In situ hybridization (ISH) with biotin-labeled probes is increasingly used in histology, histopathology and molecular biology, to detect genetic nucleic acid sequences of interest, such as viruses, genetic alterations and peptide-/protein-encoding messenger RNA (mRNA). In situ polymerase chain reaction (PCR) (PCR in situ hybridization = PISH) and the new in situ self-sustained sequence replication-based amplification (3SR) method even allow the detection of single copies of DNA or RNA in cytological and histological material. However, there is a number of considerable problems with the in situ PCR methods available today: False positives due to mis-priming of DNA breakdown products contained in several types of cells causing non-specific incorporation of label in direct methods, and re-diffusion artefacts of amplicons into previously negative cells have been observed. To avoid these problems, super-sensitive ISH procedures can be used, and it is well known that the sensitivity and outcome of these methods partially depend on the detection system used.

An atmospheric correction algorithm for medium-resolution satellite data over general water surfaces (open/coastal, estuarine and inland waters) has been assessed in Australian coastal waters. In situ measurements at four match-up sites were used with 21 Landsat 8 images acquired between 2014 and 2017. Three aerosol sources (AERONET, MODIS ocean aerosol and climatology) were used to test the impact of the selection of aerosol optical depth (AOD) and Ångström coefficient on the retrieved accuracy. The initial results showed that the satellite-derived water-leaving reflectance can have good agreement with the in situ measurements, provided that the sun glint is handled effectively. Although the AERONET aerosol data performed best, the contemporary satellite-derived aerosol information from MODIS or an aerosol climatology could also be as effective, and should be assessed with further in situ measurements. Two sun glint correction strategies were assessed for their ability to remove the glint bias. The most successful one used the average of two shortwave infrared (SWIR) bands to represent sun glint and subtracted it from each band. Using this sun glint correction method, the mean all-band error of the retrieved water-leaving reflectance at the Lucinda Jetty Coastal Observatory (LJCO) in north east Australia was close to 4% and unbiased over 14 acquisitions. A persistent bias in the other strategy was likely due to the sky radiance being non-uniform for the selected images. In regard to future options for an operational sun glint correction, the simple method may be sufficient for clear skies until a physically based method has been established.

Abstract Background Mammographic microcalcifications are considered early signs of breast cancer (BC). We examined the association between microcalcification clusters and the risk of overall and subtype-specific BC. Furthermore, we studied how mammographic density (MD) influences the association between microcalcification clusters and BC risk. Methods We used a prospective cohort (n = 53,273) of Swedish women with comprehensive information on BC risk factors and mammograms. The total number of microcalcification clusters and MD were measured using a computer-aided detection system and the STRATUS method, respectively. Cox regressions and logistic regressions were used to analyse the data. Results Overall, 676 women were diagnosed with BC. Women with ≥3 microcalcification clusters had a hazard ratio [HR] of 2.17 (95% confidence interval [CI] = 1.57–3.01) compared to women with no clusters. The estimated risk was more pronounced in premenopausal women (HR = 2.93; 95% CI = 1.67–5.16). For postmenopausal women, microcalcification clusters and MD had a similar influence on BC risk. No interaction was observed between microcalcification clusters and MD. Microcalcification clusters were significantly associated with in situ breast cancer (odds ratio: 2.03; 95% CI = 1.13–3.63). Conclusions Microcalcification clusters are an independent risk factor for BC, with a higher estimated risk in premenopausal women. In postmenopausal women, microcalcification clusters have a similar association with BC as baseline MD.

The in-situ gamma-ray spectrometry is a well suited method for seabed mapping applications, since it provides rapid results in a cost effective manner. Moreover, the in-situ method is preferable to the commonly applied laboratory measurements, due to its beneficial characteristics. Therefore, the development of in-situ systems for seabed measurements continuously grows. However, an efficiency calibration of the detection system is necessary for obtaining quantitative results in the full spectral range. In the present work, an approach for calculating the full-energy peak efficiency of an underwater insitu spectrometer for measure- ments on the seabed is presented. The experimental work was performed at the coastal site of Vasilikos (Cyprus). The experimental full-energy peak efficiency of the in-situ was determined in the energy range 1400–2600 keV, by combining the in-situ and laboratory reference measurements. The experimental effi- ciency results were theoretically reproduced by means of Monte Carlo (MC) simulations, using the MCNP5 code.

A regional ocean reanalysis system of China coastal waters and adjacent seas, called CORA (China ocean reanalysis), has been recently developed at the National Marine Data and Information Service (NMDIS). In this study, based on CORA, the impact of Argo profiles on the regional reanalysis is evaluated using a twin-experiment approach. It is found that, by assimilating Argo observations, the reanalysis quality is much improved: the root mean square (RMS) error of temperature and salinity can be further reduced by about 10% and the RMS error of current can be further reduced by 18%, compared to the case only assimilating conventional in situ temperature and salinity observations. Consistent with the unique feature of Argo observations, the temperature is improved in all levels and the largest improvement of salinity happens in the deep ocean. Argo profile data have a significant impact on the regional ocean reanalysis through improvements of both hydrographic and dynamic fields.
