AbstractThe abundance and distribution of intertidal canopy-forming macroalgae are threatened by the increase in sea surface temperature and in the frequency and intensity of heatwaves caused by global warming. This study evaluated the physiological response of predominant intertidal macroalgae in the NW Iberian Peninsula (Bifurcaria bifurcata, Cystoseira tamariscifolia and Codium tomentosum) to increased seawater temperature during immersion and increased air temperatures during consecutive emersion cycles. We combined field mensuration and laboratory experiments in which we measured mortality, growth, maximum quantum yield and C:N content of the macroalgae. Air temperature was a critical factor in determining physiological responses and survivorship of all species, whereas high seawater temperature had sublethal effects. Cystoseira tamariscifolia suffered the greatest decreases in Fv/Fm, growth and the highest mortality under higher air temperatures, whereas C. tomentosum was the most resistant and resilient species. Two consecutive cycles of emersion under atmospheric heatwaves caused cumulative stress in all three macroalgae, affecting the physiological performance and increasing the mortality. The potential expansion of the warm-temperate species B. bifurcata, C. tamariscifolia and C. tomentosum in the NW Iberian Peninsula in response to increasing seawater temperature may be affected by the impact of increased air temperature, especially in a region where the incidence of atmospheric heatwaves is expected to increase.

Mycosporines and mycosporine-like amino acids have been described as natural sunscreens and antioxidant compounds presenting a great potential for health and cosmetic applications. Herein, an untargeted screening approach for mycosporines and mycosporine-like amino acids (MAAs) was developed by the coupling of zwitterionic hydrophilic interaction liquid chromatography (HILIC) with multistage electrospray mass spectrometry MS2/MS3 using an Orbitrap analyzer and fragment ion search (FISh). This method was applied to study the mycosporine and MAA contents of five algae extracted using a 50% methanol solution and sonication. Candidate-MAAs were detected by mining eight characteristic fragment ions in their HILIC data-dependent MS2 mass spectrum. Their exact masses were measured with 3 ppm mass accuracy and their structures were elucidated on the basis of the MS3/MS4 mass spectra. The method developed was validated with a targeted analysis using an extract of Gymnogongrus devoniensis which confirmed the detection of 14 MAAs reported in the literature. In addition, 23 previously unreported MAAs were detected and the structures could be assigned for seven of them. The developed method was applied to the analysis of four algae: Gelidium sesquipedale, Halopithys incurva, Porphyra rosengurtii and Cystoseira tamariscifolia allowing the detection of MAAs, including some reported here for the first time.

The application of natural compounds extracted from seaweeds is a promising eco-friendly alternative solution for harmful algae control in aquatic ecosystems. In the present study, the anti-cyanobacterial activity of three Moroccan marine macroalgae essential oils (EOs) was tested and evaluated on unicellular Microcystis aeruginosa cyanobacterium. Additionally, the possible anti-cyanobacterial response mechanisms were investigated by analyzing the antioxidant enzyme activities of M. aeruginosa cells. The results of EOs GC–MS analyses revealed a complex chemical composition, allowing the identification of 91 constituents. Palmitic acid, palmitoleic acid, and eicosapentaenoic acid were the most predominant compounds in Cystoseira tamariscifolia, Sargassum muticum, and Ulva lactuca EOs, respectively. The highest anti-cyanobacterial activity was recorded for Cystoseira tamariscifolia EO (ZI = 46.33 mm, MIC = 7.81 μg mL−1, and MBC = 15.62 μg mL−1). The growth, chlorophyll-a and protein content of the tested cyanobacteria were significantly reduced by C. tamariscifolia EO at both used concentrations (inhibition rate >67% during the 6 days test period in liquid media). Furthermore, oxidative stress caused by C. tamariscifolia EO on cyanobacterium cells showed an increase of the activities of superoxide dismutase (SOD) and catalase (CAT), and malondialdehyde (MDA) concentration was significantly elevated after 2 days of exposure. Overall, these experimental findings can open a promising new natural pathway based on the use of seaweed essential oils to the fight against potent toxic harmful cyanobacterial blooms (HCBs).
