langmuir isotherm model and the pseudo second order kinetics model were the best fit for the experimental data. The monolayer adsorption capacity of FFS for Pb(II) and Cu(II) was observed to be 96.15 mg/g and 100 mg/g respectively. Conclusion: The study revealed that the competitive biosorption of heavy metals can be achieved (at a good adsorption capacity) using functionalised Micropogonias undulates fish scales.

This paper presents a two-step methodology for the stochastic generation of snapshot peak demand scenarios in water distribution networks (WDNs), each of which is based on a single combination of demand values at WDN nodes. The methodology describes the hourly demand at both nodal and WDN scales through a beta probabilistic model, which is flexible enough to suit both small and large demand aggregations in terms of mean, standard deviation, and skewness. The first step of the methodology enables generating separately the peak demand samples at WDN nodes. Then, in the second step, the nodal demand samples are consistently reordered to build snapshot demand scenarios for the WDN, while respecting the rank cross-correlations at lag 0. The applications concerned the one-year long dataset of about 1000 user demand values from the district of Soccavo, Naples (Italy). Best-fit scaling equations were constructed to express the main statistics of peak demand as a function of the average demand value on a long-time horizon, i.e., one year. The results of applications to four case studies proved the methodology effective and robust for various numbers and sizes of users.

In contrast to many other cases in microbial development, Aspergillus nidulans conidiophore production initiates primarily as a programmed part of the life cycle rather than as a response to nutrient deprivation. Mutations in the acoD locus result in "fluffy" colonies that appear to grow faster than the wild type and proliferate as undifferentiated masses of vegetative cells. We show that unlike wild-type strains, acoD deletion mutants are unable to make conidiophores under optimal growth conditions but can be induced to conidiate when growth is nutritionally limited. The requirement for acoD in conidiophore development occurs prior to activation of brlA, a primary regulator of development. The acoD transcript is present both in vegetative hyphae prior to developmental induction and in developing cultures. However, the effects of acoD mutations are detectable only after developmental induction. We propose that acoD activity is primarily controlled at the posttranscriptional level and that it is required to direct developmentally specific changes that bring about growth inhibition and activation of brlA expression to result in conidiophore development.

After having studied in great detail the observational HR diagram (log Teff, Mbol) composed by 40 main sequence stars of the Hyades (Perryman et al.,1997, A&A., in press), we have tried to apply the same method to the observational main sequences of the three next nearest open clusters: Coma Berenices, the Pleiades, and Praesepe. This method consists in comparing the observational main sequence of the clusters with a grid of theoretical ZAMSs. The stars composing the observational main sequences had to have reliable absolute bolometric magnitudes, coming all from individual Hipparcos parallaxes, precise bolometric corrections, effective temperatures and metal abundances from high resolution detailed spectroscopic analyses. If we assume, following the work by Fernandez et al. (1996, A&A,311,127), that the mixing-lenth parameter is solar, the position of a theoretical ZAMS, in the (log Teff, Mbol) plane, computed with given input physics, only depends on two free parameters: the He content Y by mass, and the metallicity Z by mass. If effective temperature and metallicity of the constituting stars of the 4 clusters are previously known by means of detailed analyses, one can deduce their helium abundances by means of an appropriate grid of theoretical ZAMS’s. The comparison between the empirical (log Teff, Mbol) main sequence of the Hyades and the computed ZAMS corresponding to the observed metallicity Z of the Hyades (Z= 0.0240 ± 0.0085) gives a He abundance for the Hyades, Y= 0.26 ± 0.02. Our interpretation, concerning the observational position of the main sequence of the three nearest clusters after the Hyades, is still under way and appears to be greatly more difficult than for the Hyades. For the moment we can say that: ‒ The 15 dwarfs analysed in detailed in Coma have a solar metallicity: [Fe/H] = -0.05 ± 0.06. However, their observational main sequence fit better with the Hyades ZAMS. ‒ The mean metallicity of 13 Pleiades dwarfs analysed in detail is solar. A metal deficient and He normal ZAMS would fit better. But, a warning for absorption in the Pleiades has to be recalled. ‒ The upper main sequence of Praesepe, (the more distant cluster: 180 pc) composed by 11 stars, analysed in detail, is the one which has the best fit with the Hyades ZAMS. The deduced ‘turnoff age’ of the cluster is slightly higher than that of the Hyades: 0.8 Gyr instead of 0.63 Gyr.

We describe the actinomycete strain IMV-70 isolated from the soils of Kazakhstan, which produces potent antibiotics with high levels of antibacterial activity. After the research of its morphological, chemotaxonomic, and cultural characteristics, the strain with potential to be developed further as a novel class of antibiotics with chemotherapeutics potential was identified asStreptomycessp. IMV-70. In the process of fermentation, the strainStreptomycesspp. IMV-70 produces the antibiotic no. 70, which was isolated from the culture broth by extraction with organic solvents. Antibiotic compound no. 70 was purified and separated into individual components by HPLC, TLC, and column chromatography methods. The main component of the compound is the antibiotic 70-A, which was found to be identical to the peptolide etamycin A. Two other antibiotics 70-B and 70-C have never been described and therefore are new antibiotics. The physical-chemical and biological characteristics of these preparations were described and further researched. Determination of the optimal growth conditions to cultivate actinomycete-producer strain IMV-70 and development of methods to isolate, purify, and accumulate preparations of the new antibiotic no. 70 enable us to research further the potential of this new class of antibiotics.

Hyaluronan (HA), a polymeric glycosaminoglycan ubiquitously present in higher animals, is hydrolyzed by hyaluronidases (HAases). Here, we used bee HAase as a model enzyme to study the HA-HAase interaction. Located in close proximity to the active center, a bulky surface loop, which appears to obstruct one end of the substrate binding groove, was found to be functionally involved in HA turnover. To better understand kinetic changes in substrate interaction, binding of high molecular weight HA to catalytically inactive HAase was monitored by means of quartz crystal microbalance technology. Replacement of the delimiting loop by a tetrapeptide interconnection increased the affinity for HA up to 100-fold, with a KD below 1 nm being the highest affinity among HA-binding proteins surveyed so far. The experimental data of HA-HAase interaction were further validated showing best fit to the theoretically proposed sequential two-site model. Besides the one, which had been shown previously in course of x-ray structure determination, a previously unrecognized binding site works in conjunction with an unbinding loop that facilitates liberation of hydrolyzed HA.

Aeromonas proteolytica was grown for various time periods in nutrient broth, tryptic soy broth, a semisynthetic medium, and 1 and 5% peptone under different conditions involving temperature and in continuous shake and stationary flasks. The cell-free culture filtrates were tested for hemolytic, endopeptidase, and dermonecrotic activity and optimal growth conditions for their production were determined. The dermonecrotic activity and endopeptidase activity was found to be parallel in all tests, while hemolysin was independent of the other two. Studies on the thermal stability of the culture filtrate revealed that hemolysin and dermonecrotic and endopeptidase activity were destroyed at 70 °C for 30 min. Fractionation of the filtrate by Sephadex G-200 resolved three peaks at 280 nm. Peak I was inactive; peak II contained endopeptidase and dermonecrotic and hemolytic activity; peak III contained pigment and hemolysin. Evidence is presented that the endopeptidase and dermonecrotic substance found in the cell-free filtrates of A. proteolytica grown medium appear at the same time and thus may be the same entity.
