In this study, highly active Fe3O4@PDA@Au@GO surface-enhanced Raman spectroscopy (SERS) active substrate was synthesized for application in the enrichment and detection of trace polycyclic aromatic hydrocarbons (PAHs) in the environment. The morphology and structure were characterized by transmission electron microscopy (TEM), energy-dispersive spectroscopy (EDS), X-ray diffraction (XRD) and UV–visible absorption spectrum (UV–vis spectra). The effect of each component of Fe3O4@PDA@Au@GO nanocomposites on SERS was explored, and it was found that gold nanoparticles (Au NPs) are crucial to enhance the Raman signal based on the electromagnetic enhancement mechanism, and apart from enriching the PAHs through π–π interaction, graphene oxide (GO) also generates strong chemical enhancement of Raman signals, and polydopamine (PDA) can prevent Au from shedding and agglomeration. The existence of Fe3O4 aided the quick separation of substrate from the solutions, which greatly simplified the detection procedure and facilitated the reuse of the substrate. The SERS active substrate was used to detect phenanthrene in aqueous solution with a detection limit of 10−7 g/L (5.6 × 10−10 mol/L), which is much lower than that of ordinary Raman, it is promising for application in the enrichment and detection of trace PAHs.

AbstractPesticides are a safety issue globally and cause serious concerns for the environment, wildlife and human health. The handheld detection of four pesticide residues widely used in Basmati rice production using surface-enhanced Raman spectroscopy (SERS) is reported. Different SERS substrates were synthesised and their plasmonic and Raman scattering properties evaluated. Using this approach, detection limits for pesticide residues were achieved within the range of 5 ppb-75 ppb, in solvent. Various extraction techniques were assessed to recover pesticide residues from spiked Basmati rice. Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERs) acetate extraction was applied and characteristic spectral data for each pesticide was obtained from the spiked matrix and analysed using handheld-SERS. This approach allowed detection limits within the matrix conditions to be markedly improved, due to the rapid aggregation of nanogold caused by the extraction medium. Thus, detection limits for three out of four pesticides were detectable below the Maximum Residue Limits (MRLs) of 10 ppb in Basmati rice. Furthermore, the multiplexing performance of handheld-SERS was assessed in solvent and matrix conditions. This study highlights the great potential of handheld-SERS for the rapid on-site detection of pesticide residues in rice and other commodities.

This study aimed to optimize the detection conditions for surface-enhanced Raman spectroscopy (SERS) of single-stranded DNA (ssDNA) in four different buffers and explore the interaction between gonyautoxin (GTX1/4) and its aptamer, GO18. The influence of the silver colloid solution and MgSO4 concentration (0.01 M) added under four different buffered conditions on DNA SERS detection was studied to determine the optimum detection conditions. We explored the interaction between GTX1/4 and GO18 under the same conditions as those in the systematic evolution of ligands by exponential enrichment technique, using Tris-HCl as the buffer. The characteristic peaks of GO18 and its G-quadruplex were detected in four different buffer solutions. The change in peak intensity at 1656 cm−1 confirmed that the binding site between GTX1/4 and GO18 was in the G-quadruplex plane. The relative intensity of the peak at 1656 cm−1 was selected for the GTX1/4–GO18 complex (I1656/I1099) to plot the ratio of GTX1/4 in the Tris-HCl buffer condition (including 30 μL of silver colloid solution and 2 μL of MgSO4), and a linear relationship was obtained as follows: Y = 0.1867X + 1.2205 (R2 = 0.9239). This study provides a basis for subsequent application of SERS in the detection of ssDNA, as well as the binding of small toxins and aptamers.

Exosomes are small nanoscale vesicles with a double-layered lipid membrane structure secreted by cells, and almost all types of cells can secrete exosomes. Exosomes carry a variety of biologically active contents such as nucleic acids and proteins, and play an important role not only in intercellular information exchange and signal transduction, but also in various pathophysiological processes in the human body. Surface-enhanced Raman Spectroscopy (SERS) uses light to interact with nanostructured materials such as gold and silver to produce a strong surface plasmon resonance effect, which can significantly enhance the Raman signal of molecules adsorbed on the surface of nanostructures to obtain a rich fingerprint of the sample itself or Raman probe molecules with ultra-sensitivity. The unique advantages of SERS, such as non-invasive and high sensitivity, good selectivity, fast analysis speed, and low water interference, make it a promising technology for life science and clinical testing applications. In this paper, we briefly introduce exosomes and the current main detection methods. We also describe the basic principles of SERS and the progress of the application of unlabeled and labeled SERS in exosome detection. This paper also summarizes the value of SERS-based exosome assays for early tumor diagnosis.

We present a facile approach for the determination of the electromagnetic field enhancement of nanostructured TiN electrodes. As model system, TiN with partially collapsed nanotube structure obtained from nitridation of TiO2 nanotube arrays was used. Using surface-enhanced Raman scattering (SERS) spectroscopy, the electromagnetic field enhancement factors (EFs) of the substrate across the optical region were determined. The non-surface binding SERS reporter group azidobenzene was chosen, for which contributions from the chemical enhancement effect can be minimized. Derived EFs correlated with the electronic absorption profile and reached 3.9 at 786 nm excitation. Near-field enhancement and far-field absorption simulated with rigorous coupled wave analysis showed good agreement with the experimental observations. The major optical activity of TiN was concluded to originate from collective localized plasmonic modes at ca. 700 nm arising from the specific nanostructure.

Surface-enhanced Raman spectroscopy (SERS) is a promising tool that can be used in the detection of molecular changes triggered by disease development. Cardiovascular diseases (CVDs) are caused by multiple pathologies originating at the cellular level. The identification of these deteriorations can provide a better understanding of CVD mechanisms, and the monitoring of the identified molecular changes can be employed in the development of novel biosensor tools for early diagnostics. We applied plasmonic SERS nanosensors to assess changes in the properties of erythrocytes under normotensive and hypertensive conditions in the animal model. We found that spontaneous hypertension in rats leads (i) to a decrease in the erythrocyte plasma membrane fluidity and (ii) to a decrease in the mobility of the heme of the membrane-bound hemoglobin. We identified SERS parameters that can be used to detect pathological changes in the plasma membrane and submembrane region of erythrocytes.
