Objective: To observe inflammation of interstitial connective tissue space caused by Lead acetate in rat testis and ameliorative after math caused by Ficuscarica. Study Design: Laboratory based experimental study. Place and Duration of Study: Anatomy Department, Army Medical College Rawalpindi together with NIH (National Institute of Health) Islamabad, from Mar to Nov 2017. Methodology: Sprague Dawley male rats, 30 in quantity were chosen and 10 animals each partited into 3 groups. Treatments were given for 8 weeks, once daily. Group A was control group. Group B was treated with dosage of 30 mg/kg of Lead acetate. Group C was given dosage of 30 mg/kg of Lead acetate as well as 80 mg/kg of Ficuscarica. Twenty four hours after the concluding dose, animals were vivisected. For histological study, testis were fixed and stained with Haematoxylin and eosin. Interstitial connective tissue space thickness was morphometrically and assessed by SPSS version 22. A p-value <0.05 was considered significant in results. Results: Interstitial space thickness was significantly increased due to inflammation (>3 times normal) in group B in comparison to groups A and C. Thickness of space was slightly increased (<2 times normal) in group C in comparison to groups A due to reduction in inflammation. Conclusion: There was increased thickness of interstitium due to inflammation, cellular congestion and lymphocytic infiltration in rat’s testis because of lead acetate but concomitant dose of Ficuscarica protects against inflammation, venous congestion of interstitial space.

The formation of dentin and enamel in the abdominal wall in young pups was achieved by transplantation of the soft tissues of the developing tooth germ. An interesting finding was the cytomorphosis of the epithelium of the enamel organ. When this was transplanted so that the ameloblasts were in contact with the odontoblasts the cylindrical character of the epithelial cells was preserved and enamel was produced; otherwise the cylindrical shape of these cells was lost and a stratified epithelium resulted, resembling the gingival and certain tumors (the adamantinoma) of the jaw and related structures. This degenerated epithelium did not produce enamel and had an important characteristic of not forming cysts in a closed connective tissue space, instead forming islands and cords of cells with epithelial pearl formation. Thus the influence of mesodermic connective tissue derivatives on the form and function of epithelium is presented. The odonto-blasts were found capable of survival as such and readily formed new dentin in transplantation; the stellate cells of the pulp were inert from the standpoint of inducing calcification.

The objective of this study was to evaluate whether demineralized bovine bone (Gen-ox®) alters bone neoformation in rats submitted to alcoholism. Forty male rats were separated into two groups of 20 rats and distributed as follows: Group E1, which received 25% ethanol and a surgical cavity filled only by a blood clot, and Group E2, which received 25% ethanol and a surgical cavity filled with Gen-ox®. The animals were euthanized at 10, 20, 40 and 60 days after surgery and necropsy was performed. The histomorphological and histometric analyses of the area of connective tissue and bone neoformation showed that the reorganization of the bone marrow and full repair of the surgical cavity in Group E1 occurred more quickly than in Group E2. It was also noted that in the final period the animals in Group E2 showed areas of connective tissue and thick bone trabeculae around the particles of the implant. It can be concluded that the use of Gen-ox® delayed the process of bone repair in alcoholic rats, although it can be used as filling material because it shows osteoconductive activity, as evidenced by bone tissue formation around the graft particles.

AbstractCadmium (Cd) and diazinon (DZN) are known to be environmental risk factors for various bone diseases including osteoporosis. Selenium (Se), an essential constituent of many antioxidant enzymes, has in higher concentrations negative effects on the bone. The present study was aimed to investigate possible changes in femoral bone of adult male rats after their acute and subchronic exposures to Cd, DZN and Se. A total of 30 male Wistar rats were randomized into three experimental groups. The rats in the group A (4-months-old) were injected intraperitoneally with a mixture of 2 mg CdCl2 kg−1, 20 mg DZN kg−1 and 2 mg Na2SeO3 kg−1 body weight and killed 36 h after xenobiotics had been injected. In the group B, young males (1-month-old) were administered with a combination of 30 mg CdCl2 L−1, 40 mg DZN L−1 and 5 mg Na2SeO3 L−1 in their drinking water, for 90 days. Ten 4-months-old males without toxicant supplementation served as a control group (C). After treatment period, detailed histological analysis of femoral bone was performed in each group. Our results revealed apparent osteoporotic symptoms (resorption lacunae, osteoporotic fractures) in rats from groups A and B. Moreover, histomorphometrical evaluation showed reduced bone vascularization (constricted primary osteons’ vascular canals and Haversian canals) and weakness mechanical properties of bones (smaller size of the secondary osteons) in these rats in comparison with those of the control group. Our study demonstrates for the first time that acute and subchronic co-administrations to Cd, DZN and Se induce evident manifestation characteristics of osteoporosis in male rats.

We studied the effect of silymarin and dimercaptosuccinic acid (DMSA), a chelating agent that was administered individually or in combination against lead (Pb) toxicity in rats. Wistar rats (200 ± 20) were randomly divided into five groups. Group A served as a control. Groups B–E were exposed to 2000 ppm of lead acetate in drinking water for 8 weeks. Group B served as a positive control. Group C received silymarin (100 mg kg−1 orally) for 8 weeks. Group D received DMSA (75 mg kg−1 orally) once daily for the last 5 days of treatment. Group E received DMSA and silymarin as groups C and D, respectively. The effect of Pb was evaluated and accordingly the treatments on blood lead levels (BLLs), renal system, and genotoxic effects were calculated using comet assay. The BLLs were significantly increased following the exposition of lead acetate. The administration of silymarin and DMSA provided reduction in BLLs. Silymarin and DMSA provided significant protection on the genotoxic effect of Pb. The toxic effect of Pb on kidneys was also studied. Our data suggest that silymarin and DMSA improve the renal histopathological lesions.

The toxicity of lead acetate (L. A.) concerned to public health disruptor due to its persistence in the environment and it has the adverse influence on the human and animal health as well. It causes physiological,biochemical, and neurological dysfunctions in humans. Histologically it has a negative effect on the liver which is considered one of the major target organs where acts as detoxification machine by elimination the toxic substance from the blood in rich with it.  As well as it affects kidneys that are the two of the most filtering organs. Therefore the present study was aimed to investigate the histopathological effect of L.A. on liver and kidney tissues in male rats. Twenty male rats involved in the study were equally and randomly divided into two groups each of them involved 10 animals. Group I (castrated rats) and Group II (control) each group received 80mg/L of lead acetate dissolved in one liter distilled water by drinking for 15 days. Histological sections showed some alterations including abnormal architecture, cell degeneration, nuclear degeneration, hyperchromatic hepatocytes, immune cells, degeneration in tubules, dilation in sinusoids, dilation in central vein of liver increased bowman's space glomerular atrophy degeneration of tubular cells in liver and kidney tissues of rats in castrated rats from control group. But the size of degenerated tissue was more severe in castrated male rats. It was concluded that the castration process could produce a hypogonadism and decreased testosterone which owns many receptors in kidney and liver may produce adverse influence with L.A. administration.
