We repeatedly sampled eight sites on the Crooked River in British Columbia’s Arctic watershed for adult and nymph mayflies (Ephemeroptera) over the course of two years. Using taxonomic keys and DNA-barcoding we report eight new species records for the province. These are five Baetidae (Acerpenna pygmaea, Baetis phoebus, Baetis vernus, Iswaeon anoka, and Procloeon pennulatum), one Heptageniidae (Leucrocuta hebe), one Leptohyphidae (Tricorythodes mosegus), and one Siphlonuridae (Siphlonurus alternatus). Three of these – Acerpenna, Iswaeon, and Leucrocuta – are also new genus records for the province. In total we detected 40 species in eight families as indicated by clustering into BINs (Barcode Index Numbers), by morphological keys, and by matches in the Barcode of Life Database. One of those species, Ameletus vernalis, is of conservation concern. Our analysis indicated that a number of other specimens may represent new species or genus records for BC. In addition this unique and anthropogenically impacted river may contain cryptic species of Baetis tricaudatus (Baetidae), Leptophlebia nebulosa (Leptophlebiidae), and Paraleptophlebia debilis (Leptophlebiidae).

DNA barcoding has been recently promoted as a method for both assigning specimens to known species and for discovering new and cryptic species. Here we test both the potential and the limitations of DNA barcodes by analysing a group of well-studied organisms—the primates. Our results show that DNA barcodes provide enough information to efficiently identify and delineate primate species, but that they cannot reliably uncover many of the deeper phylogenetic relationships. Our conclusion is that these short DNA sequences do not contain enough information to build reliable molecular phylogenies or define new species, but that they can provide efficient sequence tags for assigning unknown specimens to known species. As such, DNA barcoding provides enormous potential for use in global biodiversity studies.Key words: DNA barcoding, species identification, primate, biodiversity.

Betapista dekkerae, new genus and new species, is described. The generic diagnoses for Laphania and Scionides are improved after inspection of the holotypes of the type species. Eupolymnia heterobranchia (syn., E. crescentis) is redescribed. Additions to the descriptions, based on study of type material, are made for Laphania boecki (new record, Northeast Pacific), Neoamphitrite robusta (syn., Scionides dux), Neoleprea californica and N. spiralis, Pista brevibrunchiata (new record, British Columbia [B.C.], Washington [WA], Japan) and P. fratrella, and Polycirrus californicus (new record, B.C., WA, syn., P. perplexus). Other additions to the descriptions are provided for Amaeana occidentalis (new record, B.C.) and Pista cristata. Two further Pista species (one from the Skagerrak) and five Polycirrus species are charcterized but not named. Other new records are Lanassa venusta venusta (B.C.), Lysilla loveni, and Neoleprea japonica (the two latter for Northeast Pacific). Lysilla pacifica, Pista fasciata, and P. fratrella are shown not to be members of the fauna of British Columbia and Washington. Presumably, neither is Polycirrus caliendrum. Abstract Background The northeastern part of the Qinghai-Tibet Plateau (QTP) presents a high number of plateau loach species. As one of the three major groups of fishes distributed on the QTP, plateau loach has high ecological value. However, the taxonomy and systematics of these fish are still controversial, and a large number of new species have been reported. The reason for this phenomenon is that the degree of morphological variation is low, the phylogenetic information provided by morphological and anatomical features used for species identification is relatively poor, and many cryptic species are observed. Based on the high-density sampling points from the biodiversity hotspots surveyed, this study aims to evaluate the biodiversity of plateau loach in the northeastern part of the QTP and reveal the hidden diversity by comparing morphological species with molecular operational taxonomic units (MOTUs). Results After careful identification and comparison of the morphology and DNA barcoding of 1630 specimens, 22 species were identified, with 20 considered valid local species and two identified as new species that had not been previously described. Based on the combination of morphological and molecular methods, a total of 24 native species were found, two of which were cryptic species: Triplophysa robusta sp1 and Triplophysa minxianensis sp1. Fourteen of the 24 species form clusters of barcodes that allow them to be reliably identified. The remaining cases involved 10 closely related species, including rapidly differentiated species and species that seemed to have experienced incomplete lineage sorting or showed introgressions. Conclusions The results highlight the need to combine traditional taxonomies with molecular methods to correctly identify species, especially closely related species, such as the plateau loach. This study provides a basis for protecting the biodiversity of plateau loach.

Two new species, A. coniferana from British Columbia and Alberta and A. morrisana from New Brunswick, are distinguished from A. borealis by characters of maculation and especially of male and female genitalia.

Anthropogenic pressures on aquatic systems have placed a renewed focus on biodiversity of aquatic macroinvertebrates. By combining classical taxonomy and DNA barcoding we identified 39 species of caddisflies from the Crooked River, a unique and sensitive system in the southernmost arctic watershed in British Columbia. Our records include three species never before recorded in British Columbia: Lepidostoma togatum (Lepidostomatidae), Ceraclea annulicornis (Leptoceridae), and Cheumatopsyche harwoodi (Hydropsychidae). Three other specimens may represent new occurrence records and a number of other records seem to be substantial observed geographic range expansions within British Columbia.

The family Eosentomidae from Japan is revised. A new genus, Osientomon, is erected and a new species is ascribed to this genus. This new genus is characterized by the short, setiform sensillum g, absence of sensilla e and b'1 on foretarsus, and S-shaped processus sternalis and duck’s head type caput processus on the female squama genitalis. Two new species are added to the genus Pseudanisentomon and 19 new species and 1 new record of the genus Eosentomon are described. As a result, 43 species belonging to 4 genera are now recognized from Japan. Taxonomic keys are provided for the identification of the genera and species from Japan.

AbstractWe describe three new genera and four new species (three named) of siricomorph sawflies (Hymenoptera: Symphyta) from the Ypresian (early Eocene) Okanagan Highlands: Pamphiliidae,Ulteramus republicensisnew genus, new speciesfrom Republic, Washington, United States of America; Siricidae,Ypresiosirex orthosemosnew genus, new speciesfrom McAbee, British Columbia, Canada; and Cephidae,Cuspilongus cachecreekensisnew genus, new speciesfrom McAbee and another cephid treated as Cephinae species A from Horsefly River, British Columbia, Canada. These are the only currently established occurrences of any siricomorph family in the Ypresian. We treat the undescribed new siricoid from the Cretaceous Crato Formation of Brazil as belonging to the Pseudosiricidae, not Siricidae, and agree with various authors that the YpresianMegapterites mirabilisCockerell is an ant (Hymenoptera: Formicidae). The Miocene speciesCephites oeningensisHeer andC. fragilisHeer, assigned to the Cephidae over a century and a half ago, are also ants. Many of the host plants that siricomporphs feed upon today first appeared in the Eocene, a number of these in the Okanagan Highlands in particular. The Okanagan Highlands sites where these wasps were found also had upper microthermal mean annual temperatures as are overwhelmingly preferred by most modern siricomorphs, but were uncommon in the globally warm Ypresian, only found then in higher elevations and highest latitudes.
