Ameliorative effects of aqueous extracts of ginger and garlic on Hibiscus sabdariffa-induced testicular damage in male Wistar rats

Background: The incidence of cognitive decline has been proposed to rise exponentially in coming years. Therapies targeting molecular pathways involved in enhancement of memory and energy regulation could be a major breakthrough in prevention or management of dementia in susceptible populations. Objectives: This study investigated the effects of aqueous extracts of Cola nitida (AECONS) and Garcinia kola (AEGAK) on glutamate level and Na+/K+-ATPase activity in the hippocampus and hypothalamus of male Wistar rats. Methods: Adult male Wistar rats (170-200) were randomly allotted into groups (n=5/group); control (distilled water p.o.), AECONS1 (200 mg/kg), AECONS2 (400 mg/kg), AEGAK1 (200 mg/kg), AEGAK2 (400 mg/kg), AECONS1+AEGAK1 and AECONS2+AEGAK2. The extract was prepared and the administration was done daily for 6 weeks. Results and Discussion: Administration of AECONS or AEGAK increased plasma, hippocampal and hypothalamic glutamate, Na+/K+-ATPase activity, NO, SOD except hippocampal glutamate in AECONS1/AEGAK1, Na+/K+-ATPase activity and SOD in AEGAK1, hypothalamic glutamate and SOD in AECONS1 when compared with control. Besides, MDA level decreased in AEGAK2 and hippocampal but not hypothalamic MDA decreased in AEGAK1 compared with control. However, concomitant administration of AECONS and AEGAK enhanced plasma, hippocampal and hypothalamic biomarkers except hypothalamic MDA level. The present study demonstrates that AECONS and AEGAK synergistically enhances hippocampal and hypothalamic glutamate and Na+/K+-ATPase activity, which are accompanied by NO and SOD-dependent antioxidant enrichment. Conclusion: These findings therefore suggest that AECONS+AEGAK could be a better therapeutic candidate in hippocampal-hypothalamic-related neurodegenerative diseases.

Aim: The study was aimed at evaluating the effects of ethanolic extract of Sesamum indicum seeds on sperm morphology and viability in ketoconazole induced testicular damage in Wistar rats. Methods: Forty eight adult male Wistar rats were divided into eight Groups of six rats each. Groups 1,2,3,4,5,6,7 and 8 were administered 1ml of normal saline, 100 mg /kg body weight (bwt) of Ketoconazole only, 500 mg/kg bwt of ethanolic extract of sesamum indicum (EESI) seeds then100 mg/kg bwt of Ketoconazole, 100 mg/kg bwt of Ketoconazole then 250 mg /kg body weight of EESI seeds,100 mg/kg bwt of Ketoconazole then 500 mg/kg bwt of EESI, 100 mg/kg body weight of Ketoconazole then 0.36 mg/kg body weight of Mesterolone, 0.36 mg/kg body weight of Mesterolone, 500 mg/kg body weight of EESI respectively. All administrations were carried out orally once per day. The sperm smears were collected for semen analyses. Result: A significant increase in the abnormalities of sperm morphology and viability respectively were noted in the group administered ketoconazole alone (Group 2) compared with other experimental Groups (P ≤ 0.05). Conclusions: The administration of ethanolic extract of Sesamum indicum appeared to have minimized the damages and sperm deaths caused by ketoconazole which may be due to its androgenic properties.

Abstracts Background Chemotherapy is associated with male infertility. Cisplatin (CP), an antineoplastic agent has been successfully used for the treatment of diverse kinds of malignancies, however, the use of this effective agent could induce oxidative stress injury, nephrotoxicity, hepatotoxicity, and testicular damage. Combined CP chemotherapy with plant extracts can diminish the toxicity and enhance the antitumor efficacy of the drug. The objective of the study was to determine the protective effect Lophira lanceolata leaf extract (LLLE) on CP-induced toxicity on male reproductive organs. Methods The study was carried out with 30 (n = 30) male Wistar rats (Rattus norvegicus). The rats were randomly assigned into 6 groups of 5 rats each. Rats in group 1 (Control) were administered distilled water per os. Rats in group 2 were administered 5 mg/kg of CP intraperitoneally (i.p). Rats in groups 3 and 4 were administered per os LLLE at the doses of 200 and 400 mg/kg body weight and rats in groups 5 and 6 were administered 5 mg/kg body weight of CP + LLLE at the doses of 200 and 400 mg/kg body weight respectively. Results The results showed a significant decrease in the sperm parameters in the group treated with CP alone when compared with the control and there in the sperm parameters in the groups administered CP + LLLE. The body and organ weights of the rats were significantly (p < 0.05) decreased in the CP treated group relative to the control. However, there was an increase in the weight of the organs in the LLLE pretreated groups. The photomicrographs showed degenerative changes in the testicular tissues of the rats administered CP alone whereas the group pretreated with the LLLE showed amelioration induced by the CP. Our study revealed that CP treatment has deleterious effects on sperm parameters and testicular tissues and the accessory sex organs (Epididymis, prostate, seminal vesicles) of the rats. Oral administration of LLLE at 200 and 400 mg/kg bodyweight for 26 days conferred protective effects against testicular damage induced by CP. Conclusion This study revealed that pretreatment with LLLE protected against CP-induced testicular toxicity.

Herbal teas ofMoringa oleifera(Moringa) andGlycyrrhiza glabra(Licorice)were known to have many beneficial effects, their possible hypolipidemic effects were investigated in this paper. Albino wistar rats were fed with aqueous extracts ofM. oleifera,G. glabraand their blend teas(10, 30 and 50 mg/kg.BW/ml). The teas were found to lower the serum cholesterol, triglycerides, VLDL, LDL, and atherogenic index at their higher concentration, but were found to slightly increase the HDL as compared to the corresponding normal healthy rats fed with clean water (control). The phytochemicals screening of the teas aqueous extract were also investigated, the chemicals (tannins, saponins, flavonoids, terpenoids and glycosides) which were known to exhibit cholesterol lowering effect by inhibiting its absorption and simultaneous increase its excretion were found to be present in the teas extract. Thus, the study demonstrates and validates thatM.oleifera,G. glabraand their blend possess hypolipidemic effect at higher concentration.

Aim: To perform phytochemical screening of Plumbago zeylanica L. root extracts and assess the claim of its use in traditional management of erectile dysfunction in Uganda through evaluation of the aphrodisiac effect of its aqueous extracts in male Wistar rats. Study Design: This study employed both qualitative and quantitative research designs. Place and Duration of Study: All research work were performed at the Department of Biological Sciences, Gulu University, Gulu, Uganda between August 2019 and December 2019. Methodology: Classical phytochemical screening of aqueous and methanolic extracts of Plumbago zeylanica roots were performed following standard methods. Aqueous extracts were administered to Male Wistar rats and the effect of the extracts on the mounting and intromission frequencies were determined. Results: Glycosides, phenols, saponins, quinones, terpenoids and steroids were present in both methanolic and aqueous extracts. Alkaloids were present in methanolic extracts only while tannins and phlobatannins were only present in aqueous extracts. Flavonoids, coumarins and anthraquinones were not detected in both extracts. Aqueous root extracts of Plumbago zeylanica produced pro-sexual stimulatory effects in male rats when administered at 150, 300 and 450 mg/kg body weight. Conclusion: The results support the use of Plumbago zeylanicaroots by indigenous people in Uganda to increase libido, treat premature ejaculation and erectile dysfunction. The extracts had low sexual enhancement in sexually inexperienced male rats and therefore, further studies using experienced animal models are needed to better apprehend the prosexual effects of P. zeylanica roots. The toxicity of the extracts as well as structural elucidation and pharmacological evaluation of the responsible bioactive compounds merit further studies.
