The current experiment was conducted in the Fish Laboratory of Aquaculture Unit, College of Agriculture, University of Basrah from 28th January to 19th April 2021. Growth criteria of grass carp Ctenopharyngodon idella were investigated using four water temperatures (15, 20, 25 and 30 oC). Grass carp fingerlings (4.92 g) were brought from earthen ponds of the Aquaculture Unit in Al-Hartha Station for Agricultural Researches, North Basrah. The results indicated that the highest final weight (7.96 g) was reached by fishes at 15 0C, while the lowest final weight (7.22 g) was at 30 0C. Weight increments for fishes were 3.02, 2.97, 2.76 and 2.22 g for water temperatures of 15, 20, 25 and 30 0C, respectively. Fishes had the highest daily growth rate (0.0425 g/day) at 15 0C and the lowest (0.0312 g/day) at 30 0C. Fishes at 20 0C had the best feed conversion rate of 4.03, while fishes at 30 0C had the worst feed conversion rate of 5.29. It can be concluded that the best growth criteria were obtained from fishes at 15 0C and 20 0C, while the worst was observed at 30 0C.

Aquaculture has become a primary method to produce various aquatic products, and intensive aquaculture technologies have become commercially important. To improve the efficiency of intensive aquaculture per unit area without reducing the growth rate of cultured fish, the present study explored the potential of artificial substrata in ponds. Our results showed that the concentrations of total nitrogen (TN) and total phosphorous (TP) in the ponds with different stocking densities of grass carp were lower than those in the control group in most cases. Further, the feed conversion rate of grass carp was significantly reduced by introducing these artificial substrata, and the culture density could be significantly increased without reducing the growth rates of these fish. Artificial substrata also significantly enriched specific bacteria and changed the structure of the microbiota in pond water. The relative abundance of Proteobacteria was significantly increased, and bacteria closely related to N and P cycles, such as Hyphomicrobium, Chitinimonas, Legionella, Shewanella, Roseiflexus, and Planktothrix were significantly enhanced. These results showed that the artificial substratum could increase TN and TP removal in aquaculture pond water by enriching N and P cycle-related bacteria, thus significantly increasing the specific growth rate of grass carp and significantly reducing their feed conversion rate. Finally, the stocking density of grass carp and the yield per unit area of pond could be increased without reducing the growth rate.

ABSTRAKPenelitian bertujuan untuk mengetahui penggunaan fermentasi bungkil kelapa sawit dalam pakan buatan.Penelitian menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari 4 perlakuan 3 ulangan.Susunan perlakuan adalah Perlakuan A : bungkil kelapa sawit 12%, Perlakuan B : fermentasi bungkil kelapa sawit 15%, Perlakuan C : fermentasi bungkil kelapa sawit 18%, Perlakuan D : fermentasi bungkil kelapa sawit 21%. Penelitian ini dilakukan selama 60 hari untuk mengetahui laju pertumbuhan spesifikberat, laju pertumbuhan harianpanjang, konversi pakan, kelangsungan hidup dan kualitas air sebagai penunjang.Hasil penelitian menunjukkan pemberian pakan buatan dengan fermentasi bungkil kelapa sawit menghasilkan respon yang baik terhadap pertumbuhan ikan gurami. Pemberian pakan buatan dengan fermentasi bungkil kelapa sawit 18% (perlakuan C), memberikan hasil terbaik dengan laju pertumbuhan dengan rata – rata laju pertumbuhan spesifik berat 1.502%, laju pertumbuhan harian panjang 4.06%, nilai konversi pakan rata – rata 3.48 dan kelangsungan hidup rata – rata 90%.Kata Kunci : Fermentasi Bungkil Kelapa Sawit, Ikan Gurami, Laju PertumbuhanThe study aims to determine the use of fermentation residue oil palm in artificial feed .Research using a completely randomized design ( CRD ), which consists of 4 treatment three replications.The composition of the treatment is the treatment  A : 12 % of palm oil cake , Treatment B : fermented palm oil cake 15 % , Treatment C : fermented palm oil cake 18 % , Treatment D : fermentation residue palm oil 21 %.This research was conducted for 60 days to determine the specific growth rate of heavy , long daily growth rate , feed conversion , survival and quality of water as a supporter.The results showed the artificial feeding with fermented palm cake to produce a good response to the growth of carp.Feeding artificially by fermentation cake palm oil 18 % ( treatment C ) , gives the best results with a growth rate with the average - average growth rate of the specific weight of 1.502 % daily growth rate long- 4:06 % , the value of feed conversion average - average 3:48 and median survival - average of 90 % .Keywords : Fermentation Palm Kernel Oil, Fish carp, Growth Rate

The grass carp reovirus (GCRV) causes severe hemorrhagic disease with high mortality and leads to serious economic losses in the grass carp (Ctenopharyngodon idella) industry in China. Oral vaccine has been proven to be an effective method to provide protection against fish viruses. In this study, a recombinant baculovirus BmNPV-VP35-VP4 was generated to express VP35 and VP4 proteins from GCRV type Ⅱ via Bac-to-Bac baculovirus expression system. The expression of recombinant VP35-VP4 protein (rVP35-VP4) in Bombyx mori embryo cells (BmE) and silkworm pupae was confirmed by Western blotting and immunofluorescence assay (IFA) after infection with BmNPV-VP35-VP4. To vaccinate the grass carp by oral route, the silkworm pupae expressing the rVP35-VP4 proteins were converted into a powder after freeze-drying, added to artificial feed at 5% and fed to grass carp (18 ± 1.5 g) for six weeks, and the immune response and protective efficacy in grass carp after oral vaccination trial was thoroughly investigated. This included blood cell counting and classification, serum antibody titer detection, immune-related gene expression and the relative percent survival rate in immunized grass carp. The results of blood cell counts show that the number of white blood cells in the peripheral blood of immunized grass carp increased significantly from 14 to 28 days post-immunization (dpi). The differential leukocyte count of neutrophils and monocytes were significantly higher than those in the control group at 14 dpi. Additionally, the number of lymphocytes increased significantly and reached a peak at 28 dpi. The serum antibody levels were significantly increased at Day 14 and continued until 42 days post-vaccination. The mRNA expression levels of immune-related genes (IFN-1, TLR22, IL-1β, MHC I, Mx and IgM) were significantly upregulated in liver, spleen, kidney and hindgut after immunization. Four weeks post-immunization, fish were challenged with virulent GCRV by intraperitoneal injection. The results of this challenge study show that orally immunized group exhibited a survival rate of 60% and relative percent survival (RPS) of 56%, whereas the control group had a survival rate of 13% and RPS of 4%. Taken together, our results demonstrate that the silkworm pupae powder containing baculovirus-expressed VP35-VP4 proteins could induce both non-specific and specific immune responses and protect grass carp against GCRV infection, suggesting it could be used as an oral vaccine.
