You Are Only as Good as You Are Behind Closed Doors

Virtues are standardly characterized as stable dispositions. A stable disposition implies that the virtuous actor must be disposed to act well in any domain required of them. For example, a politician is not virtuous if s/he is friendly in debate with an opponent, but hostile at home with a partner or children. Some recent virtue theoretic accounts focus on specific domains in which virtues can be exercised. I call these domain-variant accounts of virtue. This paper examines two such accounts: Randall Curren and Charles Dorn’s (2018) discussion of virtue in the civic sphere, and Michael Brady’s (2018) account of virtues of vulnerability. I argue that being consistent with the standard characterization of virtue requires generalizing beyond a domain. I suggest four actions the authors could take to preserve their accounts while remaining consistent with the standard characterization. I also discuss how virtue education could be enhanced by domain-variant accounts.

      This study deals with the colonial perspectives in Dave Eggers’s A Hologram for The King (2012), according to the postcolonial approach. Although colonialism era is over by now, colonial perspectives remain strong in some literary works. Since its advent in the second half of the twentieth century, postcolonial theory confronts colonial attitudes and experiences as colonialism has been justified in many works of Western writers and scholars who have distorted the real image of non-Europeans and non-Westerners via different means and techniques in masquerade of orientalism. Postcolonial discourse opposes the misrepresentation of non-Europeans and argues that such falsification is driven by political, social, religious and economic motives. In the current study, the researcher aims at explaining the notions of colonialism, otherization and other falsified images of non-Westerners in A Hologram for the King. This paper mainly questions Eggers’s portrayal of the protagonist, Alan Clay, who after bankruptcy and failure at home, flies to Saudi Arabia and capitalizes on the physical and moral assets of the Orientals in this country to convert his story of failure to a success. The characterization of the oriental world and its setting show Eggers’s being biased against the Eastern world and ironically mirror clear hints of colonialism and eurocentrism.

This chapter explores the question of the nature of the alienation of the bourgeoisie under capitalism. In particular, it considers the distinction made by Karl Marx in The Holy Family between the alienation endured by the worker and the alienation endured by the capitalist in bourgeois society. According to Marx: “The possessing classes and the class of the proletariat present pictures of the same human self-estrangement. But the former class feels at home in and confirmed by this self-estrangement.” The chapter analyzes the meaning of this passage by focusing on a characterization of the human essence in The German Ideology and on the doctrine of alienation articulated in the Paris Manuscripts. It also discusses the worker's alienation in his relation to the machine, and the capitalist's alienation in his relation to money, as well as the latter's relation to his capital. Finally, it restates the contrast between bourgeois and proletarian.

Abstract To begin unraveling the functional significance of calcium channel diversity, we identified mutations in Dmca1D, a Drosophila calcium channel α1 subunit cDNA that we recently cloned. These mutations constitute the l(2)35Fa lethal locus, which we rename Dmca1D. A severe allele, Dmca1DX10, truncates the channel after the IV-S4 transmembrane domain. These mutants die as late embryos because they lack vigorous hatching movements. In the weaker allele, Dmca1DAR66, a cysteine in transmembrane domain I-S1 is changed to tyrosine. Dmca1DAR66 embryos hatch but pharate adults have difficulty eclosing. Those that do eclose have difficulty in fluid-filling of the wings. These studies show that this member of the calcium channel α1 subunit gene family plays a nonredundant, vital role in larvae and adults.

Conjugative pili are extracellular filaments elaborated by Gram-negative bacteria expressing certain type IV secretion systems. They are required at the earliest stages of conjugal DNA transfer to establish specific and secure cell–cell contacts. Conjugative pili also serve as adsorption organelles for both RNA and DNA bacteriophages. Beyond these facts, the structure, formation and function of these filaments are poorly understood. This paper describes a rapid, quantitative assay for F-pili encoded by the F plasmid type IV secretion system. The assay is based on the specific lateral adsorption of icosahedral RNA bacteriophage R17 by F-pili. Bacteriophage particles conjugated with a fluorescent dye, Alexa 488, and bound to F-pili defined filaments visible by immunofluorescence microscopy. F-pili attached to F+ cells and free F-pili were both visible by this method. For quantification, cell-bound bacteriophage were separated from free bacteriophage particles by sedimentation and released by suspending cell pellets in 0·1 % SDS. Fluorescence in cell-free supernatant fractions was measured by fluorometry. The authors present a characterization of this assay and its application to F-pilus formation by cells carrying mutations in the gene for the F-pilus subunit F-pilin. Each mutation introduced a cysteine, which F-pilin normally lacks, at a different position in its primary structure. Cysteine residues in the N-terminal domain I abolished filament formation as measured by fluorescent R17 binding. This was confirmed by measurements of DNA donor activity and filamentous DNA bacteriophage infection. With one exception (G53C), cysteines elsewhere in the F-pilin primary structure did not abolish filament formation, although some mutations differentially affected F-pilus functions.

This chapter explores the twinned emergence in the British novel of a critique of plantation slavery and commercial imperialism with a proto-feminist questioning of the ‘commerce of the sexes’. The discourses of racial and sexual oppression resonate with one another, helping to establish connections between inequalities at home and the sufferings of distant others. It has been argued that novelistic representations of violence and suffering are central to an ‘imagined empathy’ which in turn assisted the development in the eighteenth century of humanitarian sentiment. While it might be charged that the mid-eighteenth-century novel failed to grant full humanity to the enslaved and that it was somewhat instrumentalist in its handling of slavery reform, it can be demonstrated that the versatility of the figure of slavery enabled fuller characterization of the colonized and enslaved, as well as the more explicit imagining of colonial violence.

Cryptosporidium parvum is a protozoan parasite that can cause moderate-to-severe diarrhea. Insulinase-like proteases (INS) are one of the largest protein families within the small proteome of the pathogen. However, their roles in C. parvum biology remain un-elucidated. In this study, a member of the protein family, INS-15 of C. parvum encoded by cgd3_4260, was cloned, expressed and characterized to understand its function. INS-15 and its domain I were expressed in Escherichia coli and polyclonal antibodies against the domain I and one specific polypeptide were prepared in rabbits. The role of INS-15 protein in the C. parvum invasion was preliminarily studied. Recombinant INS-15 protein and its domain I were successfully expressed in E. coli, together with various degraded products. The cgd3_4260 gene had a peak expression at 2 h of in vitro C. parvum culture, while the INS-15 protein was expressed in the mid-anterior region of sporozoites and the area of merozoites opposite to the nucleus. Anti-INS-15 domain I antibodies reduced the invasion of C. parvum sporozoites by over 40%. The anterior location of INS-15 in invasion stages and partial reduction of in vitro growth indicate that INS-15 plays some roles in the invasion or early development of C. parvum.
