Cryopreservation of germplasm from chickens kept in Canadian research institutions

Twenty-three genetically distinct lines of chickens are maintained as living populations by Agriculture and Agri-Food Canada and Canada’s eight Faculties of Agriculture. Historically, cryogenic storage of avian genetic material has been difficult, but we have developed techniques of gonadal transplantation to allow recuperation of stored genetic material into living birds. Gonads from 1660 day-old chicks or late-term embryos (810 females and 850 males) from 18 chicken populations from four Canadian institutions were harvested and cryopreserved using dimethylsulfoxide as a cryoprotectant. Future efforts will be directed to completing the collection of the populations kept in Canadian publicly-funded institutions that conduct agricultural research. SummaryBrazil has various species of domestic animals which developed from breeds brought by the Portuguese settlers soon after the discovery. Over the last five centuries, these breeds have been submitted to natural selection in particular environments and therefore today, they present characteristics adapted to the specific environmental conditions. From the beginning of this century, some exotic breeds, selected in temperate regions, have begun to be imported. Although more productive, these breeds lack adaptation traits, such as resistance to disease and parasites found in breeds considered to be“native” but even so, little by little, they have substituted the native breeds to such an extent that the latter are, today, in danger of extinction. To avoid the loss of this important genetic material, Brazil created an Animal Genetics Resource Conservation Programme, coordinated by the National Research Centre for Genetic Resources and Biotechnology (Cenargen) of the Brazilian Agricultural Research Corporation (EMPRAPA). The conservation has been carried out by various Research Centres of EMPRAPA, Universities, State Research Corporations, as well as by private farmers, with a single coordinator at national level, Cenargen. The conservation is being carried out through Conservation Nuclei, situated in the habitats where the animals have been subjected to natural selection (in situ), and by the storage of semen and embryos (ex situ). The recently created Animal Genetics Laboratory of Cenargen allowed genetic characterisation studies on cattle and horse breeds to begin, and, in the near future, work with asses, buffalo and sheep will be conducted‥ From the results of this research it will be possible to compare the native breeds and estimate genetic distances between them. The harmonisation of chosen micro-satellites with those which have been used in other Latin America and Iberian Peninsula countries will be extremely useful for comparative studies and will allow future exchange of germplasm between countries.

Abstract The structure of raspberry cultivars and genetic resources in the Baltic countries have been influenced by the historical political situation in the 20th century and climatic conditions, especially winterhardiness. The genetic resources consist of some old European and American cultivars, but mostly of cultivars and hybrids bred in Russia. Currently, targeted breeding programmes are active only in Estonia and Latvia, which aim to develop winterhardy, disease-resistant cultivars, well adapted to the local climate. Therefore, parent material for hybridisation has been chosen from local advanced hybrids and introduced cultivars suitable to the regional climatic conditions. The aim of the study was to estimate the level of genetic diversity of Rubus germplasm and assess inter-specific and intra-specific relationships using phenotypical characterisation and molecular markers. Forty one Rubus genotypes were evaluated by 41 phenotypical traits and 15 previously described SSR markers. Both characterisation approaches discovered high correspondence with pedigree and a low level of diversity. A limited amount of the diversity of raspberry genetic material has been used in various breeding programmes, despite their broad geographical origin. The obtained results indicate the need for including local wild R. idaeus plant material into breeding programmes.

Abstract Introduction Institute of Natural Fibres and Medicinal Plants (INF&MP) is involved in gathering and evaluation of genetic resources of flax (Linum usitatissimum L.), hemp (Cannabis sativa L.) and medicinal plants, because Poland has signed the Convention on Biological Diversity. Field trials were carried out in 1990–2001 in the Experimental Station in Wojciechów located in Opolskie region. The research material was accessions of genetic resources from the collection of flax, stored at the Institute of Plant Breeding and Acclimatization Institute in Radzików near Warsaw. Objective The aim of the publication was to evaluate the following biological features of flax: time of beginning of flowering and vegetation period to yellow maturity. Methods The characteristics data for flax accessions are presented according to the methodology of development of the International Flax Database. Each flax accessions from the world collection of flax genetic resources received one of the following assessments of two vegetation periods: very short, short, medium, long and very long. Descriptors of biological features of flax were used, presented in “List of flax descriptors (L. usitatissimum L.)” edited by Janka Nozkova, published in 2011. These descriptors are used to develop the International Flax Database, which in turn helps in the rapid evaluation of the L. usitatissimum genetic resource collection. Carrying out a detailed characterization of biological features of L. usitatissimum accessions is very helpful for breeders in selecting genetic material for breeding new varieties of flax. Results The following results were obtained from the time of the beginning of flowering and the vegetation period to yellow maturity. Time of beginning of flowering was the following: very short – 0 accessions, short – 97 accessions, medium – 20 accessions, long – 2 accessions and very long – 0 accessions of flax. The genotypes tested were characterized by the following results in terms of vegetation period from sowing to yellow maturity: very short – 0, short – 37, medium – 62, long – 0 and very long – 0 accessions of flax. Conclusions Time of beginning of flowering and the vegetation period to yellow maturity shall be revalorised to the International Flax Database for those L. usitatissimum accessions that did not receive the same assessment of the biological features tested in the two or three years of the study.

This chapter introduces the Consultative Group on International Agricultural Research (CGIAR) Virtual Library as a tool for linking researchers and agricultural research results. The CGIAR is a strategic alliance of countries, international and regional organizations, and private foundations supporting 15 international agricultural centers that work in partnership with national agricultural research institutions and societies. The research results generated are numerous and cover a wide range of subject fields. While these are properly documented, locating relevant and timely information across the system’s 15 centers is a long and tedious process as individual databases have to be searched. The CGIAR virtual library (CGVLibrary) project of the CGIAR Libraries and Information Services Consortium (CGIARLISC) was created in 2005 to address the difficulty of information retrieval across the various centers. It is now available via the WWW (http://vlibrary.cgiar.org/) and knowledge generated by the CGIAR can now be retrieved with a few mouse clicks.

Providing a sustained flow of improved physical, biological and social technology applicable to the country or region is essential in the strategy of agricultural development. Building research institutions, which have the capacity to supply this flow, has accounted for a large proportion of Land Grant University overseas technical assistance effort. Several papers and broad spectrum studies have attempted to review the experience to date with a view to inducing more productive effort. We shall (a) identify the main issues relating to required features of effective research institutions, (b) examine the history of American reasearch institutions for insight into the bases of effectiveness, (c) inventory institution-building attempts in Asian situations with regard to presence of these bases of effectiveness and (d) evaluate and interpret alternative policies of foreign assistance to build effective research programs in agriculture. Primary attention is focused upon one organizational aspect which appears to be both limitational and neglected – the engineering of dependable responsiveness of the institutions to the problems of their respective regions.

The current paper has considered the challenge of breeding of rice lines adapted to the climatic conditions of the Southern Federal District of Russia by pyramiding a complex of Pi-1, Pi-2, Pi-33, Pi-ta and Pi-b rice blast resistance genes which is efficient in the southern part of Russia. The control of the resistance genes present in the breeding material was carried out using a marker-associated selection (MAS). The purpose of the current study was to identify the Pi-1, Pi-2, Pi-33, Pi-ta, and Pi-b rice blast resistance genes in the rice samples developed in the laboratory of rice breeding and seed production of the FSBSI “Agricultural Research Center “Donskoy” to identify valuable genotypes. The screening of 689 breeding samples of rice resulted in identification of the allelic state of the Pi-1, Pi-2, Pi-33, Pi-ta and Pi-b rice blast resistance genes, and the genotypes with functional alleles of these genes and their combinations. There were identified 19 breeding samples of rice possessing a combination of 4 functional alleles of rice blast resistance genes in various combinations. The identified rice samples are valuable genetic material and are recommended to use as the sources of rice blast resistance in the breeding process.

An experiment was conducted at the Hill Agricultural Research Station, Khagrachari from September 2002 to February 2003 to find out the effect of mulching method (Mulching one month before planting, mulching just after planting and no mulching) and planting times (01 October, 16 October and 01 November) on the yield and yield attributes of tomato in hill slope. The highest yield (21.43 t/ha) was obtained from plant where mulch was given one month before planting. Among three planting times, the highest yield (15.27 t/ha) was obtained from 01 October planting. In case of combined effect, mulching one month before planting with 01 October planting produced the highest yield (28.06 t/ha) of tomato in hill slope.
