Transcriptional Expression of Serpin-6 Gene in Bombyx Mori Larval after Infection of Bacteria at Different Developmental Stages

Serpins can block different steps in the activation cascade of prophenoloxidase (proPO) system, and play an important role in immunity of insect. In this paper, Haemolymph was collected from the 4th molting, newly moulted 5th instar and day-3 fifth instar larval challenged by lipopolysaccharide (LPS) or Bacillus thuringiensis, respectively. The results revealed that the transcriptional level of Bmserpin-6 in different developmental stages showed a trend of rise first, then fall. Bmserpin-6 of the 4th molting larva expressed highest at 6h post-infection with LPS and 3h post-infection with Bacillus thuringiensis. Bmserpin-6 of newly moulted 5th instar larva expressed highest at 9h post-infection with LPS and 6h post-infection with Bacillus thuringiensis. Bmserpin-6 of day-3 fifth instar larva expressed highest at 9h post-infection with Bacillus thuringiensis. Bmserpin-6 was all highly induced and highly expressed in haemolymph of larval at different developmental stages. But The time to arrive the highest transcriptional level was different. This is inferred that the Serpin gene may play an important role in immunity of Bombyx mori.

A proton-translocating vacuolar-type ATPase (V-ATPase) was identified and characterized in the anterior silk gland of Bombyx mori. By incubating the intact tissue with the fluorescent dye Acridine Orange, the acidified compartment was detected at the apical pole of the epithelial cells. This was observed throughout the feeding period of the fifth-instar larva until the onset of spinning. Acidification was prevented completely and reversibly by 0.8 micromol l-1 bafilomycin A1, a specific inhibitor of V-ATPase. The presence of V-ATPase in a microsomal fraction was verified by immunoblots using an antiserum to the V-ATPase holoenzyme from Manduca sexta midgut. The antiserum localized the V-ATPase to the apical plasma membrane of the anterior silk gland cells, suggesting that the enzyme is functionally active in pumping protons out of the cell towards the glandular lumen of feeding silkworm larvae. In spinning larvae, the acidification produced by the V-ATPase appears to cease, because acidic compartments were seen rarely and only in the periphery of basal cytoplasm, and because immunocytochemical staining for the V-ATPase was greatly reduced at the apical surface. The metamorphic changes in relation to the occurrence of V-ATPase corresponded well with the ultrastructural changes in the anterior silk gland cell of Bombyx mori larvae.

NUPR1 (nuclear protein 1), also called P8 (molecular mass 8 kDa) or COM1 (candidate of metastasis 1), is involved in the stress response and in cancer progression. In the present study, we investigated whether human NUPR1 expression was regulated by TGFβ (transforming growth factor β), a secreted polypeptide largely involved in tumorigenesis. We demonstrate that the expression of NUPR1 was activated by TGFβ at the transcriptional level. We show that this activation is mediated by the SMAD proteins, which are transcription factors specifically involved in the signalling of TGFβ superfamily members. NUPR1 promoter analysis reveals the presence of a functional TGFβ-response element binding the SMAD proteins located in the genomic DNA region corresponding to the 5′-UTR (5′-untranslated region). Altogether, the molecular results of the present study, which demonstrate the existence of a TGFβ/SMAD/NUPR1 activation cascade, open the way to consider and investigate further a new mechanism enabling TGFβ to promote tumorigenesis by inducing stress resistance.

The duration of hatching, larval instars and pupal stages of Stethorus punctillum feeding on two-spotted spider mite, Tetranychus urticae were investigated in different seasons under laboratory conditions. The highest values of different developmental stages were obtained during winter. Higher temperature significantly reduced the duration of different developmental stages. No significant effect of relative humidity was exerted on the development stages of S. punctillum. The predation rate of fourth instar larva of S. punctillum was the highest whereas the first instar larva consumed the lowest number of prey. The fourth instar larva of the predator consumed 135.8 eggs, 126.4 larvae, 96.6 nymphs and 72.8 adults per day separately. But the first instar consumed 41.6 eggs, 36.2 larvae, 26.8 nymphs and 16.8 adults during the same period. The insect pathogens Bacillus sotto Ishiwata and Bacillus thuringiensis Berliner can be differentiated by means of certain cultural and morphological differences, and by a quantitative difference in pathogenicity for the larvae of Bombyx mori L., B. sotto being the more pathogenic. They are also pathogenic for the larvae of several North American Lepidoptera including Anisota rubicunda (F.), Anisota senatoria (A. and S.), Nymphalis antiopa (L.), Erannis tiliaria (Harr.), Datana integerrima (G. and R.), Datana ministra (Drury), Liparis dispar L., Protoparce quinquemaculata (Haw.), and Protoparce sexta (Johan.). Silkworm larvae ingesting material from a sporulated culture of B. sotto become sluggish, cease feeding, and suffer from a progressive paralysis that begins in the mid-gut area, spreads to affect the whole larva, and ends in death.
