Aim. Study spectrum and expressiveness of antibacterial and antifungal properties oflichen extracts. Materials and methods. Antimicrobial activity of acetone extracts from Hypogymnia physodes, Xanthoria parietina, Evernia prunastri, Ramalina pollinaria, Cladonia arbuscula lichens was determined by micro-dilution methods in broth for 4 - 500 mcg/ml concentrations against 13 strains from ATCC collection and 6 clinical isolates. Results. High antibacterial activity of H. physodes and C. arbuscular extracts against staphylococci and enterococci was detected (MIC 31 - 62 mcg/ml). Antimicrobial activity against enterobacteria and Pseudomonas aeruginosa was absent for all the extracts. E.prunastri, H.physodes and C arbuscula extracts were active against Stenotrophomonas maltophilia strains (MIC 250 - 500 mcg/ml). Antifungal activity (MIC 500 mcg/ml for 4 Candida strains) was only detected for the E. prunastri extract. Conclusion. H.physodes and C. arbuscula lichens can be examined as a perspective source of antibacterial substances, effective against antibiotics resistant staphylococci, streptococci and S. maltophilia strains.

AbstractDue to wide range of secondary metabolites, lichens were used from antiquity as sources of colorants, perfumes and medicaments. This research focuses on exploring the antioxidant, antimicrobial and cytotoxic activities of methanol, ethanol, acetone extracts and aqueous infusions of corticolous lichens sampled from Armenia. Methanol, ethanol and acetone extracts from all tested lichens were active against Gram-positive bacterial strains. The most effective solvent to retrieve antimicrobial compounds was methanol. Aqueous infusions of tested lichens didn’t show any significant antibacterial and antifungal activity. The highest antimicrobial activity was observed for methanol extract of Ramalina sinensis. The minimum inhibitory concentration of methanol extract of Ramalina sinensis were 0.9–1.8 mg mL− 1. Pseudevernia furfuracea demonstrated antifungal activity (Ø 12 mm). Methanol extract of Parmelia sulcata demonstrated largest 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging activity (71 %). The cytotoxicity was measured on human HeLa (cervical carcinoma) cell lines using microculture tetrazolium test assay. The IC50 values estimated for methanol extracts of Peltigera praetextata, Evernia prunastri, Ramalina sinensis and Ramalina farinacea species in HeLa cell line were within 1.8–2.8 mg mL− 1 and considered as non-cytotoxic. Obtained results suggest that studied lichens can be prospective in biotechnologies as alternative sources of antimicrobial and antioxidant substances.

Background: Although a great number of the targets of antimicrobial therapy have been achieved, it remains among the first fields of pharmaceutical research, mainly because of the development of resistant strains. Docking analysis may be an important tool in the research for the development of more effective agents against specific drug targets or multi-target agents 1-3. Methods: In the present study, based on docking analysis, ten tetrahydrothiazolo[2,3-a]isoindole derivatives were chosen for the evaluation of the antimicrobial activity. Results: All compounds showed antibacterial activity against eight Gram-positive and Gram-negative bacterial species being, in some cases, more potent than ampicillin and streptomycin against all species. The most sensitive bacteria appeared to be S. aureus and En. Cloacae, while M. flavus, E. coli and P. aeruginosa were the most resistant ones. The compounds were also tested for their antifungal activity against eight fungal species. All compounds exhibited good antifungal activity better than reference drugs bifonazole (1.4 – 41 folds) and ketoconazole (1.1 – 406 folds) against all fungal species. In order to elucidate the mechanism of action, docking studies on different antimicrobial targets were performed. Conclusion: According to docking analysis, the antifungal activity can be explained by the inhibition of the CYP51 enzyme for most compounds with a better correlation of the results obtained for the P.v.c. strain (linear regression between estimated binding Energy and log(1/MIC) with R 2 =0.867 and p=0.000091 or R 2 = 0.924, p= 0.000036, when compound 3 is excluded.

Background:A novel copper-catalyzed synthesis of substituted-1,2,3-triazole derivatives has been developed and performed by Huisgen 1,3-dipolar cycloaddition reaction of azides with alkynes. The reaction is one-pot multicomponent.Objective:We state the advancement and execution of a methodology allowing for the synthesis of some new substituted 1,2,3-triazole analogues with antimicrobial activity.Methods:A series of triazole derivatives was synthesized by Huisgen 1,3-dipolar cycloaddition reaction of azides with alkynes. The structures of the synthesized compounds were elucidated and confirmed by 1H NMR, IR, MS and elemental analysis. All the synthesized compounds were tested for their antimicrobial activity against a series of strains of Bacillus subtilis, Staphylococcus aureus and Escherichia coli for antibacterial activity and against the strains of Candida albicans, Aspergillus flavus and Aspergillus nigar for antifungal activity, respectively.Results and Conclusion:From the antimicrobial data, it was observed that all the newly synthesized compounds showed good to moderate level of antibacterial and antifungal activity.

We evaluated the antimicrobial activity of thirty-one nitrogen-containing 5-α-androstane derivatives in silico using computer program PASS (Prediction of Activity Spectra for Substances) and freely available PASS-based web applications (such as Way2Drug). Antibacterial activity was predicted for 27 out of 31 molecules; antifungal activity was predicted for 25 out of 31 compounds. The results of experiments, which we conducted to study the antimicrobial activity, are in agreement with the predictions. All compounds were found to be active with MIC (Minimum Inhibitory Concentration) and MBC (Minimum Bactericidal Concentration) values in the range of 0.0005–0.6 mg/mL. The activity of all studied 5-α-androstane derivatives exceeded or was equal to those of Streptomycin and, except for the 3β-hydroxy-17α-aza-d-homo-5α-androstane-17-one, all molecules were more active than Ampicillin. Activity against the resistant strains of E. coli, P. aeruginosa, and methicillin-resistant Staphylococcus aureus was also shown in experiments. Antifungal activity was determined with MIC and MFC (Minimum Fungicidal Concentration) values varying from 0.007 to 0.6 mg/mL. Most of the compounds were found to be more potent than the reference drugs Bifonazole and Ketoconazole. According to the results of docking studies, the putative targets for antibacterial and antifungal activity are UDP-N-acetylenolpyruvoylglucosamine reductase and 14-α-demethylase, respectively. In silico assessments of the acute rodent toxicity and cytotoxicity obtained using GUSAR (General Unrestricted Structure-Activity Relationships) and CLC-Pred (Cell Line Cytotoxicity Predictor) web-services were low for the majority of compounds under study, which contributes to the chances for those compounds to advance in the development.

AbstractFour water soluble azo dyes, 4-(isopropyl)-2-[(E)-(4-chlorophenyl)diazenyl]phenol (L 1), 4-(isopropyl)-2-[(E)-(2,4-dichlorophenyl)diazenyl]phenol (L2), 4-(sec-butyl)-2-[(E)-(4-chlorophenyl) diazenyl]phenol (L 3), 4-(sec-butyl)-2-[(E)-(2,4-dichlorophenyl)diazenyl]phenol (L 4), and their Cu(II) and Ni(II) complexes were synthesized and characterized using spectroscopic methods. Examination of their thermal stability revealed similar decomposition temperature of approximately 260–300°C and that they were more thermally stable than their metal complexes. Ni(II) complexes of ligands L2 and L4 were more stable than the other coordination compounds. Among the synthesized ligands, L2 and the complexes Cu(L3)2 and Ni(L4)2 showed both antimicrobial and antifungal activity. However, the other ligands and the complexes were poorly active against selected microorganisms.

In vitro, the photoprotective, cytotoxic and photomodifying properties of hexane and benzene extracts from the widely distributed in Belarus lichens Cladonia arbuscula, Evernia prunastri, Hypogymnia physodes, Ramalina pollinaria and Xanthoria parietina in the culture of human keratinocytes (HaCAT), are estimated. It was found that these extracts are not photoprotective, they have a photomodifying effect upon irradiation of keratinocyte cultures with ultraviolet light. Hexane extract of E. prunastri and benzene extracts of R. pollinaria and H. physodes at a concentration of 2.5 μg/ml are photoprotectors that weaken the effect of ultraviolet radiation by 1.6 ÷ 1.8 times. Extracts of C. arbuscula and X. parietina are photosensitizers – they increase the toxic effect of ultraviolet radiation by 10 or more times, regardless of the concentration of the extract in the nutrient medium. Hexane and benzene extracts of C. arbuscula and H. physodes lichens are cytotoxic to the HaCAT culture. With an increase in the dose of ultraviolet irradiation of keratinocytes from zero to lethal values, the benzene extract of R. pollinaria acted as a photoprotector at a concentration of 2.5 μg / ml, and as a photosensitizer at higher concentrations. The hexane and benzene extracts of E. prunastri and H. physodes exhibited sensitizing properties that increased with an increase in the concentration of lichen extracts. C. arbuscula hexane extract and X. parietina benzene extract were the most potent photosensitizers.

Цель – первое лихенологическое обследование техногенных отвалов Михайловского горно-обогатительного комбината (Железногорский район, Курская область) с прилегающими природными и урбанизированными территориями. Материалы и методы. Сборы материалов проводились маршрутным методом в течение 2017 и 2019-2020 годов, камеральная обработка осуществлялась с применением общепринятых лихенологических методик. Идентифицированные образцы хранятся в гербариях Центрально-Черноземного заповедника им. проф. В. В. Алехина и Станции юных натуралистов г. Железногорска. Результаты и обсуждение. Выявленная лихенобиота включает 88 видов (преимущественно макролишайников, вследствие методики сбора) из 38 родов, 18 семейств. Наиболее распространенные, металл-толерантные виды: геоплезные Cladonia fimbriata, C. coniocraea, эпигеидные C. rei, C. mitis, C. cornuta, C. furcata, C. phyllophora и C. gracilis, эпифито-эпиксильные Evernia prunastri, Parmelia sulcata, Xanthoria parietina, Hypogymnia physodes и Physcia adscendens. Составлен аннотированный список в различной степени редких видов, среди которых 4 новых для Центрального Черноземья (Cladonia cervicornis, Dibaeis baeomyces, Usnea dasopoga, U. lapponica); 5 новых для Курской области (Bryoria fuscescens, Cladonia carneola, C. deformis, Melanohalea septentrionalis, Peltigera extenuata); 6 занесенных в Красную книгу Курской области (Cladonia subrangiformis, C. subulata, Peltigera praetextata, Platismatia glauca, Pseudevernia furfuracea и Usnea subfloridana); еще 5 редки в области или на более обширных территориях (Acarospora veronensis, Protoparmeliopsis muralis, Ramalina pollinaria, Trapelia coarctata, Usnea hirta). Сосредоточение значительного числа редких видов в техногенных ландшафтах, по-видимому, обусловлено рядом причин: наличием крупных выходов редкого в области и Центральном Черноземье субстрата (ожелезненного песчаника); слабой конкуренцией со стороны сосудистых растений на бедных и обогащенных металлами почвах и грунтах; ботанико-географическим положением территории; спецификой локального загрязнения среды, представленного преимущественно «кислыми» поллютантами и частицами металлов. Заключение. Отмечена приуроченность регионально редких кустистых и листоватых эпифитов из экологической группы ацидофилов к насаждениям березы в изученных техногенных ландшафтах. Предложены некоторые меры охраны выявленных редких видов лишайников.

Mn(II) and Co(II) complexes were prepared by reaction of the metal chlorides with acetylacetone in ammonical aqueous medium. The metal complexes were prepared in order to investigate their antimicrobial activity on some selected pathogens. The characterisation of the complexes was on the basis of various spectroscopic techniques like infrared and ultraviolet studies. The compounds were subjected to antimicrobial activity screening using serial broth dilution method. Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal/Fungicidal Concentration (MBC/MFC) were determined. Mn(II) complex has shown significantly both antibacterial and antifungal activity with a MIC of 1.25 μg/mL while Co(II) complex was noticeable for antifungal activity at the same concentration. Whereas Mn(II) acetylacetonate is a more potent bactericide while Co(II) acetylacetonate is a more potent fungicide, both with MBC/MFC value of 2.5 μg/mL. Antimicrobial agent of the ligand has enhanced on complexation with Mn(II) and Co(II) ions. Though, the potency of the prepared antibiotics on the tested microbes is less compared to the standard drugs (Ciprofloxacin and Fulcin).

Four new Schiff bases were synthesized from condensation of aldehyde ( 2-hydroxy benzaldehyde ) with aromatic amines (4-introaniline ; 4,4-oxydianiline ; 4,4-diaminodipleny sulfone and sulfanilamide) . These Schiff bases were characterized by IR, NMR , GC. mass and element analysis. Moreover, the Schiff bases were screened for their antibacterial and antifungal activity against various microorganisms and compared with standard compounds. In general , the results were indicated that some Schiff bases had good antimicrobial activity.
