Research background: The aim of this work was to evaluate utilization of sweet potato peel as a source of bioactive compounds. The effect of solvents (acetone and acetone/ethanol mixture) on extraction efficiency of total carotenods and phenolics from sweet potato flesh tuber and peel, and antioxidant activity were investigated. SPP extract standed out in terms of antioxidant activity and was choosen for encapsulation by spray and freeze drying techniques. Experimental approach: Encapsulation is an effective method to improve phytochemical stability by entrapping the core material with a coating agent. In this study, spray and freeze-drying techniques were applied for improving the stability of bioactive compounds (carotenoids and phenolics) using whey protein as a coating material. The main advantages of applied techniques over the other encapsulation techniques are simplicity, continuity, effectiveness, availability, and applicability. Results and conclusions: Physicochemical characteristics revealed that spray drying resulted in the formation of lower size particles, better flowing properties, and encapsulation efficiency of carotenoids. The retention of encapsulated and non-encapsulated bioactive compounds was monitoring during storage at daylight and dark conditions. Storage conditions affected the carotenoid retention, whereas daylight exhibited the higher degradation rate for all samples. Phenolic compounds exhibited higher retention for all investigated samples. Degradation kinetic parameters suggest the longer shelf life of spray dried encapsulates and potent method for bioactives stabilization. Novelty and scientific contribution: This study demonstrates that the spray drying technique and utilization of sweet potato peel have big potential in functional additives development, with improved nutritional, color and bioactive properties.

Chayote (Sechium edule (Jacq.) Swartz) and kohlrabi (Brassica oleracea var gongylodes L.) are medicinal plants widely distributed in Thailand. Several traditional medicines usually contain these extracts due to their pharmaceutical activities. However, appropriate technologies that are used for protection, stabilization, and slow release of plant extracts are a lot desired in terms of food application. In this study, chayote and kohlrabi extracts were encapsulated by several kinds of wall materials (maltodextrin, and the combination of maltodextrin and gum arabic or alginate) and drying methods (freeze-drying and tray-drying techniques). Thus, the objective of this research was to determine morphological and physicochemical properties, wall materials releasing, and antioxidant activity of encapsulated chayote and kohlrabi extracts powder. The morphology of all encapsulated chayote and kohlrabi extracts powder showed irregular spherical shape, monodispersity, and smooth surface. The encapsulated chayote and kohlrabi extracts powder with tray-drying technique tend to have more darkness and redness in color than the freeze-drying technique. Wall material releasing was expressed in glucose liberation of encapsulated extracts powder after amylolytic enzyme digestion. Encapsulation using maltodextrin as wall material provided higher wall material releasing than the other samples. After digestion analysis, the digested residues were examined for antioxidant activity. The results showed that the combination of maltodextrin and alginate for both freeze-drying and tray-drying techniques provided higher antioxidant activity after 60 and 120 min of digestion. Thus, the combination of maltodextrin and alginate, and drying with the freeze-drying technique was the best treatment in this experiment. This data can lead to a better understanding of wall materials types and releasing characteristics, which are used to control bioactive compounds liberation in the gastrointestinal tract.

Background: Tetrahydroindazole, a member of the fused-pyrazole system, is a least studied class of heterocyclic compounds owing to its scarcity in nature. However, a large number of synthetically prepared tetrahydroindazoles are known to show a variety of biological activities such as interleukin- 2 inducible T-Cell kinase inhibitors, AMPA receptor positive allosteric modulators, antitumor, antituberculosis, anti-inflammatory and antimicrobial activities. Vilsmeier-Haack reaction is one of the most important chemical reactions used for formylation of electron rich arenes. Even though Vilsmeier- Haack reaction was studied on a wide variety of hydrazones derived from active methylene compounds, literature lacks the examples of the use of 4-substituted cyclohexanones as a substrate for the synthesis of 4,5,6,7-tetrahydroindazoles. The study of the reaction of Vilsmeier-Haack reagent with hydrazones derived from cyclic keto compounds having active methylene has been considered the interested topic of investigation. In the present study, ethyl cyclohexanone-4-carboxylate was treated with one equivalent of various hydrazines for two hours and the resulted hydrazones were further treated with an OPC-VH reagent (Vilsmeier-Haack reagent isolated from phthaloyl dichloride and N,Ndimethylformamide) afforded 4,5,6,7-tetrahydroindazoles in excellent yields. The synthesized compounds 4a-f and 5a-f were screened for their antioxidant activities using the DPPH radical scavenging assay. The target compounds were synthesized regioselectively using 4+1 approach in excellent yields. A number of experiments using both conventional heating as well as microwave irradiation methods were tried and on comparison, microwave irradiation method was found excellent in terms of easy work up, high chemical yields, shortened reaction times, clean and, no by-products formation. Some of the synthesized compounds showed significant antioxidant activity. The microwave assisted synthesis of 4,5,6,7-tetrahydroindazoles from ethyl cyclohexanone-4-carboxylate has been reported under mild conditions in excellent yield. Easy work up, high chemical yield, shortened reaction times, clean and no by-products formation are the major advantages of this protocol. These advantages may make this method useful for chemists who are interested in developing novel 4,5,6,7-tetrahydroindazole based drugs.

Sulfated polysaccharide fucoidan isolated from brown algae shows a wide range of biological activities that are significantly dependent on its chemical composition, which is closely related to the applied technique and extraction parameters. Therefore, the objective of this study was to evaluate the influence of microwave assisted extraction (MAE) and pressurized liquid extraction (PLE) parameters (solvent, temperature, time, and number of cycles) on the Fucus virsoides and Cystoseira barbata polysaccharide yield (%PS) and chemical composition (total sugar, fucose, and sulfate group). The optimal MAE parameters that resulted in the highest polysaccharide extraction from F. virsoides and C. barbata were 0.1 M H2SO4 for 10 min at 80 °C, while the optimal PLE parameters were 0.1 M H2SO4, for two cycles of 15 min at 140 °C. Furthermore, the %PS, chemical structure, molecular properties, and antioxidant activity of the F. virsoides and C. barbata polysaccharide extracts obtained with MAE, PLE, and conventional extraction (CE) performed under previously determinate optimal conditions were compared. PLE resulted in a significantly higher %PS from F. virsoides, while for C. barbata, a similar yield was achieved with CE and PLE, as well as CE and MAE, for both algae. Furthermore, the polysaccharides obtained using PLE had the highest polydispersity index, fucose, and sulfate group content, and the lowest uronic acid content; however their antioxidant activity was lower.

AbstractThe grape pomace is a by-product from the industrial processing of grape juice, which can be used as a source of bioactive compounds. The aim of this study was to separate the phenolic compounds from grape pomace using an acidic aqueous solution with 2 % citric acid as a solvent, using both ultrasound-assisted extraction, with powers of 250, 350 and 450 W and times of 5, 10 and 15 min, and microwave-assisted extraction using powers of 600, 800 and 1,000 W and times of 5, 7 and 10 min. The results showed that for both methods of extraction, the contents of total phenolic compounds and antioxidant activity by ABTS and DPPH increased with time, and microwave at 1,000 W for 10 min corresponded to the best extraction condition. However, the contents of phenolic compounds and antioxidant activity were lower than exhaustive extraction using acidified methanol solution.
