Synthesis and Anticonvulsant activity of new 5-((1-(2-(4-substituted phenacyl bromide)-2-oxoethyl)-2-phenyl-1H-indol-3yl) methylene) pyrimidine-2, 4, 6(1H, 3H, 5H)-trione Derivatives

In the current scenario we have plant to study to synthesize novel indole derivatives (4a-d) and (5a-b) which have the target activity like anti-convulsion and anti microbial etc. In the primary phase, we synthesized 2-phenyl-1H-indole reacted with Phosphorus oxychloride and N, N’-dimethylformamide by using vilsmeier reagent formylation of a new compound 2-phenylindole-3-carbaldehyde (2). The 2-phenylindole-3-carbaldehyde was reacted with substituted phenacyl bromide, K2CO3 and TBAB (Tetra butyl ammonium bromide) into the presence of N, N’-dimethylformamide to gives [1-(2-(4-substituted phenacyl bromide)-2-oxoethyl)-2-phenylindole-3-carbaldehyde (3a-d). The compound [1-(2-(4- substituted phenacyl bromide)-2-oxo-ethyl)-2-phenylindole-3-carbaldehyde was permitted to mixed with various substituted phenacyl bromide gives corresponding indole derivatives. The indole derivatives (3a-d) were reacted with barbituric acid and thiobarbituric acid dissolved into the methanol at reflux for 4h to gives afforded compounds (4a-d) and (5a-b). The final indole barbituric derivatives (4a-d) and (5a-b) were synthesized from substituted phenacyl bromide by the react with methanol and recrystalized from ethanol. The last compound has been completed on the basis of spectra FT-IR and 1H NMR. All the value of FT-IR, 1H NMR, Solubility and TLC were considered to be prominent. The pharmacological screening through ear electrode induced current 50mA for 0.2 seconds in electro-convulsiometer for anticonvulsant activity. The synthesize compounds were establish to be 4b, 4c, 4d, and 5a. The compound 4c and 5a were established to be the most potent compound through compare to standard drugs phenytoin sodium.

A series of 2, 5-disubstituted - 1, 3, 4 oxadiazoles (4a-o) were synthesized on refluxing hydrazine carbothioamides with iodine and potassium iodide in ethanolic sodium hydroxide solution starting from methyl-3-amino-4-hydroxy benzoate via synthesis of an intermediate methyl-2-substitutedaryl-1, 3-benzoxazole-5-carboxylates and 2-substitutedaryl-1, 3-benzoxazole-5-carbohydrazides. The newly synthesized compounds were characterized on the basis of spectral (FT-IR, 1H-NMR, MS) and elemental analysis. All these compounds were screened for anticonvulsant activity using Maximal Electroshock (MES) and subcutaneous pentylenetetrazole (scPTZ) method. Anticonvulsant activity was shown by majority of the synthesized compounds when given i.p. to mice. Among the tested compounds 4e, 4j and 4o were considered to have potent anticonvulsant activity comparable to that of standard drugs Phenytoin and Carbamazepine. Compounds 4e, 4g, 4h, 4i, 4k and 4m passed the rota rod test successfully without any sign of neurological deficit.

In the work we presented the behavior of 5-(4-(4-X-phenylsulfonyl)phenyl)-4-(n-propyl)-2H-1,2,4-triazole-3(4H)-thiones (X= Cl or Br) with some alkylation agents. Thus, new S-alkylated 1,2,4-triazole derivatives were synthesized by reaction of the corresponding 1,2,4-triazole-3-thione derivatives with different �-halogenated compounds (ethyl bromide, ethyl chloroacetate or phenacyl bromide), in basic medium. The structures of synthesized compounds were elucidated by spectral data (1H-NMR, 13C-NMR, mass spectrometry) and elemental analysis.

Background & Objective: A series of novel 3-Substituted-1,3,4,5-Tetrahydro-2H-benzo [b] azepine-2-one Derivatives (4, 5, 7, 10, 12, 5a-j, 8a-e) were synthesized from 1,2,3,4-Tetrahydro-1- naphthalenone. The structures of these compounds were confirmed by IR, 1H NMR, 13C NMR, MASS spectra and elemental analysis. Their anticonvulsant activity was evaluated by the maximal electroshock (MES) test, subcutaneous pentylenetetrazol (scPTZ) test, and their neurotoxicity was evaluated by the rotarod neurotoxicity test. Compound 4 showed the maximum anticonvulsant activity against the maximal electroshock test (ED50=26.4, PI =3.2) and against the subcutaneous pentylenetetrazol test (ED50=40.2, PI =2.1). Conclusion: Possible structure-activity relationship was discussed.

Reactions of the labile complex (Me3tach)M(CO)3 (Me3tach = 1,3,5-trimethyl-1,3,5-triazacyclohexane; M = W, Mo) with bis(phenylthio)alkanes ( = PhS(CH2)nSPh), (2:3 molar ratio) in CH2Cl2 as solvent (at 35 oC M = W, 0 oC M = Mo) afforded in good yields bimetallic tricarbonyl complexes triply- bridged by bis(phenylthio)alkanes of the type [M(CO)3]2(μ-PhS-(CH2)n-SPh)3 [n = 3 (a), 4 (b), 5 (c), 6 (d); M = W (1), Mo (2)]. These new triply-bridged complexes were characterized by 1H NMR, FT-IR and elemental analysis. Additionally, it has been observed that the thermodynamic stability of some of these new complexes in chlorinated hydrocarbons depends mainly on the nature of the metal center and varies in the order W >> Mo.

تم في هذا البحث تحضير وتشخيص بعض مركبات 5،2-ثنائي المعوض-4،3،1-اوكسادايازول المشتقة من حامض الدايكلوفيناك (H1). حيث تضمن العمل تحضير الاستر المثيلي لحامض الدايكلوفيناك (H2) من مفاعلته مع الميثانول بطريقة فشر ثم تحويل الاستر المذكور الى هيدرازايد الحامض المقابل (H3) ثم مفاعلة هذا الهيدرازايد مع بعض الحوامض الكاربوكسيلية بوجود اوكسي كلوريد الفسفور للحصول على مشتقات 5,2-ثنائي المعوض-43،1- اوكسادايازول (H4-H7). ثم مفاعلة المركب (H7) مع بعض الامينات الأولية للحصول على مركبات 5,2-ثنائي المعوض-4،3،1-اوكسادايازول امين (H8-H12). وقد شخصت هذه المركبات باستخدام تقنيات FT-IR و 1H-NMR و 13C-NMR علاوة على قياس درجات الانصهار لها. أظهرت نتائج التقييم الاولية لفعاليتها البيولوجية ضد نوعين من البكتريا، الاولى (Pseudomonas aeruginosa)، والثانية (Staphylococcus aureus) بأن للمركبات المحضرة ( H4 ، H6، H11، H12) فعالية تثبيطية متوسطة غالباً ضد هذين النوعين، بينما لوحظ بأن للمركبات (H7 ، H8، H9، H10) فعالية تثبيطية جيدة ضدهما على الاغلب، وكلاهما مقارنة بالمثبط القياسي المستعمل. وقد أظهرت نتائج التقييم الاولية للفعالية البيولوجية ضد نوعين من البكتريا، الاولى (Pseudomonas aeruginosa)، والثانية (Staphylococcus aureus) بأن للمركبات المحضرة ( H4 ، H6، H11، H12) فعالية تثبيطية متوسطة غالباً ضد هذين النوعين، بينما لوحظ بأن للمركبات (H7 ، H8، H9، H10) فعالية تثبيطية جيدة ضدهما على الاغلب، وكلاهما مقارنة بالمثبط القياسي المستعمل.

In this paper, Ca2+-montmorillonite (Ca2+-Mt) was modified with cationic surfactant hexadecyl trimethyl ammonium Bromide (CTAB) directly, without changing Ca2+-Mt into Na+-montmorillonite (Na+-Mt). X-ray diffraction (XRD), thermogravimetric analysis (TGA), Fourier transform infrared spectroscopy (FT-IR), contact angle tests and rotary viscosity tests were used to characterize the structure and property of the organo-montmorillonite. The results of XRD show that interlayer space (d001) of the product is 4.05nm. The results of TGA indicate the ratio of weight loss of the organic components in the product is 37.7%. The results of FT-IR indicate there are organic components and hardly any water molecules in the product. All these findings indicate that the CTAB has intercalated into the galleries of Ca2+-Mt. Contact angle tests shows the product is well hydrophobic and the hydrophobic property is as good as that of organo-montmorillonite modified from Na+-Mt. Rotary viscosity tests show that the viscosity of the product at the rotational speed of 6R/min is 1226mPa·s, much better than that of the organo- montmorillonite modified from Na+-Mt which is 1070 mPa·s. The thixotropy index of the product is 9.22, much better than that of the organo-montmorillonite modified from Na+-Mt which is 5.88. It can be concluded that organo-montmorillonite with perfect interlayer expansion and thixotropic property was prepared by modifying Ca2+-Mt with CTAB directly.
