The actual product assembly process mainly relies on manual assembly by workers, and the personal experience of workers is difficult to effectively reuse. Ontology as a knowledge management and expression tool is gradually applied in the field of assembly. However, the manual construction of the ontology is time-consuming and labor-intensive, and the automatic construction of the ontology requires a large number of corpora for training, both of which are difficult to obtain a good assembly case ontology. This paper proposes a method in which automatically extracts relevant knowledge from case assembly process files to generates case database and integrates ontology framework of assembly domain to construct ontology. It shows that the accuracy can be guaranteed on the basis of the rapid construction of case ontology. The feasibility of this method is proved by a practical case.

Abstract Several strength grades of sulphoaluminate cement concrete were designed, and the mechanical properties with different mix ratios were studied. This paper mainly analyzes the mechanical properties of sulphoaluminate cement concrete with the setting time test, compressive strength test and flexural test. The test results show that the setting time of cement concrete can be controlled by mixing different admixtures. One hand, the initial setting time increases from 34 min to 340 min, and the final setting time increases from 57 min to 580 min when the incorporated borax content changes from 0 to 1.0 %. Other hand, the initial setting time decreased from 34 min to 11 min, and the final setting time increased from 57 min to 18 min, when the incorporated borax content changed from 0 to 0.5 %. Furthermore, the compressive strength can reach 40 MPa, and the flexural strength can reach more than 2.5MPa after 6 hours of curing. The experiment results illustrate that the setting time and the mechanical properties satisfy the needs of the rapid construction requirements under normal temperature conditions.

Abstract Background: Designer cellulosomes are self-assembled chimeric enzyme complexes that can be used to improve lignocellulosic biomass degradation. They are composed of a synthetic multimodular backbone protein, termed the scaffoldin, and a range of different chimeric docking enzymes that degrade polysaccharides. Over the years, several functional designer cellulosomes have been constructed. Since many parameters influence the efficiency of these multi-enzyme complexes, there is a need to optimise designer cellulosome architecture by testing combinatorial arrangements of docking enzyme and scaffoldin variants. However, the modular cloning procedures are tedious and cumbersome. Results: VersaTile is a combinatorial DNA assembly method, allowing the rapid construction and thus comparison of a range of modular proteins. Here, we present the extension of the VersaTile platform to facilitate the construction of designer cellulosomes. We have constructed a tile repository, composed of dockerins, cohesins, linkers, tags and enzymatically active modules. The developed toolbox allows us to efficiently create and optimise designer cellulosomes at an unprecedented speed. As a proof of concept, a trivalent designer cellulosome able to degrade the specific hemicellulose substrate, galactomannan, was constructed and optimised. The main factors influencing cellulosome efficiency were found to be the selected dockerins and linkers and the docking enzyme ratio on the scaffoldin. The optimised designer cellulosome was able to hydrolyse the galactomannan polysaccharide and release mannose and galactose monomers. Conclusion: We have eliminated one of the main technical hurdles in the designer cellulosome field and anticipate the VersaTile platform to be a starting point in the development of more elaborate multi-enzyme complexes.

To implement the balance system of cultivated land in occupation and supplement and to adhere to the principle of “supplement the occupied cultivated land of high quality with the one bearing same quality”, in the thesis, a field experiment was conducted to study the effects of woody peat on soil physical, chemical, and biological properties of the plough layer and its crop yield. Furthermore, the correlation between soil indexes and crop yield under the best fertilization mode through the addition of the natural material of woody peat instead of lengthy cultivation of the plough layer to rapidly construct a high-quality plough layer and solve the practical problem that the natural endowment of newly reclaimed cultivated land is far less than the occupied high-quality cultivated land was explored. The land remediation project of Fuping County, Hebei Province, was taken as the experimental area, and the five most representative and effective datasets were selected and studied. The results demonstrated that the addition of woody peat and rotten straw could reduce soil particle size and bulk density and alleviate soil viscosity and acidification. An increase in soil organic matter, soil microbial biomass carbon (MBC), alkali-hydrolyzable nitrogen, available phosphorus, and available potassium and a decrease in the heavy metal content were also observed. The results indicated that the application of woody peat achieved the expected effect of the rapid construction of a high-quality plough layer. The best mode of fertilization was A2, which provided a good reference for the rapid construction of a high-quality plough layer in the future. The analysis of the correlation between soil indexes and crop yield illustrated that the organic matter content, soil available nutrients, and crop yield had a significant positive correlation; the organic matter content and soil available nutrients showed the same tendency, which suggests that soil organic matter content and soil fertility level are closely related and that soil fertility plays a decisive role in crop yield under the same external conditions. Woody peat exerted an eminent influence on the organic matter content and soil available nutrients to determine the change in crop yield, which provides a reliable basis for future research on land improvement projects to increase crop yield.

Sulfur(VI) fluoride exchange (SuFEx) click chemistry provides a powerful tool for rapid construction of modular connections. Here, we report a novel catalytic silicon-free SuFEx reaction with sulfonyl fluorides. Under the catalysis of 10 mol% N-heterocyclic carbene (NHC), a range of phenols and alcohols react with different sulfonyl fluorides to afford sulfonate esters in 49-99% yields. In addition, Under the relay catalysis of 10 mol% N-heterocyclic carbene and 10 mol% 1-hydroxybenzotriazole (HOBt), a variety of primary and secondary amines react with different sulfonyl fluorides to produce sulfonamides in 58%-99% yields. More than 140 sulfonylated products, including 17 natural product derivatives have been prepared through this method. Mechanism study showed that NHCs might act as a carbon-centered Brønsted base to catalyse the SuFEx click reactions via the formation of hydrogen bonding with phenols or alcohols.
