Acute and subchronic co-administrations to cadmium, diazinon and selenium induce apparent osteoporotic symptoms in adult male rats

AbstractCadmium (Cd) and diazinon (DZN) are known to be environmental risk factors for various bone diseases including osteoporosis. Selenium (Se), an essential constituent of many antioxidant enzymes, has in higher concentrations negative effects on the bone. The present study was aimed to investigate possible changes in femoral bone of adult male rats after their acute and subchronic exposures to Cd, DZN and Se. A total of 30 male Wistar rats were randomized into three experimental groups. The rats in the group A (4-months-old) were injected intraperitoneally with a mixture of 2 mg CdCl2 kg−1, 20 mg DZN kg−1 and 2 mg Na2SeO3 kg−1 body weight and killed 36 h after xenobiotics had been injected. In the group B, young males (1-month-old) were administered with a combination of 30 mg CdCl2 L−1, 40 mg DZN L−1 and 5 mg Na2SeO3 L−1 in their drinking water, for 90 days. Ten 4-months-old males without toxicant supplementation served as a control group (C). After treatment period, detailed histological analysis of femoral bone was performed in each group. Our results revealed apparent osteoporotic symptoms (resorption lacunae, osteoporotic fractures) in rats from groups A and B. Moreover, histomorphometrical evaluation showed reduced bone vascularization (constricted primary osteons’ vascular canals and Haversian canals) and weakness mechanical properties of bones (smaller size of the secondary osteons) in these rats in comparison with those of the control group. Our study demonstrates for the first time that acute and subchronic co-administrations to Cd, DZN and Se induce evident manifestation characteristics of osteoporosis in male rats.

Cottonseed oil is an extract from the seeds of cotton plant, that is rich in protein, carbohydrate and fiber.          The objective of the research was to study the effect of cottonseed oil on the histology of the testis.          Materials and methods. Twelve adult male Wistar rats weighing 200 – 250g, were randomly sorted into four groups (I - IV), each containing three animals. Cottonseed oil was administered orally to the experimental animals by means of an orogastric tube. The rats in Group I (the control group) received standard rat pellet and oral normal saline ad libitum only. The animals in Groups II, III, and IV received 0.2 ml, 0.6 ml and 1.0 ml of cotton seed oil per body weight respectively. On the 21st day, the animals were sacrificed by cervical dislocation. The testes were excised and, then, fixed with appropriate fixatives for routine tissue processing.          Results. The results revealed an increase in the luminal diameter of seminiferous tubules, a decrease in the Leydig cell population, the disorientation of spermatogenic series and their detachment from the germinal epithelium.          Conclusions. The present study demonstrated that oral administration of cottonseed oil to male rats displayed a deleterious effect on their testis by disrupting spermatogenesis and the histological architecture of the testis.

Background: Tranylcypromine is a monoamine oxidase inhibitor. It inhibits the breakdown of dopamine, serotonin, epinephrine and other monoamines. The aim of this study was to investigate the adverse effects of trarylcypromine on the spermatogenesis in adult male rats. Methods: In this study, 50 adults male Wistar rats, weighing 200-250g, were used and divided into 5 groups of 10. Control group that was not given the treatment; the sham group received normal saline as a solvent and the experimental groups 1, 2 and 3 received 10, 20 and 40 mg/kg of trarylcypromine orally for 21 days. At the end of the trial period, the testes of rats were removed, tissue sections were prepared, stained with hematoxylin and eosin, and the histological features were examined under light microscopy. The statistical analyses performed were ANOVA and Tukey tests, using SPSS software, version 19. Results: The results demonstrated that the number of spermatogonials, primary spermatocytes, spermatids and Leydig cells in the experimental groups 2 and 3 receiving tranylcypromine significantly decreased compared to those in the control group. However, the number of Sertoli cells in the experimental groups did not change significantly at any dose of the drug compared to those in the control group (P≤0.05). Conclusion: Tranylcypromine reduced spermatogenesis and the respective indices in rat testicular tissues.

Hepatocellular carcinoma (HCC) is the most common cancer in humans. Despite advances in its treatment, liver cancer remains one of the most difficult cancers to treat. This study aimed to investigate the ameliorative action and potential mechanism of Aspergillus awamori (ASP) administration against the initiation process of liver carcinogenesis induced by diethylnitrosamine (DEN) in male Wistar rats. Seventy-two male rats were divided equally into eight groups as follows, Group 1: untreated control; Group 2: DEN (200 mg/kg bw) intra-peritoneally for the initiation of HCC; Groups 3–5: DEN + ASP at a dose of 1, 0.5, and 0.25 mg/kg bw and groups 6–8: ASP at a dose of 1, 0.5, and 0.25 mg/kg bw. Supplementation of A. awamori significantly lightened the adverse impacts induced by DEN via restoring the leukogram to normal, lowering the elevated serum aspartate aminotransferase (AST), alanine transaminase (ALT), and γ-glutamyl transferase (GGT), and alkaline phosphatase (ALP). Furthermore, it enhanced the hepatic antioxidant capacity through increasing the reduced glutathione (GSH) level and catalase (CAT) activity with a marked reduction in malondialdehyde (MDA) level. In addition, it decreased the positive GST-P foci. Likewise, a significant alteration of DEN-associated hepatocarcinogenesis occurred through inhibiting cytochrome P450 (Cyp19) and activating p53 gene expression. In conclusion, supplementation of A. awamori counteracts the negative effects of DEN, inhibits the early development of GST-P-positive foci and could be used as a new alternative strategy for its chemo-preventive effect in liver cancer. To the best of our knowledge, the present study is the first to report the hepato-protective effect of A. awamori in induced hepatocarcinogenesis.

Background: Silver nano-particles have anti-fungal properties. In the present study, silver nano-particles were synthesized by electrochemical method and its effects on the serum levels ofT3 and T4 hormones were investigated in adult male rats. Methods: In this experimental study, 28 adult male Wistar rats weighing approximately 180-220g were divided into 4 groups of 7 rats. The control group (no treatment). The experimental groups 1and 2 received silver intraperitoneal nano-particle doses of 25 and 100 mg/kg, respectively, for 14 days. The nano-aprticles had been synthesized at 75 seconds interval. The experimental group 3 received silver nano-particles that were synthesized at 300 seconds interval with an intraperitoneal dose of 25 mg/kg for 14 days. At the end of this period, blood samples were obtained from the rats’ hearts, and the serum levels of T3 and T4 hormones were measured. The results were statistically analyzed using ANOVA and Duncan tests. Results: At the completion of the study, there was no significant difference in the mean body weights in all experimental groups compared to control group. The results showed that the mean serum levels of T3 hormone in the experimental group1 increased significantly relative to the control group. However, there was no significant difference in the mean serum levels of T4 hormone in all experimental groups compared to that in the control group (P < 0.05). Conclusions: Silver nano-particles increased the serum T3 hormone level in male Wistar rats.

Hibiscus sabdariffa is a common garden plant native to warm-temperate, subtropical and tropical regions throughout the world, used in traditional medicine. It is mostly cultivated for its flowers. The calyces are used as a refrigerant in form of tea (especially in making the sorrel drink popularly known as ‘zobo’ in Nigeria), jellies and jam. Aim: This study evaluates the effects of aqueous leaves extract of Hibiscus sabdariffa on the body, testis weight, histology of the testes and sperm parameter of adult male Wistar rats. Methodology: Twenty-five adult male wistar rats of 10-12 weeks and weighing about 120-140g were divided into four groups (A, B, C and D) (five rats / group), Group A was kept as control and B, C, D were administered 250 mg, 500 mg, 1000 mg  of Hibiscus sabdarfia leaf extract per Kg body weight for 8 weeks. The rats in each group were fed with rat feed and water ad libitum. Administration was by means of an oral cannula. At the end of each experimental period, the rats were sacrificed by cervical dislocation, the testes were harvested and immediately fixed in Bouin’s fluid for histological procedure. However, as soon as the animals were sacrificed, the cauda epididymis was removed and semen analysis was carried out immediately. Results: There was a significant decrease in body weight of all the test groups at (P <0.05) and a significant decrease in the testes of the rats in group D when compared with the control group. Statistical analysis revealed decrease in sperm count, motility and viability with a significance decrease (P <0.05) in group D only as compared to control group. The histoarchitecture revealed significant degenerative changes characterized by vacuolization in the intestitium and seminiferous epithelium when compared with the control group. Conclusion: The aqueous leaves extract of Hibiscus sabdariffa appears to have adverse effects on the fertility of male rats especially when taken over a long period of time.

Background: Purslane is an edible widely distributed shrub and one of the herbs used in decoctions for the treatment of different ailments including infertility. However, there is a shortage of evidence to validate its reproductive effects. Objective: To investigate the effect of methanolic extract of Portulaca oleracea (MEPO) on the reproductive system of male rats. Materials and Methods: Fifteen 10-wk old male Wistar rats with an average weight of 183 gr were randomly divided into three groups (n = 5/each). Group A (the control group) received distilled water only; group B received 400 mg/kg MEPO; and group C received 800 mg/kg MEPO for 14 days. The animals fasted overnight after the 14th day of administration and euthanized by cervical dislocation. Blood samples, sperm, testes, and epididymis were collected for serum hormones, sperm, and histological analyses. Results: There was no significant change in the serum luteinizing hormone and testosterone levels across all groups when compared to the control. However, group C showed a significant increase (p = 0.020) in follicle-stimulating hormone levels when compared to the control. There was a significant reduction (p = 0.006) in the sperm count in group C when compared with the control group. There was also a significantly reduced (p = 0.003) sperm motility in MEPO-treated groups compared to the control. While the testis showed no abnormalities in its histoarchitecture across groups, the epididymis showed some blood congestion in MEPO-treated groups. Conclusion: Portulaca oleracea showed the ability to reduce sperm count and motility at higher doses. Key words: Portulaca oleracea, Purslane, Testis, Epididymis, Rat, Sperm motility.

Abstract. Alterations of essential elements in the brain are associated with the pathophysiology of many neuropsychiatric disorders. It is known that chronic/overwhelming stress may cause some anxiety and/or depression. We aimed to investigate the effects of two different chronic immobilization stress protocols on anxiety-related behaviors and brain minerals. Adult male Wistar rats were divided into 3 groups as follows ( n = 10/group): control, immobilization stress-1 (45 minutes daily for 7-day) and immobilization stress-2 (45 minutes twice a day for 7-day). Stress-related behaviors were evaluated by open field test and forced swimming test. In the immobilization stress-1 and immobilization stress-2 groups, percentage of time spent in the central area (6.38 ± 0.41% and 6.28 ± 1.03% respectively, p < 0.05) and rearing frequency (2.75 ± 0.41 and 3.85 ± 0.46, p < 0.01 and p < 0.05, respectively) were lower, latency to center area (49.11 ± 5.87 s and 44.92 ± 8.04 s, p < 0.01 and p < 0.01, respectively), were higher than the control group (8.65 ± 0.49%, 5.37 ± 0.44 and 15.3 ± 3.32 s, respectively). In the immobilization stress-1 group, zinc (12.65 ± 0.1 ppm, p < 0.001), magnesium (170.4 ± 1.7 ppm, p < 0.005) and phosphate (2.76 ± 0.1 ppm, p < 0.05) levels were lower than the control group (13.87 ± 0.16 ppm, 179.31 ± 1.87 ppm and 3.11 ± 0.06 ppm, respectively). In the immobilization stress-2 group, magnesium (171.56 ± 1.87 ppm, p < 0.05), phosphate (2.44 ± 0.07 ppm, p < 0.001) levels were lower, and manganese (373.68 ± 5.76 ppb, p < 0.001) and copper (2.79 ± 0.15 ppm, p < 0.05) levels were higher than the control group (179.31 ± 1.87 ppm, 3.11 ± 0.06 ppm, 327.25 ± 8.35 ppb and 2.45 ± 0.05 ppm, respectively). Our results indicated that 7-day chronic immobilization stress increased anxiety-related behaviors in both stress groups. Zinc, magnesium, phosphate, copper and manganese levels were affected in the brain.

Abstract Background Dependence and tolerance to morphine are major problems which limit its chronic clinical application. Purpose This study was aimed to investigate the attenuation effect of Cerebrolysin, a mixture of potent growth factors (BDNF, GDNF, NGF, CNTF etc,), on the development of Morphine-induced dependence and tolerance. Methods Male Wistar rats were selected randomly and divided into different groups (n=8) including: a control group, groups received additive doses of morphine (5–25 mg/kg, ip, at an interval of 12 h until tolerance completion), and groups pretreated with Cerebrolysin (40, 80 and 160 mg/kg, ip, before morphine administration). Development of tolerance was assessed by tail-flick test and the attenuation effect of Cerebrolysin on morphine-induced dependence was evaluated after injection of naloxone (4 mg/kg, ip, 12 h after the morning dose of morphine). Seven distinct withdrawal signs including: jumping, rearing, genital grooming, abdominal writhing, wet dog shake and teeth grinding were recorded for 45 min and total withdrawal score (TWS) was calculated. Results Results showed that administration of Cerebrolysin could prolonged development (10 and 14 days in administration of 80 mg/kg and 160 mg/kg Cerebrolysin) and completion (4, 10 and 14 days in administration of 40, 80 and 160 mg/kg Cerebrolysin, respectively) of tolerance. Results also indicated that administration of Cerebrolysin (40, 80 and 160 mg/kg) could significantly decreased the TWS value (62±2, 77±4 and 85±6%, respectively). Conclusion In conclusion, it was found that pretreatment with Cerebrolysin could attenuated morphine-induced tolerance and dependence.

The effect of cachexia on insulin secretion was examined in adult male rats. Isolated islets of Langerhans from Walker 256 tumor-bearing rats secreted less insulin by glucose stimuli as compared with the control group; this was accompanied by significant change in 45Ca2+ outflow rate. Reduced insulin secretion to glucose stimuli in tumor-bearing rats probably led to low insulinemia (one-third). These findings indicate that reduced insulin secretion is probably an important factor for the development of cachexia in Walker 256 tumor-bearing rats.

This study examined the effects of increasing levels of dietary fat fed isocalorically on body weight, body composition, and adipose distribution. Adult male rats were weight matched into four groups. One group that was fed a low-fat diet (12%) served as reference controls. The other three groups were fed diets of 24, 36, or 48% fat in amounts to equal the energy intake of the control group. After 6 wk, body weights of the four groups were not significantly different. Intrascapular brown fat did not differ between groups. Total body fat and adipose depot weights, however, increased in proportion to the level of fat in the diet. Total body fat and retroperitoneal and mesenteric depot weights of the 48% fat group were greater than controls (P < 0.05). Mesenteric fat in this group was also significantly increased over all other groups (P < 0.05). These results show that high-fat diets fed to adult animals cause increased body fat in the absence of significant changes in body weight and that mesenteric fat is increased disproportionately.
