The purpose of the research is study of pinniped helminth fauna in Chukotka, and the analysis of the fish of the main commercial families infected with pathogens of helminthozoonoses based on modern literature.Materials and methods. The helminths were collected in autumn of 2019 from pinnipeds caught in the Mechigmenskaya Guba of the Bering Sea in the Chukotka Autonomous Okrug by the method of partial helminthological dissection per Skryabin (gastrointestinal tract). Samples were examined from 6 walruses and 26 seals (13 spotted seals and 13 ringed seals). The helminths found were fixed in 70% alcohol. The helminth species were identified at the Department of Parasitology and Veterinary and Sanitary Examination of the MVA named after K. I. Skryabin using reference literature.Results and discussion. All pinnipeds were infected with nematodes of the family Anisakidae. Mature Pseudoterranova desipiens were found in the walrus (Infection Prevalence = 16.7% with Infection Intensity = 3 specimens/animal), mature Ps. desipiens, as well as Contracoecum osculatum and Anisakis simplex larvae (IP = 30.8% with II from 5 to 57 specimens) were found in the spotted seal, and Ps. desipiens larvae and mature Ps. desipiens were found in the ringed seal (IP = 15.4% with II from 1 to 4 specimens). Thus, only Ps. desipiens were represented by mature stages (females and males), and two other species of anisakids, C. osculatum and A. simplex, were found in the seals in the larval stage.

Abstract Spotted seal (Phoca largha) is a critically endangered pinniped in China and South Korea. The conventional method to protect and maintain the P. largha population is to keep them captive in artificially controlled environments. However, little is known about the physiological differences between wild and captive P. largha. To generate a preliminary protein expression profile for P. largha, whole blood from wild and captive pups were subjected to a label-free comparative proteomic analysis. According to the results, 972 proteins were identified and predicted to perform functions related to various metabolic, immune, and cellular processes. Among the identified proteins, the expression level of 51 were significantly different between wild and captive P. large pups. These differentially expressed proteins were enriched in a wide range of cellular functions, including cytoskeleton, phagocytosis, proteolysis, the regulation of gene expression, and carbohydrate metabolism. The abundances of proteins involved in phagocytosis and ubiquitin-mediated proteolysis were significantly higher in the whole blood of wild P. largha pups than in captive individuals. In addition, heat shock protein 90-beta, were determined as the key protein associated with the differences in the wild and captive P. largha pups due to the most interactions of it with various differentially expressed proteins. Moreover, wild P. largha pups could be more nutritionally stressed and have more powerful immune capacities than captive pups. This study provides the first data on the protein composition of P. largha and provides useful information on the physiological characteristics for research in this species.

Rescuing seal pups is an important conservation action for spotted seals Phoca largha. Gut microbiota are directly associated with host health and diet metabolism. Therefore, knowledge gained from gut microbiota variations of spotted seal pups held in captivity after rescue can help formulate comprehensive rescue plans for the future. In this study, we collected feces from a rescued spotted seal pup every 3 d during the rescue process. Fecal bacterial communities were measured by high-throughput sequencing based on 16S rRNA amplification. Firmicutes were the most dominant bacteria, comprising >70% of the total gut microbiota. Moreover, differences in fecal bacterial communities of the rescued spotted seal pup between rescue and release were compared. At release, the abundances of 2 potential bacteria related to gut health, Blautia producta and Cetobacterium somerae, were remarkably lower, while Clostridium perfringens, a key mammalian pathogen, was significantly higher in the feces of the released pup. Moreover, the pup experienced a bout of diarrhea during its time in captivity, which resulted in a momentary change in its gut microbiota. Fusobacterium was recognized as a potential causative pathogen for the diarrhea. This study contributes to our understanding of gut microbiota variations in spotted seal pups during the rescue period.
