We describe the actinomycete strain IMV-70 isolated from the soils of Kazakhstan, which produces potent antibiotics with high levels of antibacterial activity. After the research of its morphological, chemotaxonomic, and cultural characteristics, the strain with potential to be developed further as a novel class of antibiotics with chemotherapeutics potential was identified asStreptomycessp. IMV-70. In the process of fermentation, the strainStreptomycesspp. IMV-70 produces the antibiotic no. 70, which was isolated from the culture broth by extraction with organic solvents. Antibiotic compound no. 70 was purified and separated into individual components by HPLC, TLC, and column chromatography methods. The main component of the compound is the antibiotic 70-A, which was found to be identical to the peptolide etamycin A. Two other antibiotics 70-B and 70-C have never been described and therefore are new antibiotics. The physical-chemical and biological characteristics of these preparations were described and further researched. Determination of the optimal growth conditions to cultivate actinomycete-producer strain IMV-70 and development of methods to isolate, purify, and accumulate preparations of the new antibiotic no. 70 enable us to research further the potential of this new class of antibiotics.

The objective of this study was to determine the effects of ozone on membrane lipids and on the electron-density patterns of cell membranes in electron micrographs. Analysis of fatty acids from tobacco leaves fumigated with ozone indicated that there was no significant difference between the ozone-treated and the control plants in the relative amounts of the fatty acids. This suggests that if the primary site of ozone action is unsaturated lipids in membranes then the amounts of affected unsaturated fatty acids are too small to be detected by gas chromatography. In support of this, characteristic electron-microscopic images of membranes are observed in cells of fumigated leaves. However, measurements of the length and width of the chloroplasts and the determination of axial ratios indicated that the ozone treatment resulted in a shrinkage of the chloroplasts. In contrast, mitochondrial changes are apparently explained in terms of ozone-induced swelling.

Currently available techniques for the separation and characterization of different glycerolipids are complicated and/or time consuming. By modulating the stationary phase in a solid-phase extraction (SPE) manifold, efficient and rapid separation of plant membrane lipids was achieved. The glycerolipids from rice and maize tissues were separated into seven classes (monogalactosyldiacylglycerol, digalactosyldiacylglycerol, phosphatidylethanolamine, phosphatidylcholine, sulphoquinovosyldiacylglycerol, phosphatidylinositol and phosphatidylglycerol). The pigments present in the rice and maize leaves and rice stems were successfully removed from the total lipid extracts. Pigment-free plant tissue (rice roots) was also analysed. The fatty acid profile of each lipid class isolated by SPE agreed well with those obtained by other separation techniques. The recovery of glycerolipids was at least 87°.

AbstractThe aim of the study was to investigate the effect of application of exogenous hyaluronic acid (HA) of various molecular weights on the cells of human immune system. Two cell lines HL-60 and U-937 with various ability to differentiation were chosen. HL-60 cells were differentiated to macrophages and granulocytes, whereas U-937 only to macrophages. For all investigated cell systems the most cytotoxic effect, indicated as a decrease of cell viability, was found at HA dose equal to 200 mg/L. However, greater effect was observed for differentiated cells and at longer exposure to HA. The possibility of HA interaction with both specific receptors and membrane lipids was tested by determination of biotin-labelled HA binding to cell surface and analysis of physicochemical parameters of model membranes.

A mass spectrometric method is described for the quantitative determination of dimers of polyunsaturated fatty acids (PUFA) formed in the hepatic endoplasmic reticulum of rats upon inhalation of tetrachloromethane. The results show that dimers account for a considerable fraction of microsomal PUFA which disappear during CCl4 metabolism. Cross-linking of the membrane lipids of the endoplasmic reticulum seems to be a significant process with respect to cell toxicity.

It is now possible to determine proper motions of high-velocity objects in such a way as to obtain with some accuracy the velocity vector relevant to the Sun. If a potential field of the Galaxy is assumed, one can compute an actual orbit. A determination of the velocity of the globular clusterωCentauri has recently been completed at Greenwich, and it is found that the orbit is strongly retrograde in the Galaxy. Similar calculations may be made, though with less certainty, in the case of RR Lyrae variable stars.
