Este presente estudo teve como objetivo investigar o potencial antimicrobiano, leishmanicida e citotóxico do extrato em diclorometano das partes aéreas de Mitracarpus frigidus. Além de identificar as principais classes de metabólitos especiais desse extrato. Para realização dos ensaios biológicos foram testadas nove linhagens de microrganismos (Staphylococcus aureus, Pseudomonas aeruginosa, Salmonella typhimurium, Shigella sonnei, Klebsiella pneumoniae, Escherichia coli, Bacillus cereus, Candida albicans e Cryptococcus neoformans), duas espécies de Leishmanias (L. amazonensis e L. chagasi). A citotoxicidade foi testada frente ao microcrustáceo Artemia salina. Os resultados mostraram que o extrato em diclorometano apresentou expressiva atividades leishmanicida, frente a ambas formas promastigotas das espécies de Leishmanias, e antimicrobiana, principalmente contra B. cereus e C. neoformans. O ensaio de bioautografia identificou que as classes que possuem atividade antifúngica foram alcaloides e triterpenos. Os resultados apresentados abrem perspectivas para o futuro isolamento, purificação e identificação das substâncias bioativas de Mitracarpus frigidus que possam ser utilizadas no tratamento de infecções microbianas e leishmanioses.

O presente estudo teve como objetivo extrair e avaliar o potencial antimicrobiano e citotóxico de frações ricas em alcaloides obtidas a partir das partes aéreas de Mitracarpus frigidus. O isolamento de alcaloides foi realizado por meio de extração ácido-base e cromatografia em coluna com Sephadex LH-20 como fase estacionária. A atividade antimicrobiana foi avaliada pelo método de microdiluição em caldo para determinação da concentração inibitório mínima (CIM) e também por parâmetros quantitativos antimicrobianos (atividade total, percentual de atividade e índice de susceptibilidade microbiano) contra nove linhagens de microrganismos: Staphylococcus aureus, Pseudomonas aeruginosa, Salmonella typhimurium, Shigella sonnei, Klebsiella pneumoniae, Escherichia coli, Bacillus cereus, Candida albicans e Cryptococcus neoformans. A citotoxicidade foi testada frente a Artemia salina e duas linhagens de células leucêmicas, HL60 e Jurkart. A fração mais rica em alcaloides apresentou atividade expressiva para S. aureus, S. typhimurium, B. cereus, P. aeruginosa, S. sonnei e C. neoforman com CIM abaixo de 0,100 mg/mL. As amostras não apresentaram efeito citotóxico frente a A. salina e em relação as linhagens de células tumorais, somente as frações mais ricas em alcaloides apresentaram uma moderada atividade com cerca 50 e 35% de diminuição da viabilidade celular para HL60 e Jurkart, respectivamente. Os resultados apresentados abrem perspectivas para o futuro isolamento, purificação e identificação de substâncias bioativas de M. frigidus que possam ser utilizadas, principalmente no tratamento de infecções microbianas.

Мета роботи. Здійснити синтез деяких нових піразолзаміщених 7H-[1,2,4]триазоло[3,4-b][1,3,4]тіадіазинів та провести дослідження антимікробних властивостей синтезованих сполук. Матеріали і методи. Органічний синтез, ЯМР-спектроскопія, елементний аналіз, фармакологічний скринінг. Результати й обговорення. У результаті взаємодії eтил (2Z)-хлоро(фенілгідразоно)ацетатів з ацетилацетоном було отримано етил 4-ацетил-5-метил-1-феніл-1H-піразол-3-карбоксилати. Зазначені сполуки піддали бромуванню, що дозволило одержати цільові бромкетони. Синтезовані на даній стадії етил 1-арил-4-(бромацетил)-5-метил-1Н-піразол-3-карбоксилати було введено у взаємодію з 4-аміно-5-арил(гетарил)-2,4-дигідро-3Н-1,2,4-триазол-3-тіонами з подальшим формуванням 1,3,4-тіадіазольного циклу та отриманням відповідних етил 1-арил-4-{3-арил(гетарил)-7H-[1,2,4]триазоло[3,4-b][1,3,4]тіадіазин-6-іл)}-5-метил-1H-піразол-3-карбоксилатів. Структура синтезованих сполук підтверджена даними елементного аналізу та ЯМР спектроскопією. В рамках міжнародного проекту "The Community for Antimicrobial Drug Discovery" (CO-ADD) за підтримки Wellcome Trust (Великобританія) і університету Квінсленда (Австралія) для синтезованих сполук здійснено скринінг антимікробної активності. Як тестові мікроорганізми використовували п'ять штамів бактерій: Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 700603, Acinetobacter baumannii ATCC 19606, Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 43300 та двох штамів грибків: Candida albicans ATCC 90028 і Cryptococcus neoformans ATCC 208821. Встановлено, що досліджувані сполуки виявляють різноманітну дію, від практично повної її відсутності до виразного антимікробного ефекту. Висновки. Здійснено синтез 12 нових етил 1-арил-4-{3-арил(гетарил)-7H-[1,2,4]триазоло[3,4-b][1,3,4]тіадіазин-6-іл)}-5-метил-1H-піразол-3-карбоксилатів. Зазначені речовини отримані шляхом взаємодії відповідних етил 1-арил-4-(бромацетил)-5-метил-1Н-піразол-3-карбоксилатів з 4-аміно-5-арил(гетарил)-2,4-дигідро-3Н-1,2,4-триазол-3-тіонами. Дослідження антимікробної активності синтезованих сполук демонструють потенціал пошуку антимікробних агентів серед зазначеного класу сполук.

The aim of the study was to determine antimicrobial activity of rhaponticum and shrubby cinquefoil extracts. Material and Methods. Ethanol extract from the leaves of rhaponticum (Rhaponticum carthamoides D.C. Iljin) and shrubby cinquefoil (Potentilla fruticosa L.) was produced at the Department of Food Technology, Kaunas University of Technology. The antimicrobial activity of the viscous extract or rhaponticum and shrubby cinquefoil was evaluated using standard microorganism cultures (bacteria Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 33499, Pseudomonas aeruginosa ATCC 27853, Proteus mirabilis ATCC 12459, Bacillus subtilis ATCC 6633, Bacillus cereus ATCC 8035 and fungi Candida albicans ATCC 60193). The minimum inhibitory concentration (MIC) of the examined preparations was determined. Results. Both studied preparations – rhaponticum (Rhaponticum carthamoides D.C. Iljin) and shrubby cinquefoil (Potentilla fruticosa L.) – demonstrated similar antimicrobial activity. The highest sensitivity to the studied preparations was observed in microbes with eukaryotic cell structure: Candida albicans, which is a fungus, and a spore-forming prokaryotic bacterium, Bacillus cereus. The highest resistance was observed in Escherichia coli and Klebsiella pneumoniae. Conclusions. The studied preparations – viscous extracts of rhaponticum and shrubby cinquefoil – are substances with antimicrobial activity against gram-positive (Staphylococcus aureus and Enterococcus faecalis) and gram-negative (Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Proteus mirabilis) bacteria, spore-forming bacteria (Bacillus subtilis and Bacillus cereus), and fungi (Candida albicans).

Ecuador es uno de los países más ricos en biodiversidad y endemismo del mundo, y cerca de 3200 especies de plantas tienen usos medicinales. El objetivo de esta investigación fue evaluar la composición química y el efecto antimicrobiano del aceite esencial de Lasiocephalus ovatus Schltdl. (Asteraceae) colectada en la provincia de Chimborazo, Ecuador. Las partes aéreas de L. ovatus fueron sometidas a hidrodestilación para obtener el aceite esencial, el cual fue analizado mediante cromatografía de gases acoplada a espectrometría de masas. La actividad fue evaluada frente a cinco bacterias y una levadura usando la técnica de microdilución en caldo en microplacas de 96 pozos. El rendimiento del aceite fue de 0,05 % y 27 compuestos fueron identificados, representando 95,45 % de la composición total con un elevado contenido de monoterpenos oxigenados (52,17 %). Los compuestos mayoritarios fueron alcanfor (40,48 %) y 1,2,5,5-tetrametil-1,3-ciclopentadieno (11,90 %), seguido por p-menta-1,5-dien-8-ol (5,23 %) y 1,6-dimetilhepta-1,3,5-trieno (4,69 %). Las bacterias más sensibles a la acción del aceite fueron Staphylococcus aureus ATCC 25923 y Escherichia coli ATCC 25922 con concentraciones mínimas inhibitorias de 200-400 μg/mL y bactericidas de 800 μg/mL. La inhibición antimicrobiana frente a las bacterias Enterococcus faecalis ATCC 29212, Pseudomonas aeruginosa ATCC 27853, Klebsiella pneumoniae ATCC 700603 y la levadura Candida albicans ATCC 10231 fue baja, con un rango de concentración mínima inhibitoria de 800 a 6400 μg/mL. Este reporte representa un primer análisis de la actividad antimicrobiana del aceite de L. ovatus, por lo tanto, una contribución importante al estudio del género Lasiocephalus.

Objective. To evaluate the antimicrobial activity of soft and purified propolis extracts. Study object and methods. Antimicrobial activity of soft and purified propolis extracts was determined with reference cultures of Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 33499, Pseudomonas aeruginosa ATCC 27853, Proteus mirabilis ATCC 12459, Bacillus subtilis ATCC 6633, Bacillus cereus ATCC 8035, and fungus Candida albicans ATCC 60193. Microbiological tests were performed under aseptic conditions. Minimum inhibitory concentration (MIC) – the highest dilution of preparation (the lowest concentration of preparation) that suppresses growth of reference microorganisms – was determined. Results. Concentration of phenolic compounds in soft propolis extract that possesses antimicrobial activity against gram-positive (Staphylococcus aureus, Enterococcus faecalis) and gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa, and Proteus mirabilis) is 0.587±0.054 mg and 0.587±0.054–0.394±0.022 mg (P>0.05) and in purified propolis extract – 0.427±0.044 mg and 0.256±0.02 mg (P>0.05). Klebsiella pneumoniae is most resistant to soft propolis extract when the concentration of phenolic compounds is 1.119± 0.152 mg and to purified propolis extract when the concentration of phenolic compounds is 1.013±0.189 mg (P>0.05). Spore-forming Bacillus subtilis bacteria are more sensitive to soft and purified propolis extracts when the concentration of phenolic compounds is 0.134±0.002 mg and 0.075±0.025 mg, respectively, and Bacillus cereus – when the concentration is 0.394±0.022 mg and 0.256±0.02 mg (P>0.05). Sensitivity of fungus Candida albicans to soft and purified propolis extracts is the same as Bacillus subtilis. Encapsulated bacterium Klebsiella pneumoniae is most resistant to antimicrobial action of soft and purified propolis extracts as compared with gram-positive Staphylococcus aureus and Enterococcus faecalis bacteria (P<0.05), gram-negative Escherichia coli, Pseudomonas aeruginosa, and Proteus mirabilis (P<0.05), sporeforming Bacillus subtilis and Bacillus cereus bacteria (P<0.05), and fungus Candida albicans (P<0.05). There is no statistically significant difference between antimicrobial effect of soft propolis extract and purified propolis extract on gram-positive bacteria, gram-negative bacteria, spore-forming bacteria, encapsulated bacteria, and Candida fungus. Conclusions. Soft and purified propolis extracts possess antimicrobial activity. They could be recommended as natural preservatives in the manufacture of pharmaceutical products.

Nosocomial infection is a rising problem in developing countries. Microorganisms Isolated from three private and public hospital environmental surfaces in Akure Metropolis, Ondo State, Nigeria were investigated in this study. Bacterial and fungal organisms were isolated and compared among the three hospitals. The study revealed that bacteria were the most predominant microorganisms found in the hospital environmental surfaces than fungi. Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Bacillus cereus were the bacterial isolates while fungi include Aspergillus fumigatus, Aspergillus niger and Candida albicans.  Staphylococcus aureus was found to be predominant bacteria but Aspergillus funmigatus was the predominant fungi. The result showed that the microbial loads of the public hospitals were higher than that of the private hospitals. The bacteria load of the male ward was found to be higher than that of the female ward while the fungal loads of each of the hospital environmental surfaces of female were higher than that of the male. The study revealed that bacteria were the most predominant microorganisms found in the hospital environment than fungi. Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Bacillus cereus were the bacterial isolates while fungi include Aspergillus fumigatus, Aspergillus niger and Candida albicans.  Staphylococcus aureus was found to be predominant bacteria. All the hospital environmental surfaces were contaminated with one or more microorganisms in the course of the research.

Decorating photocatalysts with noble metal nanoparticles (e.g., Pt) often increases the catalysts’ photocatalytic activity and biomedical properties. Here, a simple and inexpensive method has been developed to prepare a Pt-Ag3PO4/CdS/chitosan composite, which was characterized and used for the visible light-induced photocatalytic and antibacterial studies. This synthesized composite showed superior photocatalytic activity for methylene blue degradation as a hazardous pollutant (the maximum dye degradation was observed in 90 min of treatment) and killing of Gram positive bacterial (Staphylococcus aureus and Bacillus cereus) as well as Gram negative bacteria (Klebsiella pneumoniae, Salmonella typhimurium, Escherichia coli, and Pseudomonas aeruginosa) under visible light irradiation. The antibacterial activity of CdS, CdS/Ag3PO4, and Pt-Ag3PO4/CdS/chitosan against E. coli, Pseudomonas aeruginosa, Salmonella typhimurium, Klebsiella pneumoniae, Staphylococcus aureus, and Bacillus cereus showed the zone of inhibition (mm) under visible light and under dark conditions at a concentration of 20 µg mL−1. Furthermore, the cell viability of the CdS/chitosan, Ag3PO4, Ag3PO4/CdS/chitosan, and Pt-Ag3PO4/CdS/chitosan were investigated on the human embryonic kidney 293 cells (HEK-293), Henrietta Lacks (HeLa), human liver cancer cell line (HepG2), and pheochromocytoma (PC12) cell lines. In addition, the results indicated that the photodegradation rate for Pt-Ag3PO4/CdS/chitosan is 3.53 times higher than that of CdS and 1.73 times higher than that of the CdS/Ag3PO4 composite. Moreover, Pt-Ag3PO4/CdS/chitosan with an optimal amount of CdS killed large areas of different bacteria and different cells separately in a shorter time period under visible-light irradiation, which shows significantly higher efficiency than pure CdS and other CdS/Ag3PO4 composites. The superb performances of this composite are attributed to its privileged properties, such as retarded recombination of photoinduced electron/hole pairs and a large specific surface area, making Pt-Ag3PO4/CdS/chitosan a valuable composite that can be deployed for a range of important applications, such as visible light-induced photocatalysis and antibacterial activity.

L’activité antimicrobienne des huiles essentielles de galles de Pistacia atlantica récoltées dans trois régions (Aïn-Oussara, Laghouat et Kheneg) a été évaluée sur sept souches pathogènes (Escherichia coli, Bacillus cereus, Pseudomonas aeruginosa, Staphylococcus aureus, MRSA+, Salmonella typhi et Candida albicans), en utilisant les méthodes de diffusion sur gélose, de microdilution et celle de microatmosphère (seulement pour Candida albicans). Les huiles essentielles ont révélé un effet antimicrobien remarquable contre les micro-organismes testés, avec des valeurs de CMI et de CMB variant de 0,13 à 7,99 μl/ml et de 0,25 à 7,99 μl/ml, respectivement. Une activité importante a été observée pour les échantillons de la station Kheneg contre l’espèce Candida albicans avec une zone d’inhibition de 68 mm (50 μl). Ces résultats suggèrent que les huiles essentielles testées pour leur activité antimicrobienne peuvent être classées comme bactéricides.

Objective: In vitro investigated the potential of methanol extracts of micro-propagated C. orchiodes in the antimicrobial property against the three gram-negative bacteria, two gram-positive and one fungal filament.Methods: The micro propagated callus methanol extract was examined against Escherichia coli, Proteus vulgaris, Salmonella typhimurium, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus and Candida albicans. The zone of inhibitions are determined at 10 mg/ml concentration of methanol extracts of callus on agar well plate and MIC against tested microorganism.Results: The highest antibacterial activity recorded in Staphylococcus aureus Bacillus cereus and followed by Candida albicans. Antibacterial activity of leaf extracts of A. reticulata was also significant against the tested microorganisms Escherichia coli, Salmonella typhi, Proteus vulgaris, Pseudomonas aeruginosa compared to ciprofloxacin.Conclusion: Based on the above observations, these extracts were further evaluated for their effect on microorganisms causing infections like typhoid fever, urinary tract infections, septicemia, toxic shock syndrome, skin infection, nosocomial infection, arthritis and diarrhoea. The results also suggest that these plants serve a therapeutic purpose in the treatment bacterial infections.

<p>Os cateteres intravenosos são de uso amplamente difundido em hospitais e por estarem em contato direto com a circulação sanguínea, tornam-se um potencial risco para a ocorrência de contaminação e infecção local e sistêmica, aumentando a mortalidade, o tempo de internação e consequentemente custos hospitalares. O presente estudo teve como objetivo realizar levantamento epidemiológico da prevalência de bactérias em pontas de cateteres provenientes de sete hospitais do município de Cascavel - Paraná. Foi realizado um estudo transversal por meio da coleta de dados em laudos emitidos por um laboratório clínico privado do mesmo município, entre os anos de 2011 a 2014. Verificou-se positividade em 21,68% dos laudos analisados (248). O grupo dos bacilos Gram negativos (BGN) foi o mais prevalente, sendo as principais bactérias encontradas <em>Escherichia coli</em> e <em>Klebsiella pneumoniae</em> (84 laudos), seguidos de BGN não-fermentadores (79 laudos) e cocos Gram positivos (67 laudos). Leveduras foram encontradas em 18 laudos. <em>Staphylococcus aureus</em> apresentaram uma resistência de 31,81% para oxacilina, sendo para <em>Staphylococcus</em> coagulase-negativa resistência de 86,96%. <em>Klebsiella pnemoniae</em> apresentou discreta resistência para ertapenem (9,52%), imipenem (7,14%) e meropenem (7,14%), sugerindo presença de carbapenemase. Espécies de <em>Enterobacter</em> foram resistentes a diversos antimicrobianos, não constatado resistência aos carbapenêmicos ertapenem, imipenem e meropenem. <em>Acinetobacter baumanii</em> e <em>Pseudomonas aeruginosa</em> apresentaram elevada resistência para imipenem e meropenem (77,14% e 45,16%; 80% e 51,61%, respectivamente). <em>Candida albicans</em> apresentou 100% de suscetibilidade aos antifúngicos utilizados. Esses resultados demonstram a grande importância no manejo eficiente do controle de infecção hospitalar e uso responsável de antimicrobianos por hospitais.</p>
