Background: Today, nanoparticles hold great promise in biomedical researches and applications including bacteria detection. The rapid and sensitive outcomes of bacteria detection strategies using nanoparticle conjugates become determinative, especially in bacterial outbreaks. In the current research, we focused on detecting V. cholera bacteria and its toxin using a thiocyanate/Au nanoparticle. Thiocyanate adsorbed strongly on the surface of gold nanoparticles and changed the surface by enhancing surface plasmon resonance of gold nanoparticles. Objective: This method is tried to introduce a simple and fast procedure to assay vibrio cholera. So, it is observed by the naked eyes as well. Methods: We used two antibodies (Ab) for V. cholera detection: a) a primary antibody conjugated to magnetic nanoparticles (MNPs) for trapping V. cholera bacterial cells, and b) a secondary Abconjugated thiocyanate-GNPs as a colorimetric detector. Then, an immuno-magnetic separation system connected to a colorimetric assay was designed based on the GNPs. The results were measured by ultraviolet-visible (UV-Vis) spectroscopy. Results: The results showed that gold nanoparticles are an appropriate optical assay for detecting biological samples in a minimum concentration and also it can be easily seen by the naked eyes. The linear range of this biosensor is 3.2×104 to 28×104 cells per ml. Conclusion: In this research, a colorimetric immune assay based on gold nanoparticles was designed to improve the sensitivity of V. cholera detection. Also, this method can be used for the detection of other biological agents.

Background—Endoscopic retrograde cholangiopancreatography sphincterotomy is increasingly performed in younger patients undergoing laparoscopic cholecystectomy. However, the safety of endo- scopic sphincterotomy in this age group, relative to that in older patients, is unknown.Aim—To determine whether the development of short term complications following endoscopic sphincterotomy is age related.Patients and methods—A prospective multicentre audit of 958 patients (mean age 73, range 14–97, years) undergoing a total of 1000 endoscopic sphincterotomies.Results—Two deaths occurred, both from postsphincterotomy acute pancreatitis. Postprocedural complications developed in 24 patients: pancreatitis in 10, ascending cholangitis in seven, bleeding in four, and retroperitoneal perforation in three. There were six complications (five cases of pancreatitis and one bleed; 2.2%) and no deaths in the 281 (29.3%) patients aged under 65 years. In comparison, 18 (2.6%) of the 677 patients aged over 65 years developed a complication (cholangitis in seven, pancreatitis in five, bleeding in three, and perforation in three). Patients under 35, 45, 55, and 65 years were not at significantly increased risk of complication than those over these ages (relative risk for those under compared with those over 65 years 0.83, 95% confidence intervals 0.41–1.67, p=0.74).Conclusion—Short term complications following endoscopic sphincterotomy are not related to age. Younger patients undergoing laparoscopic cholecystectomy need not be denied endoscopic sphincterotomy for fear that the risks are greater than if they undergo surgical exploration of the common bile duct.

On July 20, 2010, three cases of cholera were reported from a district hospital in Ca Mau province, Vietnam. We investigated the likely source and mode of transmission of the outbreak. All hospitals in the province were requested to notify cases of acute watery diarrhoea. Epidemiological, clinical, and laboratory data were collected. Between July 12 and 22, seven cases with positive culture for Vibrio cholera were identified. Six cases were epidemiologically linked to the index case. Basic infection control practices were not in place at the hospital. Clinicians and public health staff should consider the possibility of nosocomial cholera transmission even in non-endemic areas.

<p><strong>Objective: </strong>To evaluate the chemical composition, antibacterial and antioxidant properties of stem bark essential oil of <em>Litsea glutinosa </em>C. B. Rob.</p><p><strong>Methods: </strong>The essential oil isolated from stem bark of <em>L. glutinosa </em>and their chemical composition was analyzed by gas chromatography coupled with mass spectrometry detector. The <em>in vitro </em>antibacterial activity of the stem bark essential oil was investigated against eight human pathogenic bacterial clinical isolates using agar disc diffusion method and MIC value was determined by modified resazurin microtitre-plate assay. The antioxidant activity of essential oil was measured by 2, 2-diphenyl-1-picrylhydrazyl radical (DPPH), 2, 2-azinobis-3-ethylbenzothiazoline-6-sulphonate radical cation (ABTS) and β-carotene bleaching assay.</p><p><strong>Results: </strong>GC-MS analysis of stem bark essential oil resulted in the identification of 37 compounds, off which 9,12-octadecadienoic acid (62.57%), hexadecanoic acid (12.68%), stigmast-5-en-3-ol (6.87%) and vitamin E (2.51%) were the main constituents representing 84.63% of the oil. The determination of <em>in vitro</em> antibacterial activity of stem bark essential oil resulted in significant inhibition zone (15.00±0.57 mm) and MIC value (0.15±0.15×10<sup>-2</sup> mg/ml) against the pathogenic bacteria <em>Vibrio cholera</em> followed by <em>Pseudomonas aeruginosa</em> and <em>Salmonella typhi. </em>The results of DPPH radical scavenging (IC<sub>50</sub>:4.540±0.06 µg/ml), ABTS (IC<sub>50</sub>:256.02±0.06 µg/ml) and β-carotene bleaching assay (%I: 78.51±0.42 <strong>%</strong>) showed significant <em>in vitro</em> antioxidant property.</p><p><strong>Conclusion: </strong><em>L. glutinosa</em> stem bark essential oil showed potential antibacterial activity against the <em>Vibrio cholera</em>. The results of this investigation supported the ethnomedical claim of essential oil as a demulcent, antidiarrheal and antioxidant drug.</p>
