Prevalence of Cryptosporidiosis Detected by Enzyme Immunoassay Coproantigen among People Living with HIV/AIDS Attending Selected Hospitals in Maiduguri, Nigeria

Background: Cryptosporidium species are intestinal parasites that infect both humans and animals; it causes cryptosporidiosis which usually resulted to diarrhea especially among those with impaired immunity. It was observed that enzyme linked immunosorbent assay (ELISA) be ascertain was never been used as one of the techniques in the detection of Cryptosporidium parvum antigen in Maiduguri, Borno state. Materials and Methods: Four hundred stool and blood samples were collected in four selected hospitals in the study area. Stool samples were analyzed by Enzyme link immunosorbent assay to detect fecal Cryptosporidium antigen, while the blood samples were analyzed with Partec sysmex ® flow cytometric machine for CD4 T-lymphocyte counts. Results: Demographically, female are 275 while male 125 in number. The result of the stool samples have shown that, of the four hundred samples, seventy nine (79) patients  were positive for Cryptosporidium  species with an infection rate of 19.8%. According to gender, females have the highest infection rate of 14.25% while males 5.5%.based on age, patients between the ages of 20-39 have the highest prevalence of 9.5% followed by 40-59 with 7.5% and lastly 0-19 and >60 old are1.25 and 1.5% respectively. Traders (businessmen and women) demonstrated the highest infection rate of 8.0% followed by housewives 6.25%, civil servants 4.25% and lastly student and farmer with 1.0 and 0.25% respectively. In terms of CD4-T-lymphocyte count, those with CD4 <200cells/µl has an occurrence rate of 8.25% followed by 201-499 with 7.0% and >500 with 4.5%. on the bases of clinical details, the results have shown that patients with diarrhea has a prevalence of 13.0% while those without diarrhea has 6.75%, however, according to the hospitals the patients attended, the result has that, USUMH has 7.0%.Followed by UMTH 6.5%, SSHM 3.75%, and MSMH 2.5% respectively. Conclusion: It was concluded that those with cd4 count bellow 200 are at risk of contacting the parasites and hence is one of the causes of diarrhea among HIV patients. It is recommended that proper hygiene practice should be encouraged.

A survey of helminth parasites was carried out on 198 dogs living in almost complete liberty in villages in the northeast of Gabon. Faeces and blood samples were collected and analysed. Dirofilariaimmitis antigen was detected in 13.6 % of dogs using the SNAP 3Dx® test, a commercially available enzyme-linked immunosorbent assay (ELISA). Faecal examination revealed that 91.4 % of dogs were infected by intestinal helminths. Ascarids were found in 58.5 % of the samples. Trichuris vulpis was observed in 49.5 % of cases, and Uncinaria spp. and Ancylostoma spp. in 34.8 %, Spirocerca lupi in 25.3 % and Capillaria spp. in 10.6 %. Cestode embryophores were found in 8.6 % of the samples.

SUMMARYPassive immunity against measles decreases during the first months of life. The objective of this study was to determine titres of measles antibodies in children aged 9–14 months and their mothers before vaccination, and the children's response to vaccination. Blood samples were collected by capillary puncture before and 28 days after vaccination. Samples were obtained between February and June 2007 during an ongoing measles outbreak. Titres of specific measles IgG antibodies were determined by enzyme-linked immunosorbent assay. Seroconversion was defined as the presence of antibodies after vaccination in subjects without antibodies before vaccination. Maternal antibodies were present in 37·7% of all 69 children included and in 45·1% of children aged 9 months. Of the 51 children in whom a second sample was obtained, 31 (60·8%) were seronegative before vaccination and 61·3% seroconverted. Interference of maternal antibodies was 30%. Advancing the first dose of measles vaccination from 15 to 12 months is a correct strategy, given the increase in the time of susceptibility of infants to measles.

Prolactin has been reported to be a remarkable index of stress response, both acute and chronic, in several species. The use of biological matrixes other than blood is receiving increasing interest in the study of hormones, due to the lower invasiveness in collection. This research aimed to investigate the possibility of using a commercial ELISA (enzyme-linked immunosorbent assay) kit for measuring canine prolactin in blood for the quantification of canine prolactin in saliva. Study 1 consisted of a validation protocol, using saliva samples collected from lactating and non-lactating dogs. Study 2 was conducted to investigate a possible correlation between prolactin concentration in saliva and plasma in sheltered dogs by using the same kit. Prolactin values were reliably read only when they came from blood samples, not from saliva, but tended to be low in most of the cases. Study 1 showed that saliva had a matrix effect. In study 2, saliva prolactin levels were low and in 42.9% of cases, not readable. No correlation between prolactin values in plasma and saliva was found (ρ=0.482; p=0.274). These findings suggested that the determination of prolactin in dog saliva through an ELISA kit created for measuring prolactin in dog blood was unreliable.

Out of 428 faecal 178 (41.59%) animals were found positive for Cryptosporidium parvum oocysts in the bovine faeces in modified Ziehl-Neelsen (mZN) staining and/ or commercial plate and dipstick- enzyme linked immunosorbent assay (ELISA) kits (Cypress diagnostics, Langdorp, Belgium) with statistically non-significant difference in the occurrence rate in cattle and buffalo calves. Seasonal prevalence difference was statistically significant in cattle calves while non-significant in buffalo calves, respectively. The highest overall prevalence was recorded during rainy season (45.15%) and almost same per cent during winter and summer. Prevalence of 67.26, 37.11, 30 and 17.65% was recorded in calves aged below 1 month, 1-3 months, 4-8 months and 9-12 months, respectively (p>0.05). There was no significant difference in the age group prevalence between cattle and buffalo calves. Both sexes of bovines are equally susceptible to the cryptosporidiosis (p>0.05). The prevalence of cryptosporidiosis was 59.54 and 29.41% for diarrhoeic and non-diarrhoeic samples, respectively (p>0.05). Both diarrhoeic and non-diarrhoeic groups of calves aged between 1-30 days were equally susceptible to the infection of Cryptosporidium spp. Almost same per cent prevalence of the infection was observed in dairy calves reared in organized (40.76%) and unorganized farms (42.21%) and the difference was non-significant in both cattle and buffalo calves. Highest prevalence of cryptosporidiosis was found in HF cross calves. The tests failed to detect the oocysts in infected soil samples. There was highly significant difference in the prevalence of cryptosporidiosis estimated by plate/ dipstick-ELISA and mZN staining with highest sensitivity and specificity in plate-ELISA.

AbstractThe present study was conducted to determine the prevalence of canine dirofilariosis in Hatay province, south of Turkey. A total of 269 blood samples were collected from owned dogs in this multi-centre survey between March and July 2006. Blood samples were examined by modified Knott and enzyme-linked immunosorbent assay (ELISA) techniques to detect circulating microfilariae and antigens of Dirofilaria immitis, respectively. Seventy out of 269 dogs (26.0%) were positive for D. immitis. However, 61.4% of positive dogs had occult infection. The prevalence of canine dirofilariosis was heterogeneous in Hatay province, with higher values in shoreline (33.0%) and riverside (30.9%) areas followed by the lowland (25.5%) or mountainous (15.2%) areas. No statistically significant differences were observed in relation to sex (females, 33.3%; males, 24.4%, P>0.05). When evaluating the prevalence of D. immitis by age, the highest prevalence was observed in dogs older than 4 years of age. Large breeds (29.6%) and the dogs living outdoors (30.2%) showed a higher prevalence in comparison to small breeds (21.4%) and the dogs living indoors (10.5%), respectively. In conclusion, according to the results of this study canine dirofilariosis had a high prevalence in Hatay province. Therefore, prophylaxis against heartworm is advisable to decrease the incidence of canine dirofilariosis.

Objectives: the main aim of this study determine the occurrence of the seroepidemiological toxoplasmosis in camels in waist province of Iraq from November 2016 to April 2017. Materials and Methods: blood samples collected of animals randomly were included six different groups of animals were diagnosed by A Latex agglutination test (LAT) and indirect enzyme linked immunosorbent assay (ELISA) kits.
