Phytochemical Evaluation and In-vitro Antioxidant Potential of whole plant of Caralluma adscendens

The current line of investigation was focused at perusing the presence of phytochemical constituents, investigation of total phenol and flavonoid content, the antioxidant potential of various extracts of Caralluma adscendens whole plant using various in-vitro assays. The dried plant powder was extracted with various solvents based on polarity (Pet ether, Chloroform, Ethyl acetate, Ethanol and Aqueous) by hot continuous extraction in Soxhlet's apparatus and Extracts were dried. Phytoconstituents present in each extract was examined by performing preliminary phytochemical screening. Total Phenolic Content (TPC), Total Flavonoid Content (TFC) and Antioxidant potential for crude extracts were studied by DPPH, nitric oxide scavenging and FRAP methods. The total phenolic content and flavonoid content of Ethanolic extract of plant was found to be 80.08±0.629mg and 70.88±1.170mg of GAE and Quercetin equivalents respectively. The Ethanolic extract exhibited potent antioxidant activity as determined by 2,2-diphenyl-1-picrylhydrazyl(DPPH), nitric oxide scavenging and ferric reducing antioxidant power assays(FRAP) than the other extracts. The IC50 values for the Ethanolic extract of Caralluma adscendens was found to be 214.765±0.224 µg/ml and 215.928±0.506µg/ml by DPPH and nitric oxide scavenging assays respectively.

The present study aimed to study the anti-oxidant potential of isolated flavonoids from the ethanol extract (70%v/v) of Cassia tora leaves. Three flavonoids were isolated viz Luteolin-7-O-β-glucopyranoside (I), quercetin-3-O-β-d-glucuronide (II) and Formononetin-7-O-β-D-Glucoside (III) from the ethanol extract (70%v/v) of Cassia tora leaves in the previous study. A comprehensive study on the phytochemical contents - total phenolic content and flavonoid content was accessed using Folin-Denis and AlCl3 method respectively. The antioxidant potential of the samples was evaluated using inhibition of hydroxyl radical, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and nitric oxide scavenging methods. The total phenolic content and flavonoid content was found to be 18.60 % w/w and 9.5% w/w respectively. The IC50 values of ethanol extract against hydroxyl, DPPH and nitric oxide radical were found to be 270 µg/mL, 190 µg/mL and 130 µg/mL respectively. The radical scavenging activity of the isolated flavonoids decreased in the following order: quercetin (IC50 values 15, 14, 18 µg/mL) >formononetin (IC50 values 19, 21, 14 µg/mL) > luteolin (IC50 values 20, 23, 18 µg/mL) respectively.

The current investigation is intended to evaluate the content of phytochemical constituents and antioxidant potential of hydroalcoholic extracts of stem and root of Grewia serrulata DC (HAESGS & HAERGS) and leaf and bark of Grewia Nervosa (Lour.) panigrahi (HAELGN & HAEBGN). Initially, all the extracts at different concentrations were estimated for their total phenolic content and total flavonoid content. The study was further extended for their antioxidant potential evaluation using various in vitro methods such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical scavenging assays. The total phenolic content (mg gallic acid equivalent per gram of extract) was high in HAELGN (170.82±0.19) and HAERGS (123.00±0.48) than HAESGS (111.2±0.26) and HAEBGN (119.60±0.23). The total flavonoid content (mg quercetin equivalent per gram) is greater in HAERGS (71.24±0.50) and HAESGS (65.68±0.27) than HAELGN (55.82±0.35) and HAEBGN (62.38±0.45). The IC50 values (µg/ml) of different plant extracts inferred that DPPH radical scavenging activity is greater in HAELGN (42.91±0.88) and HAEBGN (53.87±0.35) than HAESGS (126.73±1.20) and HAERGS (88.87±1.25). However, hydroxyl and superoxide radical scavenging activity is more in HAERGS (135.41±1.19 & 88.00±1.42) and HAELGN (172.28±1.91 & 108.163±1.09) than HAESGS (237.3±1.65 & 110.074±0.87) and HAEBGN (204.7±1.04 & 125.54±1.07). The results of present comprehensive analysis demonstrated that both the plants Grewia serrulata DC and Grewia Nervosa (Lour.) panigrahi possess high phenolic, flavonoid contents and potential antioxidant activity, and could be used as a valid source of natural antioxidants and might be utilized for pharmacological screening of various therapeutic activities. Natural substances extracted from plants have been gaining attraction as protective agents due to their safety and responsible for multiple biological effects on skins. The present study was to determine the total phenolic and flavonoid contents, Sun Protection Factor (SPF), and antioxidant potential of different solvent extracts of Graptophyllum pictum leaves. The dried powdered leaves were extracted by the cold maceration method, using three different solvents, i.e., methanol, ethanol, and water. The extracted leaves was tested for antioxidant activity using of DPPH (1,1-diphenyl-2-picryl-hydrazyl). Total phenolic content was determined using the Follin-Ciocalteu and for total flavonoid content with used the aluminium chloride. The SPF of all extracts were analyzed by ultraviolet (UV) spectrophotometry. The total phenolic content is the highest in methanol extract (377.61±0.31 µg/ml), and total flavonoid content is the highest in ethanol extract (158.06 ± 0.18 µg/ml). For DPPH free radical scavenging activity, ethanolic extract showed the maximum percentage of inhibition, and for SPF, methanol extract had displayed the highest. The study recommends that methanol is a good solvent for the extraction of sun-protective constituents from G. pictum.

In this study there were evaluated total phenolic and flavonoid contents, and ferric reducing antioxidant power (FRAP)of 3 herbs species, that are commonly used in fresh stage (summer savory –Satureja hortensisL., marjoram –Majorana hortensisM. and thyme –Thymus vulgarisL.) in dependence on time of harvest. The total flavonoid content ranged from 2.36 to 4.10 g of catechin equivalents (CE).100g−1of dry weight (dw) of plant material. The highest average total flavonoids content was ascertained in aerial part of summer savory colected in first harvest (4.10 g CE.100g−1dw) and the lowest in aerial part of summer savory collected in third harvest (2.36 g CE.100g−1dw). The highest total flavonoid content was measured by all tested species in plant material harvested in first time of harvest. The highest total phenolic content was estimated in plant material of marjoram harvested in the second time of harvest (6.74 g gallic acid equivalents (GAE).100g−1dw) and the lowest in aerial part of summer savory harvested in the third time of harvest (4.16 g GAE .100g−1dw). Ferric reducing antioxidant power (FRAP) ranged from 1,13 (summer savory, harvest No. 3) to 1.91 g GAE .100g−1dw (thyme, harvest No. 1). The best results of total flavonoid content, total phenolic content and FRAP were obtained by marjoram. Among harvest times there were the highest contents of measured compound mainly by the samples collected in the first time of harvest.

Epidendrum coryophorum belongs to the Orchidaceae family. Traditional uses of some species for this genus include infusions of the leaves used for kidney problems, treat influenza, conjunctivitis, liver pain, relieve kidney symptoms, and hypoglycemic effect. This work's objective was to determine the phytochemical profile of the ethanolic extract of Epidendrum coryophorum leaves and to evaluate the potential anti-inflammatory activity in vitro of the extract employing the erythrocyte membrane stabilization method. The phytochemical screening carried out in this work suggested phenols, coumarins, flavonoids, tannins, steroids, and sterols in the ethanolic extract of Epidendrum coryophorum leaves. Cardiotonic glycosides and carbohydrates were also found. The ethanolic extract's UV-Vis spectrum showed absorption maxima at 268 nm and 332 nm, which could correspond to flavonoids of the flavonoid classes, 3-OH substituted flavonols, or isoflavones. The quantitative determination of total phenols of the ethanolic extract was carried out using the Folin-Ciocalteu method. The total phenolic content expressed as mg Gallic acid equivalent (G.A.E.) per gram of extract was found to be 19,96 mgGAE/g of Epidendrum coryophorum. The ethanolic extract of Epidendrum coryophorum leaves showed hemolysis inhibition values ​​of 18,19% at 1,0 mg/mL, 38,98% at 1,5 mg/mL and 40,94% at 2,5 mg/mL compared with aspirin (positive control) giving values ​​of 65,33% at 1,0 mg/mL, 72,26% at 1,5 mg/mL and 73,75% at 2,5 mg/mL. The values ​​obtained for inhibition of hemolysis with ethanolic extract, compared with the values ​​obtained with a pure anti-inflammatory, are significant and demonstrate anti-inflammatory activity in Epidendrum coryophorum. Introduction: Neuroactive herbal drugs enriched with antioxidants are valuable in treating neurocognitive dysfunction and Vaccinium corymbosum, enriched with antioxidant phytochemicals, is used for treating memory disorders. Hence, the present study evaluated the neuroprotective effects of ethanolic extract of Vaccinium corymbosum (EEVC) on aluminium chloride(AlCl3)-induced Alzheimer’s type of dementia and haloperidol-induced catalepsy-associated behavioural changes. Methods:In vitro antioxidant potential was evaluated using 1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS). The total phenolic content (TPC) was quantified. For in vivo studies, AlCl3 (100 mg/kg) was orally administered for 42 days, whereas the EEVC was administered on the 21st day until the 42nd day in two doses (200 mg/kg and 400 mg/kg). In the haloperidol-induced group, EEVC was treated for 21 days, and haloperidol (1 mg/kg) was administered to induce behavioural changes. Open-field, Y-Maze and traction tests were performed, and the mice brain acetylcholinesterase (AChE) enzyme was determined. Results: IC50 values in DPPH and ABTS assays were 85.5 μg/mL and 80 μg/mL, respectively and the total phenolic content of EEVC was found to be 0.166 mg. In a behavioral study, animals treated with 200 mg/kg and 400 mg/kg of EEVC exhibited a neuroprotective impact on AlCl3-induced neurodegeneration and haloperidol-induced behavioral changes with significant inhibition (P < 0.05 and P < 0.01, respectively) in acetylcholinesterase enzyme. Conclusion: The neuroprotection by EEVC postulated that it is a promising therapeutic agent for treating behavioral and cognitive dysfunctions. Further investigations on pro-inflammatory cytokine and neuroendocrine regulation in transgenic Alzheimer’s disease (AD)models complement the therapeutic value of V. corymbosum.

Olax scandens Roxb. (family Olacaceae) available in throughout tropical India. The current study, aerial parts of different concentrates of Olax scandens was evaluated for its in-vitro antioxidant potential by FRAP assay taking ascorbic acid as the standard and estimation of total phenolic content and flavonoids content. The IC50 value was originated that methanolic concentrates of Olax scandens are more efficient in antioxidant activity by FRAP methods compared EA & PE concentrates. The methanolic concentrates of Olax scandens & ascorbate exhibited antioxidant potential possessing IC50 207µg/ml & 50µg/ml by Ferric reducing ability Power assay. The methanolic and EA concentrates of Olax scandens showed the total phenolic content (14.426 ± 0.032, 4.128 ± 0.025) respectively, and ৒lavonoids content (11.526 ± 0.054, 3.682 ± 0.042) respectively. Invitro antioxidant studies show methanolic concentrates of Olax scandens have better antioxidant activity as well as a higher content of total phenolic and flavonoids content. These results indicate that aerial parts of methanolic concentrates Olax scandens could serve as a natural antioxidant, which may be useful in preventing free radical-induced diseases.

The in vitro antioxidant capacity, total phenolic content and mineral elements of the fruit peel of Myrciaria cauliflora were investigated. The antioxidant capacity was analyzed by the diphenylpicrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP) and β-carotene methods. The assays based on the DPPH (EC50 = 3.18 g sample/g DPPH), ABTS•+ (1017 μmol Trolox/g sample), FRAP (1676 µM Fe2SO4/g sample) and β-carotene/linoleic acid (70% of oxidation inhibition) methods indicated a high antioxidant capacity of the fruit peel extract of the plant. The Folin-Denis method was more efficient in determining the total phenolic compound contents in the different solvents than the Folin-Ciocalteu one. Extractions made with 4:1 methanol-water, 4:1 ethanol-water, 3:2 ethanol-water and 3:2 acetone-water solutions using the Folin-Denis method exhibited high contents of phenolic compounds (18.95, 14.06, 12.93 and 11.99 mg GAE/g, respectively). Potassium was the major element found in the fruit peel, followed by phosphorus, calcium, magnesium and iron, in that order. As a result, the fruit peel of M. cauliflora can be considered as an important source of natural antioxidants and essential elements of easy access for the population and for application in the food industry.

Clitoria ternatea, commonly known as Butterfly pea flower or Bunga telang, is a tropical flower and notable as one of the most vital sources of polyphenols with high antioxidant capacity. C. ternatea abundantly produces anthocyanin compounds benefiting natural blue colourants. To date, a blue colourant is the most difficult to obtain since the anthocyanins responsible for this colour is the least stable and easily degraded after extraction. Their stability commonly relies on several operational parameters, such as shifted pH value, high temperature, and light exposure. Apart from colour attributes, anthocyanins have also been identified as a source of functional molecules due to antioxidant activity and beneficial health effects such as anti-cancer, anti-obesity, antimicrobial and more. This study aimed to characterize the chemical constituents of the extractable polyphenols from ethanolic extract of C. ternatea quantitatively. The numerical data were evaluated using the Minitab version 18 statistical method. The chemical profiling, including total phenolic content (TPC), total flavonoid content (TFC), total monomeric anthocyanins (TMA) were determined using standard methods. The antioxidant activity was evaluated using DPPH and ABTS methods. The results showed that the ethanolic extract of C. ternatea had 35.7 mg QE/g of TFC, 102.4 mg GAE/g280nm and 28.8 mg GAE/g750nm of TPC and 2.7 CE/g and 2.9 ME/g of TMA. The total phenolic content of C. ternatea showed a good correlation with the antioxidant activity by the DPPH method when analysed using Pearson correlation and showed an increasing trend of antioxidant power with regards to TPC in both assays. In a nutshell, this study contributes to the knowledge of ethanolic extract of C. ternatea. Future research could consider exploring other extraction methods that could enhance the stability of polyphenols particularly the anthocyanins from degradation such as using surfactants.
