In Vitro Assessment of Antibacterial Activity and Cytotoxicity of Silver Contained Antibacterial HA Coating Material

Failure of dental implant is often caused by specific pathogenic bacteria. In this paper, antibacterial HA coating was prepared by vacuum plasma spraying with silver loaded zirconium phosphate containing HA as feedstock. The antibacterial activity against Aa,Fn and Pg was evaluated using a film covering method. MTT colorimetric assay was used to measure cytotoxic effect of the antibacterial HA coatings. Results show that the HA coatings displayed significant antibacterial activity against Aa,Fn, Pg and no cytotoxicity to L929 murine cell lines when the content of silver-loaded zirconium phosphate antimicrobial is between 5wt% to 10wt%. The sensitivity of the bacteria to the antibacterial HA coating is in sequence of Pg >Fn >Aa.

Vacuum plasma sprayed (VPS) hydroxyapatite (HA) coatings with silver-loaded zirconium phosphate antimicrobial have been prepared on titanium substrate. Antibacterial effects of the coatings were studied by bacteria culturing using Porphyromonas gingivalis (Pg), Fusobacterium nucleatum (Fn) and Actinobacillus actinomycetemcomitans (Aa) as microorganisms. Simulated body fluid (SBF) test was carried out to evaluate the bioactivity of the coatings as well. The results obtained showed that the coatings exhibited a marked antibacterial effect against Pg, Fn and Aa when the content of antimicrobial was equal to or more than 5%. The antibacterial capability of the coatings decreased in the following order: Pg, Fn, Aa. Bone-like apatite layer was formed on the silver-containing HA coatings after immersed in SBF, suggesting that their bioactivities were not affected obviously by the addition of silver-zirconium phosphate antimicrobial. This study indicated that silver-containing VPS HA coating is a prospective candidate as dental material.

AbstractInfectious bursal disease (IBD) caused by non-enveloped double stranded RNA virus is an acute and contagious poultry disease. Outbreak of IBD could result in 10–75% mortality of the birds; hence it has gained socio-economic importance worldwide. Medicinal plants have shown broad spectrum anti-viral activities against RNA and DNA viruses. Moringa oleifera Lam (MOL), Phyllanthus emblicus Linn (PEL), Glycyrrhiza glabra Linn (GGL), and Eugenia jambolana Lam (EJL) are commonly available medicinal plants of the sub-continent and exhibited anti-viral potential against different viruses. Ethanolic extracts of the leaves of MOL and EJL, roots of GGL and dried fruit of PEL were investigated for their cytotoxic and anti-viral potential against IBD virus using MTT colorimetric assay and anti-viral assay. Significant anti-viral potential (P<0.001) was demonstrated at concentrations 12.5, 25, 50 and 100 µg ml−1 of GGL, PEL, MOL and EJL, respectively, with no cytotoxicity. Data also spotlighted that all tested plant extracts possess significant anti-viral potential and this trend was higher in GGL followed by PEL, MOL, and EJL. The data undoubtedly conclude that these medicinal plants contain several health beneficial phyto-chemicals which got significant anti-viral potential and effectively be utilized against IBD virus. Moreover, the outcomes of this study provide a platform on the way to discover novel anti-viral agents against IBD virus and other viruses from plant origin.

Vibriosis is one of diseases which often results in mass mortality of Penaeus monodon larval rearing systems. It attacks shrimp of all stages in zoea, mysis and shrimp postlarva stage. This disease is caused by Vibrio spp, particularly Vibrio harveyi (a luminescent bacterium). Several kinds of antibiotics and chemical material have been used to overcome the disease but they have side effects to environment and human. The searching of bioactive compounds as an alternative treatment has been done for multi purposes. In this study diethyl eter, butanol and aqueous extract of Indonesian sponges Aaptos aaptos and Callyspongia pseudoreticulata were tested for in vitro activity against Vibrio spp. and Vibrio harveyi by using disc diffusion method. The result showed that all extracts of Aaptos aaptos gave a positive antibacterial activity towards those pathogenic bacteria. Meanwhile, only butanol extract of Callyspongia pseudoreticulata obtained to exhibit an antibacterial activity on those pathogenic bacteria. The strong anti-vibrio activity were shown by butanol and aqueous extract of Aaptos aaptos with the minimum inhibitory concentration (MIC) value of 0.313 and 0.625 mg/mL, respectively. Whilst, the butanol extract of Callyspongia pseudoreticulata indicated a low antibacterial activity with the MIC value of 10 mg/mL. Toxicity of those active extracts was evaluated by Brine Shrimp Lethality Test (BST). Interestingly, butanol and aqueous extracts of Aaptos aaptos did not show any toxic effect in Artemia salina larvae up to 8 x MIC (2.504 mg/mL and 5.000 mg/mL). It is the first report for the anti-vibr io activity of both Aaptos aaptos and Callyspongia pseudoreticulata. This results suggest that Aaptos aaptos has a potential to be used as a source of alternative compound to vibriosis prevention for mariculture.

ABSTRACT-QMFS2019Poultry production provides source of protein and contributes an important income for Vietnamese farmers. Among the poultry in Vietnam, ducks account for 27.3% of head of poultry and even 55.7% in Mekong Delta region. Along with the development of rearing ducks, bacterial, viral and fungal diseases occurring in the two last decades induced bad effect for poultry producer. Escherichia coli, Salmonella enterica, Streptococcal or Pasteurella act as major pathogenic bacteria in duck. The aims of this study were to investigate the antibacterial activity of garlic Allium sativum against E. coli, Staphylococcus aureus, Salmonella Typhimurium and to evaluate the effect of garlic on growth performance of duck from 1-28 old-days. The results indicated that fresh garlic and dried garlic powder showed inhibitory effect against pathogenic tested strains from 2% and 4% w/v, respectively. The inhibition zones and the minimal inhibitory concentration (MIC) values of garlic extract ranged from 11.3-28.3 mm and 0.02-0.2 g/ml, respectively. After 28 days of diet with garlic supplemented, D3 (2% of fresh garlic in water) showed significantly different in weight gain (WG), feed conversion ratio (FCR), protein efficiency ratio (PER), average daily weight (ADW); whereas, D2 (2% of garlic powder in basal diet) only possessed a difference significant in feed consumption (FC) compared to the D1 (control without garlic supplementation). The obtained results demonstrated the potential of garlic application in poultry production.

The use of turmeric rhizome to treat diarrhea is written in the original Indonesian medicinal manuscript. Diarrhea disease is still a public health problem in Indonesia, because of its high morbidity and mortality. The morbidity survey conducted by Indonesian Ministry of Health shows an increasing incidence trend. One of the causes of diarrhea is an uncontrolled increase in the number of intestinal bacteria and infection by intestinal pathogenic bacteria. This study aims to determine the effect of the turmeric rhizome preparation process, both traditionally and by extraction method by maceration and soxhletation on antibacterial activity, especially intestinal bacteria, so that it can be applied by the traditional medicine industry as well as traditional herbal medicine sellers (“jamu gendong”). The research method used was to test the antibacterial activity of fresh turmeric juice, pre-dried turmeric juice, turmeric extract by maceration using 95% ethanol, and turmeric extract by soxhletation at 100°C using 95% ethanol. The intestinal bacteria used in this study were Escherichia coli, Yersinia enterolitica, Vibrio nonagglutinable, and Shigella flexneri. The results of this study indicate that the treatment process using the traditional method, both fresh turmeric juice and pre-dried turmeric juice, does not show any antibacterial activity. Turmeric extract by maceration showed antibacterial activity against all bacterias and the greatest against Vibrio nonagglutinable bacteria. Turmeric extract by soxhletation showed antibacterial activity against all bacterias and the greatest against Vibrio nonagglutinable bacteria. The conclusion of this study is that the most appropriate method used to process turmeric rhizome as a medicine for diarrhea caused by bacteria is the extraction method by maceration or soxhletation. The greatest antibacterial effect is against the Vibrio nonagglutinable bacteria.

Study contextual: Faced with the global problem of antimicrobial resistance, and the use of traditional medicine for the research of antibacterial biomolecules. Aim: our work focused on the valorization of a medicinal plant Dittrichia viscosa which has many therapeutic and culinary virtues worldwide. Methods: To do this, a phytochemical screening of the leafy stems of the plant is carried out according to a set of physicochemical reactions, as well as an in vitro evaluation of the antibacterial activity, of the aqueous and ethanolic extracts against multi-resistant bacterial strains, by microdilution technique on microplates. An evaluation of the synergistic interaction between extracts and weakened antibiotics against pathogenic bacteria was also highlighted in this study. Results: The tests revealed the richness of Dittrichia viscosa species by tannins, flavonoids, saponosides, sterols and triterpenes. As for the antibacterial effect, the MICs range from 0.858±0.29 to 66.66 ± 0.00 mg / ml and the MBCs from 4.300 ± 1.01 to 11.610 ± 2.31 mg / ml is an interesting antibacterial activity. Regarding the combination of extracts with antibiotics tested, it revealed a synergistic action inducing an amplification of the antibacterial power of Penicillin, Imipenem and Erythromycin with a rate that reaches 471%. Conclusion: The results of this study show that the aqueous and ethanolic extracts of Dittrichia viscosa have interesting and promising antibacterial activity in the pharmaceutical, food and cosmetic industries.

Background: In the past few decades focus of research has been toward herbal medicines because of growing bacterial resistance and side effects of antimicrobial agents. The extract derived from the plants may increase the efficacy of antibiotics when used in combination against pathogenic bacteria. In the current study, the synergistic antibacterial efficacy of plant extracts in combination with antibiotics has been assessed on selected periodontal pathogens. Methods: Ethanolic extracts were prepared from Salvadora persica (Miswak) and Cinnamomum zeylanicum (Ceylon cinnamon), by the soxhalate method. Plaque samples were collected from clinical periodontitis patients to isolate and grow the periodontal pathobionts under favorable conditions. Susceptibility of bacteria to the extracts was assessed by gauging the diameter of the inhibition zones. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of plant extracts were determined against each bacterium. Synergistic activity of plants extract in combination with antibiotics against the bacteria was also assessed by measuring the diameter of the inhibition zones. Results: Ethanolic extract of both the plants showed an inhibitory effect on the proliferation and growth of all four strains of periodontal pathobionts. Maximum antibacterial activity was exhibited by C. zeylanicum against Tannerella forsythia (MIC = 1.56 ± 0.24 mg/mL, MBC = 6.25 ± 0.68 mg/mL), whereas among all the studied groups the minimum activity was reported by C. zeylanicum against Aggregatibacter actinomycetemcomitans the (MIC = 12.5 ± 3.25 mg/mL, MBC = 75 ± 8.23 mg/mL). Combination of herbal extracts with different antibiotics revealed a synergistic antibacterial effect. The best synergism was exhibited by S. persica with metronidazole against A. actinomycetemcomitans (27 ± 1.78). Conclusions: Current in vitro study showed variable antibacterial activity by experimented herbal extracts against periodontal pathobionts. The synergistic test showed significant antibacterial activity when plant extracts were combined with antibiotics.
