Gastric ulcer is an excavation of mucosa and extending to submucosal layers due to increased gastric secretion, generation of free radicals and imbalance between protective factors and aggravating factors. The present study aimed to screen yttrium and copper oxide nanoparticles for antiulcerogenic activity. Metal oxide nanoparticles were evaluated for various biological activities considering their high surface area to volume ratio, different oxidative states, cell permeability etc. Various studies have proved metal oxide nanoparticles to be beneficial in various ailments due to their antioxidant, antibacterial, and other pharmacological activities. Nanoparticles prepared using biogenic methods were relatively more biocompatible and less toxic compared to conventional methods of synthesis. In the present study, green synthesized yttrium and copper oxide nanoparticles were evaluated for acute toxicity, antioxidant, proton pump inhibition and antiulcerogenic activity. In vitro antioxidant activity by DPPH, nitric oxide scavenging methods and H+K+ATPase assay was performed to evaluate the mechanism of action. Synthesized nanoparticles have shown no signs of acute toxicity as per OECD 423. Pyloric ligation method was performed to evaluate gastric volume, pH and ulcer severity. The present study revealed the dose-dependent antiulcer potential of yttrium oxide nanoparticles at doses of 2mg/kg and 20mg/kg. Antisecretory action of yttrium and copper oxides were evident from in vitro H+K+ATPase activity and inhibition of gastric volume. Antioxidant activities of yttrium oxide nanoparticles indicate gastric mucosal protection by free radical scavenging action. Copper oxide nanoparticles produced antisecretory activity but produced mucosal damage and hemorrhage at a dose of 20mg/kg.

The inflamed break in the skin or mucous membrane lining the elementary tract describes development of ulcer. About 10% of World’s population suffering from peptic ulcer. It is a broad term that includes ulcers of digestive tract, in the stomach or the duodenum. The presence of acid and peptic activity in gastric juice with a breakdown in mucosal defences develops peptic ulcer. It may be occurring due to regular usage of drugs, irregular food habits, stress. The ideal aims of treatment of peptic ulcer disease are to relieve pain, heal the ulcers and delay ulcer recurrence. The availability of number of synthetic drugs such as proton pump inhibitors (PPIs) and histamine-2 (H2) receptor antagonists are conventionally used for treatments of peptic ulcers, associated with adverse effects, relapses, various drug interactions are observed and expensive when compared to herbal medicines. Herbal medicines demand has increased globally. Availability of gastro-protective remedies without side effects are excellent resources for costeffective medicines. Natural compounds showed significant antiulcerogenic activity, compounds such as tannins, flavonoids, alkaloids, triterpenoids, steroids, saponins, and coumarins. The current review states some medicinal plants, being used in Ayurveda or modern science for the treatment or prevention of peptic ulcer and comparison with synthetic drugs.

Objective: To study the morphological and histological effects of vitamin E on gastric lesions produced by indomethacin.Materials and Methods: This was an animal interventional study, 48 adult healthy albino mice were selected and were split into four groups A, B, C and D. Number of animals in each group was 12. Group A was categorized as control. Evion 400mg/kg was administered to Group B. Indocid 25mg/kg was given to group C. Indocid and Evion both (25mg/kg &400 mg/kg respectively) were administered to Group D. In all group, six animals were selected and treated for three days and rest for eight days with calculated doses of drugs. Mice were sacrificed and dissection was done 24 hours after the last dose. The stomach was identified, washed, and observed under dissecting microscope to study the number and shape of ulcers. The dimension of ulcers was measured under a compound microscope.Results: No ulcers were seen in groups A and B. 35 and 10 ulcers were observed in groups C and D respectively. The mean number of ulcers in groups C and D was statistically significant (p-value=0.000). In comparison to group D, Pindot, linear, Irregular, and punched-out ulcers were more prevalent in group C and were statistically significant (p-value < 0.05). The mean linear dimension of ulcers in group C was much greater than in group D. The mean dimension of ulcers in group C1 and C2 was 262.50µm and 232.5µm respectively. Whereas in group D1 and D2 given both Indocid and vitamin E the dimensions were 56.5µm and 50µm.Conclusion: Vitamin E has an anti-ulcerogenic effect on stomach mucosa by reducing the number and dimension of ulcers.

The phytochemical constituents and the antiulcerogenic activity of methanolic extract from Ipomoea asarifolia leaf were investigated to ascertain its medicinal potentials. The study revealed that the leaf contained saponins, tannins, alkaloids, terpenoids and eugenols. The methanolic extract was used as a protective agent against gastric ulcer in Wistar rats, using the experimental model of gastric ulcer (lesion) induced by ethanol. The ulcer count for the methanol extract ranged from 0.00 to 2.00, while the ulcer index ranged between 0.33 to 2.00 for all concentrations (100, 200 and 400mg/kg). Percentage inhibition at 100, 200 and 400mg/kg were respectively 25.09%, 12.73% and 87.64%. The extract prevented acute gastric mucosal injury induced by absolute ethanol in a non dose-dependent manner. This is because the protective action was observed at the lowest and highest doses, but not an intermediate dose of the extract. The results obtained indicate that the leaf extract has antiulcerogenic importance and may be utilized for the treatment of gastric ulcers.

AbstractAllantoin (ALL) is a phytochemical possessing an impressive array of biological activities. Nonetheless, developing a nanostructured delivery system targeted to augment the gastric antiulcerogenic activity of ALL has not been so far investigated. Consequently, in this survey, ALL-loaded chitosan/sodium tripolyphosphate nanoparticles (ALL-loaded CS/STPP NPs) were prepared by ionotropic gelation technique and thoroughly characterized. A full 24 factorial design was adopted using four independently controlled parameters (ICPs). Comprehensive characterization, in vitro evaluations as well as antiulcerogenic activity study against ethanol-induced gastric ulcer in rats of the optimized NPs formula were conducted. The optimized NPs formula, (CS (1.5% w/v), STPP (0.3% w/v), CS:STPP volume ratio (5:1), ALL amount (13 mg)), was the most convenient one with drug content of 6.26 mg, drug entrapment efficiency % of 48.12%, particle size of 508.3 nm, polydispersity index 0.29 and ζ-potential of + 35.70 mV. It displayed a sustained in vitro release profile and mucoadhesive strength of 45.55%. ALL-loaded CS/STPP NPs (F-9) provoked remarkable antiulcerogenic activity against ethanol-induced gastric ulceration in rats, which was accentuated by histopathological, immunohistochemical (IHC) and biochemical studies. In conclusion, the prepared ALL-loaded CS/STPP NPs could be presented to the phytomedicine field as an auspicious oral delivery system for gastric ulceration management.
