Antimicrobial activity of enterocins from Enterococcus faecalis SL-5 against Propionibacterium acnes, the causative agent in acne vulgaris, and its therapeutic effect

Acne vulgaris is one of the most common health problem where medical treatment is sought in adults worldwide. It has been long described the integral role of Propionibacterium acnes in the pathogenesis of this disease. In this study, a group of local herbs known for their antimicrobial effects were selected for the evaluation of potential anti-acnes effects in vitro. Phenolics and flavonoid contents of methanolic extracts of Eucalyptus globulus, Mentha rotundifolia, Inula viscosa, Utrica dioica, Malva sylvestris, Quercus calliprinos, Arum palaestinum and Achille aodorata collected from different regions in Jordan during 2016-2017 were screened for antimicrobial activity against Propionibacterium acnes by disc diffusion and by broth microdilution method. Measurement of release of interleukin 1 alpha from human skin explants by ELISA was used for the evaluation of anti-inflammatory effects of the herbal preparations and extracts. M. rotundifolia and E. globulus, showed the highest phenolic and flavonoid contents in contrast to M. sylvestris which showed the least phenolic contents. Moreover, polyphenolic fractions exhibited modest anti-acne activity of herbal extracts of E. globulus and A. palaestinum (MIC 0.125 mg/ml), U. dioica (0.25 mg/ml) and I. viscosa (0.5 mg/ml), compared to not significant antimicrobial activity for others (MIC >1mg/ml). Regarding anti-inflammatory effects of the tested fractions, E. globulus and A. palaestinum extracts showed inhibition of interleukin 1 alpha release by more than 60 % for concentrations of 0.5 mg/ml respectively. The presence of anti-inflammatory and anti-acne activities in the polyphenolic extracts of local medicinal plants would increase the potential of using these herbs in the control of Acne vulgaris.

AbstractVarious Achillea species are rich in bioactive compounds and are important medicinal plants in phytotherapy. In the present study, Achillea millefolium L., Achillea moschata Wulfen, and Achillea atrata L. were compared with respect to their phenolic profile and antibacterial activity against gram-positive bacteria strains (Staphylococcus, Propionibacterium). Particular focus was given to A. atrata, which has hardly been studied so far. Based on the metabolite profile, A. atrata exhibited more similarities to A. moschata than to A. millefolium. The former two only differed in the occurrence of four compounds. The flavonols syringetin-3-O-glucoside and mearnsetin-hexoside, not reported for an Achillea species before, have been detected in A. atrata and A. moschata. All Achillea species reduced growth of the tested bacteria. A. atrata demonstrated highest activity against Propionibacterium acnes and Staphylococcus epidermidis, both being involved in the pathogenesis of acne vulgaris. Furthermore, A. atrata has a pronounced anti–methicillin-resistant Staphylococcus aureus potential. Bioassay-guided fractionation revealed that only the most polar fraction of A. moschata displayed antimicrobial activity, which was attributed to phenolics such as apigenin, centaureidin, and nevadensin, being present in high amounts in A. atrata. Thus, this alpine species shows promising antimicrobial activity and might be a potential source for developing novel dermal/topical drugs.

Jerawat  didefinisikan  sebagai  peradangan  kronik  dari  folikel  polisebasea  yang disebabkan  oleh  beberapa  faktor  dengan  gambaran  klinis  yang  khas.  Salah  satu tanaman  yang  dapat  mengatasi  jerawat  yaitu semangka.  Kulit buah semangka (Citrullus lanatus (Thunb.) Matsum & Nakai) memiliki   senyawa antibakteri di antaranya  alkaloid,  fenol,  saponin,  dan  terpenoid.  Dalam penelitian ini ekstrak limbah  kulit  buah semangka diformulasikan   dalam   sediaan   gel.   Tahap   penelitian   yang menggunakan  rancangan  acak  lengkap ini meliputi  penyiapan  dan  pengumpulan simplisia   limbah   kulit   buah   semangka,   ekstraksi,   uji kandungan senyawa, formulasi gel, evaluasi sifat fisik sediaan gel, dan uji aktivitas antibakteri  terhadap  bakteri  Propionibacterium acnes  dan  Staphylococcus  aureus. Ekstraksi yang dilakukan adalah dengan menggunakan metode maserasi dengan penyari kloroform. Uji kandungan senyawa dilakukan dengan menggunakan metode penapisan fitokimia meliputi uji alkaloid, uji triterpenoid, uji fenol, dan uji saponin. Ekstrak kulit buah semangka dengan konsentrasi 5, 10, dan 15% kemudian diformulasikan dengan carbopol 940 dan dilanjutkan dengan evaluasi sifat fisik gel, di antaranya uji organoleptis, uji homogenitas, uji daya sebar, uji daya lekat, uji pH, dan uji viskositas. Uji aktivitas antibakteri dari formulasi ekstrak kulit buah semangka dilakukan dengan menggunakan metode difusi agar. Hasil formulasi terbaik pada formulasi gel menghasilkan gel yang homogen, lekat, menyebar, pH yang cocok dengan kulit, dan memiliki viskositas yang cukup baik. Hasil terbaik dari zona hambat uji antibakteri pada bakteri Propionibacterium acnes yaitu 5,23 mm dan pada bakteri Staphylococcus  aureus yaitu 5,80 mm.

The medicinal value of the plants is due to their chemical components that bring a definite physiological action on the human body to prevent the diseases. In this work, we investigated the antimicrobial activity of leaves’ extracts of Quercus robur L., collected from the Algerian upper highlands, on ten bacterial strains and one fungal strain known to be pathogenic. First, we performed a qualitative phytochemical analysis, and second, antimicrobial activity tests performed by agar diffusion method (disc and well) with the determination of MIC by broth macro-dilution method. Given the results, it appears that obtained macerates of Quercus robur L. were rich in bioactive phytoconstituents such as alkaloids, anthraquinones, saponins, tannins, and other components. The yield of aqueous and methanolic macerates of leaves was 8.5 ± 1.41 and 22.4 ± 4.36%, respectively. The bacterial resistance was relatively important to several antibiotics, namely, ampicillin, amoxicillin + clavulanic acid for strains of Escherichia coli and Salmonella sp. However, Staphylococcus aureus strains were resistant to fusidic acid, penicillin, and oxacillin; while Enterococcus faecalis was resistant to fusidic acid, penicillin, oxacillin, and ticarcillin. The antibacterial activity of the macerates toward tested microbial strains showed that the aqueous and methanolic macerates of the leaves were proportional to the tested concentration and active not only against Gram-positive and Gram-negative bacteria but also on the fungal species Candida albicans. The estimated MIC for Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus was in the order of 10 mg/mL, which seems more effective than toward Salmonella sp., Klebsiella pneumoniae, Pseudomonas aeruginosa, and Candida albicans which were in the order of 30 mg/mL. These preliminary results confirm that the part of the studied plant had a very good antimicrobial activity that was proportional to the serial concentrations of the tested extracts.
