In vitro Antioxidant potential, Total Phenolic and Flavonoid contents in Justicia gendarussa leaf extracts

Free radicals are triggered in human body by routine metabolic pathways which are highly reactive substance entangled in various physiological functions but excess production may impart with multitude diseases such as cancer, arthritis, inflammation and various neurodegenerative disorders. As a result, previous findings supported that various photochemical constituents present in natural plants exerts prominent role in defense mechanism against rate of excess free radicals’ production. The present study was carried out to investigate phytochemical composition, quantification studies and in vitro antioxidant potential of Justicia gendarussa (JG) various solvent extracts. JG belongs to the family Acanthaceace and used as traditional healers in various ailments. JG plant material was collected, dried, powdered and subjected for soxhlation to prepare ethanolic, ethyl acetate, aqueous and n-hexane extracts. Screening of Active constituents by various qualitative tests, estimation of total phenolic (TPC) and Total flavonoid content (TFC) and in vitro antioxidant potential of plant extracts were performed by DPPH, Lipid peroxidation, Nitric oxide and Superoxide radical scavenging methods. Present investigation reveals that, presence of flavonoids, phenols, amino acids, glycosides, cardiac glycosides, proteins, carbohydrates, alkaloids, steroids, terpenoids and tannins. TPC and TFC of ethanol extract were found to be 9.47 ± 0.0216 mg Gallic acid equivalent (GAE/g) and 97.6 ± 0.0342 mg Rutin equivalent/g (RUE/g). Among all the analyzed extracts ethanolic extract of JG possessed high radical neutralizing capacity and for DPPH (IC50 32µg/ml), Lipid Peroxidation (28 µg/ml), Nitric oxide (30 µg/ml) and superoxide anion radicals (39 µg/ml) respectively and found to be more effective as ascorbic acid (26 µg/ml) which was used as standard. Overall results of the study concluded that JG leaf extracts possess beneficial Phytoconstituents, high phenol and flavonoid contents and potential antioxidant activity, which could be a viable source of natural antioxidants for treating various degenerative disorders.

Objective: To evaluate the preliminary phytochemical content and antioxidant potential of the hydroalcoholic leaf extracts of Hemionitis arifolia. Methods: Total phenolic, flavonoid and alkaloid contents were evaluated using spectrophotometric methods. The free radical scavenging activity of the leaf hydroalcoholic extract were evaluated against DPPH+, ABTS+, Reducing power assay and nitric oxide assay were determined. Results: The hydroalcoholic concentrate of H. arifolia uncovered the most elevated polyphenol content when contrasted and the other phytoconstituents. Absolute phenol content of the hydroalcoholic separate was observed to be 31.78%, flavonoid content is 1.02% and Alkaloid content is 30.40% individually. The Solvent concentrates showed huge cell reinforcement movement, with hydroalcoholic extract. ABTS Assay, DPPH assay, Reducing power assay and Nitric oxide assay where the Inhibition concentration were 667.75µg/ml, 734.25 µg/ml, 791.58 µg/ml and 899.67 µg/ml. Conclusion: This study suggests that hydroalcoholic leaf extracts of H. arifolia could be a potential source of natural antioxidant and justifies its use in ethno-medicine.

The current line of investigation was focused at perusing the presence of phytochemical constituents, investigation of total phenol and flavonoid content, the antioxidant potential of various extracts of Caralluma adscendens whole plant using various in-vitro assays. The dried plant powder was extracted with various solvents based on polarity (Pet ether, Chloroform, Ethyl acetate, Ethanol and Aqueous) by hot continuous extraction in Soxhlet's apparatus and Extracts were dried. Phytoconstituents present in each extract was examined by performing preliminary phytochemical screening. Total Phenolic Content (TPC), Total Flavonoid Content (TFC) and Antioxidant potential for crude extracts were studied by DPPH, nitric oxide scavenging and FRAP methods. The total phenolic content and flavonoid content of Ethanolic extract of plant was found to be 80.08±0.629mg and 70.88±1.170mg of GAE and Quercetin equivalents respectively. The Ethanolic extract exhibited potent antioxidant activity as determined by 2,2-diphenyl-1-picrylhydrazyl(DPPH), nitric oxide scavenging and ferric reducing antioxidant power assays(FRAP) than the other extracts. The IC50 values for the Ethanolic extract of Caralluma adscendens was found to be 214.765±0.224 µg/ml and 215.928±0.506µg/ml by DPPH and nitric oxide scavenging assays respectively.

Introduction. Propolis (bee glue) is a natural product collected by honeybees from buds of various trees, shrubs and other plant species. Extracts of propolis possess numerous biological activities, including antioxidant, antibacterial, antifungal and anticancer. For this reason, propolis is currently used in many applications, such as preparations for cold syndrome, dermatological preparations or as a constituent of nutritional supplements and health food. The chemical composition of this natural material is very complex and depending on many factors, including method of extraction and selection of the solvent for the extraction process. Aim. The aim of the study was to determine concentration of selected phenolic compounds (flavonoids and phenolic acids) in extract of Polish propolis and estimate its antioxidant activity and effect on human red blood cells. Material and methods. In the propolis extract was determined concentration of 14 flavonoids and 9 phenolic acids using ultra-performance liquid chromatography equipped with a photodiode detector and a triple quadrupole mass spectrometer. The antioxidant potential of propolis extract was evaluated applying DPPH˙ free radical scavenging activity assay and Fe3+ reducing power assay. Moreover, the cytotoxicity and cytoprotective potential of propolis extract was estimated using human erythrocytes in vitro. Results. The propolis extract contained high concentration of pinocembrin, galangin, chrysin, apigenin, kaempferol, coumaric acid and cinnamic acid. It exhibited also high antioxidant potential. The antiradical activity of examined propolis extract was equal to 75% approx. activity of both standard antioxidants used in the study, namely Trolox and BHT. The reducing power of extract was equal to 65% approx. of Trolox and 80% of BHT, respectively. The propolis extract had no hemolytic activity, moreover, effectively protected human erythrocytes against free radicals-induced damage in vitro. Conclusions. The results of this study indicate that the propolis extract of national origin is a rich source of flavonoids and phenolic acids. Therefore, the propolis extract possesses a high antioxidant potential and can protect erythrocytes against free radicals-induced oxidative hemolysis.

Rose petals have been applied as food additives in teas, cakes and flavor extracts. The aim of this research study was to explore and reveal the antioxidant potential of aqueous extracts of rose petals belonging to nine genotypes of rose (wild as well as hybrid). The in vitro antioxidant activities of roses were studied by lipid peroxidation assay, DPPH radical scavenging assay, iron chelation assay, phosphomolybdenum reduction assay and total phenolic and flavonoid contents. The aqueous extract showed inhibition against lipid peroxidation (TBARS), induced by prooxidants (10 µM FeSO4) in mice liver homogenate. The free radical scavenging activities of the extracts were determined by scavenging of the DPPH radical. Extracts also showed metal chelating activities and high antioxidant activity in the phosphomolybdenum assay. The high content of phenolics and flavonoids detected in aqueous extracts may be responsible for the antioxidant activity. Amongst the different rose genotypes, screened, Rosa moschata (musk rose) was found to carry slightly higher antioxidant potential, owing to its higher phytochemical content.

In order to facilitate the quality control of Trifolium repens flowers and leaves, a RP-HPLC method with UV detection was developed for the simultaneous quantitative determination of flavonols and isoflavones. The total flavonoid and phenolic (TPC) contents were determined spectrophotometrically in a visible part of the light spectrum at 425 and 760 nm, respectively. Additionally, evaluation of the antioxidant properties of the plant materials was performed using the DPPH in vitro test. The results showed that the flowers are the richest source of phenolics ranging from 28.7 to 38.8 mg GAE/g, and flavonoids, calculated for hyperoside, up to 20 mg HP/g, which hydrolyzed mainly to flavonols (the quercetin level greater than 6 mg/g). T. repens is poor in isoflavones; similar quantities of ca. 0.2 mg/g were detected in the flowers and leaves. The flower and leaf extracts showed antioxidant activity towards DPPH with EC50 values ranging from 72.3 to 179.3 μg/mL. Significant linear correlations were found between antioxidant potentials of the studied plant materials and total phenolic and flavonoid contents determined by HPLC and spectrophotometric methods ( R2 in the range of 0.97 - 0.99).

AbstractThe world's population is getting older. By 2050 it is projected that 22% of the global population will be over 60 years of age. Aging is characterized by a decline in physiological functions and an increase in malnutrition and associated disease. Oxidative stress is associated with several age-related conditions, including cardiovascular disease, obesity, neurodegenerative disease and cancer. The negative effects of reactive oxygen species (ROS) are neutralized by antioxidants, which can be endogenously synthesised or consumed through the diet. Cheese is a nutritious dairy food and previous research has highlighted the antioxidant potential of some cheese types. Physiological changes associated with age can impact digestion processes, thus the ability to efficiently release bioactive compounds, including antioxidants, may be impaired with age. Cheese products varying by milk source, texture and fat content were subjected to simulated gastrointestinal in vitro digestion (SGID) using a modified model to account for physiological changes associated with aging. The antioxidant potential of undigested and digested cheese (n = 11) was determined in vitro using DPPH radical scavenging, ferric reducing antioxidant power (FRAP) and total phenolic content (TPC) assays. Cellular antioxidant potential was also investigated using the reduced glutathione assay (GSH) in the human macrophage cell line (U937). Experiments were carried out in triplicate. Statistical analysis by T-test demonstrated that digestion significantly increased (P < 0.05) FRAP and TPC of all cheese products analysed. Of the eleven cheese products investigated, goats’ cheese displayed a significant increase (P < 0.05) in radical scavenging properties (89.8% inhibition). Low fat white cheddar had significantly higher (P < 0.05) GSH levels compared to full fat white cheddar (0.16 ± 0.01, 0.11 ± 0.01 μmol GSH/mg protein, respectively). Findings from this study suggest that despite physiological changes associated with age, cheese digestates retain, and in some cases, possess higher antioxidant potential than undigested samples. Cheese demonstrates potential as a functional food for the elderly by neutralizing the negative effects associated with age-related oxidative stress.

Mallotus philippinensisis an important source of molecules with strong antioxidant activity widely used medicinal plant. Previous studies have highlighted their anticestodal, antibacterial, wound healing activities, and so forth. So, present investigation was designed to evaluate the total antioxidant activity and radical scavenging effect of 50% ethanol fruit glandular hair extract (MPE) and its role on Human Erythrocytes. MPE was tested for phytochemical test followed by its HPLC analysis. Standard antioxidant assays like DPPH, ABTS, hydroxyl, superoxide radical, nitric oxide, and lipid peroxidation assay were determined along with total phenolic and flavonoids content. Results showed that MPE contains the presence of various phytochemicals, with high total phenolic and flavonoid content. HPLC analysis showed the presence of rottlerin, a polyphenolic compound in a very rich quantity. MPE exhibits significant strong scavenging activity on DPPH and ABTS assay. Reducing power showed dose dependent increase in concentration absorption compared to standard, Quercetin. Superoxide, hydroxyl radical, lipid peroxidation, nitric oxide assay showed a comparable scavenging activity compared to its standard. Our finding further provides evidence thatMallotusfruit extract is a potential natural source of antioxidants which have a protective role on human Erythrocytes exhibiting minimum hemolytic activity and this justified its uses in folklore medicines.

Abstract Background Oxidation is believed to play a vital role in the pathogenesis of diabetes mellitus by lipid peroxidation; DNA and protein damage leads to the development of vascular complications like coronary heart disease, stroke, neuropathy, retinopathy, and nephropathy. The herbal preparations are complementary and alternative medicines to allopathic drugs which are believed to cause adverse events. Therefore, the current study was aimed to identify the novel plants, which belong to the genera Argyreia (Argyreia pierreana (AP)) and Matelea (Matelea denticulata (MD)), and assess the aqueous and ethanolic leaf extracts for in vitro antioxidant and antidiabetic potential by DPPH, OH•, superoxide, and glucose uptake and gene expression (GLUT-4 and PPARγ) studies using the L-6 cell line respectively. Results The preliminary scrutiny revealed the presence of polyphenols, flavonoids, terpenoids, steroids, tannins, alkaloids, and glycosides. The total phenolic and flavonoid contents of ethanolic extracts were found higher than those of aqueous extracts. The ethanolic extracts exhibited the superior antioxidant capacity when compared with aqueous extracts. However, the ethanolic extract of MD was shown superlative glucose uptake activity (72.54%) over control (0.037%) and GLUT-4 and PPARγ gene expressions (1.17 and 1.20) in term of folds respectively over cell control (1.00). Conclusion The ethanolic leaf extracts of both plants showed significant in vitro antioxidant and antidiabetic activities compare to aqueous extracts. The Matelea denticulata ethanolic leaf extract exhibited superior activity. This superior activity might be due to their higher phenolic and flavonoid content. However, further approaches are needed to define these activities.

AbstractVaccinum myrtillus L., Ribes nigrum L., Rubus fruticosus L., Fragaria vesca L. leaves are considered an agro-waste of the berry industry. Although numerous studies indicate fruit is a rich source of bioactive compounds, the authors prove leaves can also be a valuable source of compounds used in the pharmaceutical and cosmetic industries. The study attempts to assess and compare the antioxidant and cytotoxic properties of berry leaves extracts. The total phenolic compounds, flavonoids, anthocyanins and procyanidins content were determined. Antioxidant potential was evaluated using the DPPH radical scavenging method. Cytotoxicity studies were conducted to evaluate the effect of the extracts on the metabolism and proliferation of keratinocytes and fibroblasts in vitro. The effect on the migration capacity of these cells was also assessed. The obtained results show that the examined extracts are a source of valuable bioactive agents. All tested extracts show significant ability to remove free radicals in higher concentrations. Cytotoxicity assessments have shown that leaf extracts of the analyzed plants differ in cytotoxicity, both for keratinocytes and fibroblasts. The results of the assessment of cell migration capacity correlate with cytotoxicity tests, because the concentration of extracts showing cytotoxic activity towards the tested cells also inhibited their migration.

In the present research work, in vitro, the antioxidant activity of methanolic extracts of bark of Prunus cornuta and root of Rumex obtusifolius, respectively, was assessed by DPPH free radical scavenging method. Total phenolic content (TPC) of methanolic extracts was also determined by spectrophotometric method using Folin-Ciocalteu reagent and calculated in ?g/mL gallic acid equivalent (GAE). DPPH scavenging activity results show that the root extract of Rumex obtusifolius (IC50 = 17.15) and bark extract of Prunus cornuta (IC50 = 27.32) shows good antioxidant potential. TPC of root extract was found to be 178.56 ?g/g GAE, and bark extract was 137.23 ?g/g GAE. Antioxidant effect (%) determined by using phosphomolybdate assay and superoxide anion radical scavenging percentage further supports the potential antioxidant activity.
