Epidemiological evidence indicates that plant antioxidants activity can treat or help to prevent the development of various diseases. One species with great potential as an antioxidant is Curcuma longa. However, different extraction techniques can influence isolated chemical compounds. This study investigated chemical composition and antioxidant activity of two rhizome extracts of C. longa: hydroethanolic, obtained by exhaustion (HECLex); and dried by a spray dryer (HECLsd). The phytochemical composition was evaluated by GC/MS. Antioxidant activity was evaluated using DPPH and FRAP assays. Total phenolic compounds and soil analyses were performed. The main components of HECLex were ar-turmerone, γ-curcumene, α-turmerone, and β-sesquiphellandrene. The main components of HECLsd were 9,12,15-octadecatrienoic acid, 2,3-bis([trimethylsilyl]oxy)propyl ester, verrucarol, and 1-monolinoleoylglycerol trimethylsilyl ether. HECLsd had significantly higher levels of phenolic compounds and higher antioxidant capacity compared with HECLex. In conclusion, processes of the preparation of C. longa rhizomes alter the chemical components and consequently their biological activity.

Many techniques are used today to study insect morphology, including light and electron microscopy. Most of them require to specifically prepare the sample, precluding its use for further investigation. In contrast, micro-CT allows a sample to be studied in a non-destructive and rapid process, even without specific treatments that might hinder the use of rare and hard-to-find species in nature. We used synchrotron radiation (SR) micro-CT and conventional micro-CT to prepare 3D reconstructions of Diptera, Coleoptera, and Hymenoptera species that had been processed with 4 common preparation procedures: critical-point drying, sputter-coating, resin embedding, and air-drying. Our results showed that it is possible to further utilize insect samples prepared with the aforementioned preparation techniques for the creation of 3D models. Specimens dried at the critical point showed the best results, allowing us to faithfully reconstruct both their external surface and their internal structures, while sputter-coated insects were the most troublesome for the 3D reconstruction procedure. Air-dried specimens were suitable for external morphological analyses, while anatomical investigation of soft internal organs was not possible due to their shrinking and collapsing. The sample included in resin allowed us to reconstruct and appreciate the external cuticle and the internal parts. In this work, we demonstrate that insect samples destined to different analyses can be used for new micro-CT studies, further deepening the possibility of state-of-the-art morphological analyses.

Abstract Indigenous knowledge on medicinal plants and practices is outdistancing and vulnerable to loss if not properly documented. A survey on medicinal plants and their practices was carried in the Myagdi district. Around 40 tribal people from four different villages were interviewed using a semi-structured questionnaire. Documentation of the indigenous knowledge was done in written form and the pictures of available medicinal plants were taken. The study showed that 93.51% of the respondent has used medicinal plants and 87.20% found them effective in curing several diseases and injuries. They are commonly used to cure diseases like bone fractures, abdominal pain, fever, common cold, dysentery, eye opacity, scabies, worm infection, reproductive problems, mental disorder, and cardiovascular problems. As reported, with access to modern pharmaceuticals, the use of medicinal plants has been less practiced these days. The knowledge on medicinal plants and practices are restricted to older-aged groups of the community (76.32%) reflecting that valuable indigenous knowledge is on the wane. This study portrays the commonly used medicinal plants along with their preparation techniques practiced in the study area.

Orally disintegrating tablets (ODTs) rapidly disintegrate or dissolve in the oral cavity without using water. Demand for ODTs has increased, and the field has overgrown in the pharmaceutical industry and academia. It is reported that ODTs have several advantages over other conventional tablets. Since some of them are absorbed from the mouth, pharynx, and esophagus as the saliva passes down into the stomach, in such cases, the bioavailability of the drug improves meaningfully. Furthermore, the immediate release property of ODTs makes them a popular oral dosage form in patients with swallowing challenges, children, and for cases with a need for rapid onset of action. The current review article explains the features of active ingredients and excipients used in the formulation of ODTs, discusses multiple ODT formulation and preparation techniques with their merits and demerits, and also, offers remedies for problems associated with ODTs. Moreover, quality control steps and required considerations are presented.

Tillage is an important factor affecting different soil properties and crop yields. Tillage relates to applying forces to soil using different implements for preparation of a proper seed bed. For rice crop, land preparation is a rigorous and time consuming operation that needs special attention for preparing a puddled soil condition to transplant the weak and tender rice seedlings. A field experiment was established under agro-ecological conditions of Sheikhupura to evaluate different land preparation techniques during fall in 2017 and 2018 years. The experiment was performed at Adaptive Research Farm Sheikhupura. It was laid out in randomized complete block design (RCBD) with four different land preparation techniques including T1= Cultivator (4 times) + Planking (2 times) (Farmer’s Practice), T2=MB plough (1 time) + Disc Harrow (1 time) + Planking (2 Times), T3=MB plough (1 time) + Cultivator (2 times) + Planking (2 Times) and T4= Rotavator (1 time) + Disc Harrow (1 time) + Planking (2 Times), each treatment replicated thrice during both the years. All the other agronomic and crop husbandry operations were kept uniform throughout the growing season every year. The results revealed that preparing land with MB plough (1 time), Disc Harrow (1 time) and Planking (2 times) is the best land preparation method can increase the paddy yield 13.5-17.5% as compared to the traditional method practised by the farmers. Moreover, highest plant height (134.00 cm and 132.00 cm), number of tillers (224 and 220 m-2), number of grains per spike (130 and 116) and 1000-grain weight (23.0 and 22.0 g) respectively during Kharif-18 and Kharif-19 was also recorded in the same treatment. So, preparation of land for paddy in agro-ecological conditions of Sheikhupura using MB plough (1 time), Disc Harrow (1 time) and Planking (2 times) can significantly enhance the paddy yield.

Moisture is a ubiquitous and clean resource in nature, which continuously diffuses in the atmosphere and demonstrates huge chemical potential energy that is difficult to be utilized. Recently, the generation of power from interactions between graphene and gaseous water molecules in moisture has triggered great research interest that could provide a novel energy conversion system for our society. graphene-based assemblies have been considered as ideal platforms for moist-electric generation (MEG) in many studies, because of the abundant of functional groups, controllable microstructure and diverse macro morphologies. Therefore, in this short review, we will first state the preparation techniques of graphene-based assemblies for MEG. Then, the fundamental mechanisms of MEG are discussed and the latest advances on graphene MEG are reviewed. Finally, an overview of the current challenges and future development trends in graphene MEG is provided.

While sample preparation techniques for the chemical and biochemical analysis of tissues are fairly well advanced, the preparation of complex, heterogenous samples for single-cell analysis can be difficult and challenging. Nevertheless, there is growing interest in preparing complex cellular samples, particularly tissues, for analysis via single-cell resolution techniques such as single-cell sequencing or flow cytometry. Recent microfluidic tissue dissociation approaches have helped to expedite the preparation of single cells from tissues through the use of optimized, controlled mechanical forces. Cell sorting and selective cellular recovery from heterogenous samples have also gained traction in biosensors, microfluidic systems, and other diagnostic devices. Together, these recent developments in tissue disaggregation and targeted cellular retrieval have contributed to the development of increasingly streamlined sample preparation workflows for single-cell analysis technologies, which minimize equipment requirements, enable lower processing times and costs, and pave the way for high-throughput, automated technologies. In this chapter, we survey recent developments and emerging trends in this field.
