The In vitro Evaluation of the Antimicrobial Activity of Quercus robur L. Methanolic and Aqueous Leaves’ Extracts, from the Algerian High Plateaus Against some Uropathogenic Microbial Strains

The medicinal value of the plants is due to their chemical components that bring a definite physiological action on the human body to prevent the diseases. In this work, we investigated the antimicrobial activity of leaves’ extracts of Quercus robur L., collected from the Algerian upper highlands, on ten bacterial strains and one fungal strain known to be pathogenic. First, we performed a qualitative phytochemical analysis, and second, antimicrobial activity tests performed by agar diffusion method (disc and well) with the determination of MIC by broth macro-dilution method. Given the results, it appears that obtained macerates of Quercus robur L. were rich in bioactive phytoconstituents such as alkaloids, anthraquinones, saponins, tannins, and other components. The yield of aqueous and methanolic macerates of leaves was 8.5 ± 1.41 and 22.4 ± 4.36%, respectively. The bacterial resistance was relatively important to several antibiotics, namely, ampicillin, amoxicillin + clavulanic acid for strains of Escherichia coli and Salmonella sp. However, Staphylococcus aureus strains were resistant to fusidic acid, penicillin, and oxacillin; while Enterococcus faecalis was resistant to fusidic acid, penicillin, oxacillin, and ticarcillin. The antibacterial activity of the macerates toward tested microbial strains showed that the aqueous and methanolic macerates of the leaves were proportional to the tested concentration and active not only against Gram-positive and Gram-negative bacteria but also on the fungal species Candida albicans. The estimated MIC for Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus was in the order of 10 mg/mL, which seems more effective than toward Salmonella sp., Klebsiella pneumoniae, Pseudomonas aeruginosa, and Candida albicans which were in the order of 30 mg/mL. These preliminary results confirm that the part of the studied plant had a very good antimicrobial activity that was proportional to the serial concentrations of the tested extracts.

Microorganisms, usually from the dental caries, are the main sources of diseases in dental pulp (root canals) and periapical region. Facultative bacteria and fungi have been identified in therapy resistant persistent endodontic infection. The objectives of this study was to evaluate the antimicrobial efficacy of Mineral Tri Oxide Aggregate (MTA) against therapy resistant endodontic microorganisms. The efficacy of MTA was also compared with that of calcium hydroxide. Six standard bacterial stains were used: Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, bacillus subtilis, Candida albicans and Enterococcus faecalis. The agar diffusion method on Muller- Hilton media was employed. The plates containing media were inoculated with the specified bacterial suspensions. Two standard holes were prepared on each microorganism inoculated plate with a copper puncher and one hole was completely filled with MTA & the other with Ca (OH)2 . The plates were then kept at environmental temperature for one hour to ensure prediffusion and then incubated at 370C for 24 hours. After 24 hours, the diameters of inhibition zones were measured. Tests were replicated for thirty times for each sample and mean values were taken. Zone of inhibition as measured for MTA and Ca (OH)2 were statistically analyzed with Student’s t-Test and Post Hoc Games Howell Test and were presented as mean ± SD to compare of efficacy of MTA and calcium hydroxide on different microorganisms. Both MTA and Ca(OH)2 were found to produce zone of inhibition against Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa (ATCC 27853), , Bacillus subtilis (BTCC 17 ), and Candida albicans (BTCC 493). MTA showed highest activity against S. aureus and lowest activity against P. aeruginosa which was similar to the activity range of Ca (OH)2 against the mentioned organisms. But both of them failed to produce any activity against E. coli and. E. faecalis. MTA was found to produce a lower efficacy than Ca (OH)2 while comparing the zone of inhibition between them and statistically it was significant. Mineral Tri Oxide Aggregate (MTA) showed antimicrobial efficacy against some therapy resistant microorganisms but it did not show antimicrobial efficacy against Escherichia coli and Enterococcus faecalis. Medicinal plants have served as sources of medicine to treat and suppress the diseases, because many pathogens are gaining resistance to the current synthetic drugs. In addition, high cost and adverse side effects are commonly associated with popular Synthetic drugs. Therefore, there is need for continuous search for new drugs in order to overcome this emerging resistance. Plants synthesize bioactive compounds which are of great potential in agriculture, antimicrobial and anti-insect activity. The concentration of bioactive compounds in each plant species depends on the environmental conditions, age of the plant, relative humidity of harvested materials and method of extraction. Little is known on the phytochemical and antimicrobial potential of Alba and Rosea cultivars of Catharanthus roseus ethanol extracts. The leaves of Alba and Rosea cultivars were investigated for their phytochemical and antimicrobial properties. The study was conducted at Maseno University, Kenya. Plant Leaves were collected around Maseno University. Leaves of Alba and Rosea cultivars of Catharanthus roseus were air-dried in the shade, thereafter crushed into powder and ethanol extraction done using the Rotary evaporator. Antimicrobial activity of the pathogenic microorganisms was Candida albicans and Escherichia coli.  Disc diffusion method was used for antimicrobial tests. concentrations of ethanol leaf extracts consisted of 2.5, 5and 7.5 mg/mL with three replications.  Data on growth inhibition were subjected to analysis of variance (ANOVA) . Phytochemical analysis revealed the presence of tannins, flavonoids, terpenoids, saponins, alkaloids and phenols except steroids and glycosides. The ethanol leaf extracts were active against Candida albicans and and Escherichia coli. Alba leaves extracts showed higher inhibitory zones compared to Rosea leaves. The observed differences in antimicrobial activity could be due to differences in cell wall synthesis, structure and composition. The results of present study further confirm the use of these plants traditionally for the treatment of different ailments.

ABSTRACT Sponges are a component of coral reef biota. These sea animals are known to contain compounds that have the potential to be developed in the field of medicine, including as an antimicrobial. This study aims to determine the antimicrobial activity of Phyllospongia lamellosa sponge against the growth of Staphylococcus aureus, Escherichia coli and Candida albicans collected in the waters of Tumbak, Posumaen District, Southeast Minahasa. The antimicrobial activity test was carried out by agar diffusion method. The results showed that the extract and fraction of Phyllospongia lamellose had antimicrobial activity seen in the inhibition zone formed around the paper disk against the test microbes. Ethanol extrack and fraction from Phyllospongia lamellosa sponge showed the greatest antimicrobial activity against Candida albicans with an average value of 13,33 mm was categorized as strong , than in Staphylococcus aureus with an average value of 13 mm is categorized as strong and on Escherichia coli 11 mm categorized as strong. Keywords  :Phyllospongia lamellosa, antimicrobial activity, Escherichia coli, Staphylococcus aureus, Candida albicans ABSTRAK Spons merupakan salah satu komponen biota penyusun terumbu karang.  Hewan laut ini diketahui mengandung senyawa- senyawa yang berpotensi untuk dikembangkan dalam bidang pengobatan, diantaranya sebagai antimikroba. Uji aktifitas antimikroba dilakukan dengan metode difusi agar. Hasil penelitian menunjukkan bahwa ekstrak dan fraksi spons Phyllospongia lamellose memiliki aktifitas antimikroba dilihat zona hambat yang terbentuk disekitar cakram kertas terhadap mikroba uji. Ekstrak etanol dan fraksi dari Spons Phyllospongia lamellosa menunjukkan aktivitas antimikroba paling kuat terhadap candida albicans dengan nilai rata-rata 13,33 mm dikategorikan kuat, kemudian pada Staphylococcus aureus dengan nilai rata-rata 13 mm dikategorikan kuat, dan pada Escherichia coli 11 mm dikategorikan kuat. Kata Kunci :  Phyllospongia lamellosa, aktivitas antimikroba, Escherichia coli, Staphylococcus aureus, Candida albicans

Antimicrobial activity of alcoholic extractions from the aboveground organs like bark and buds of Quercus robur L. was studied. The determination of the minimum inhibitory concentration (MIC) was performed with the use of double serial dilution in broth. The following microorganisms were used as test cultures: Bacillus cereus, Candida albicans, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus. It has been revealed that alcoholic extractions of the bark of Quercus robur have the widest spectrum of antibacterial activity in relation to Pseudomonas aeruginosa, Escherichia coli and Candida albicans. Results of microbiological analysis show that alcoholic extractions from the bark and buds have an antimicrobial effect on all of these strains with a single dilution (except for antimicrobial activity against Staphylococcus aureus with 70% extractions of bark of Quercus robur). In particular, relatively high antimicrobial activity was observed in alcoholic extracts of all buds concentrations against Candida albicans at a dilution of 2, 4 and 8 times and Escherichia coli at a dilution of 4, 8 and 16 times. The greatest antimicrobial effect against Pseudomonas aeruginosa has extracts from the bark and buds in all the studied concentrations of alcohol. A significant antimicrobial effect against Escherichia coli has 40% of extractions from the buds of Quercus robur when diluted by 2, 4, 8, 16, 32 and 64 times. The results can be used as justification for the introduction of a new type of raw material that is buds of Quercus robur, as well as drugs based on it in the State Pharmacopoeia of the Russian Federation as antibacterial phytopharmaceuticals.

The genus Lepidium L. from Brassicaceae Burnett. family covers over 150 species with an almost cosmopolitan spread. In Kazakhstan, 21 species are described, of which four species are characterized by medicinal properties (L. crassifolium Waldst. et Kit., L. perfoliatum L., L. ruderale L., and L. latifolium L.), used in folk medicine as means of antibacterial, irritant, laxative, antitumor, analgesic, and anthelmintic action. Methods. Raw materials were collected from Almaty region (Republic of Kazakhstan). Lepidium latifolium L. herb’s carbon dioxide extract (CO2 extract) was obtained by subcritical carbon dioxide extraction. A gas chromatograph with a mass spectrometric detector was used to determine the component composition of the extract. Antimicrobial activity was determined by two methods: the micromethod of serial dilution and the disc-diffusion method. Four microbial test strains were used: Staphylococcus aureus ATCC 6538-P, Escherichia coli ATCC 8739, Klebsiella pneumonia ATCC 10031, and Candida albicans ATCC 10231. Results. The technology of carbon dioxide extraction has undoubted advantages over traditional methods of extraction: it has a controlled selectivity in relation to groups of biologically active substances, allows deep extraction, and maximizes the release of rich complexes of compounds contained in plants. In this study, firstly, the CO2 extract was obtained under subcritical conditions from the aerial part of L. latifolium L., and the composition was determined. Hexane was the best solvent for CO2 extract, and 40 components were identified. Screening of antimicrobial activity of the L. latifolium’s CO2 extract showed the essential activity of all clinically significant strains tested: Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia, and Candida albicans. Conclusions. This research showed that the CO2 extract of the raw material of Lepidium latifolium L. contains biologically active compounds exhibiting an essential antimicrobial effect, and therefore it is possible to recommend for the development of various drugs for use in medical practice.

ABSTRACT Algae have long been used for the treatment of various types of diseases. Ulva lactuca is one of the algae, which contains bioactive substances as antimicrobial, antifungal, and antioxidant. This study aims to determine the microorganisms growth inhibitory activity from Ulva lactuca algae obtained from the of Lembeh Strait waters in City of Bitung against microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. Ulva lactuca algae was extracted using maceration method with ethanol solvent and fractionated with methanol, n-hexan and chloroform solvents. Testing of antimicrobial activity using agar diffusion method. The result showed that extracts and fractions of Ulva lactuca algae did not have antimicrobial activity against the microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. Hasil penelitian menunjukkan ekstrak dan fraksi dari alga Ulva lactuca tidak memiliki aktivitas antimikroba terhadap Escherichia coli, Staphylococcus aureus, dan Candida albicans. ABSTRACTSoft coral Dendronephtya sp., living in the waters of coral reefs that are rather deep, at depths below 10 meters, grow attached to a hard substrate, protected under lumps of living coral or dead coral. This study aims to determine the antimicrobial activity of soft coral Dendronephtya sp., collected from the waters of Tumbak Village, Pusomaen District, Southeast Minahasa Regency, against Escherichia coli, Staphylococcus aureus and Candida albicans. Dendronephtya sp., extracted by maceration method using 96% ethanol solvent, fractionation using liquid-liquid partition method with n-hexane, chloroform and methanol solvent, and the testing of antimicrobial activity using the Kirby Bauer diffusion method. The results showed that ethanol extract of soft corals (Dendronephtya sp.), n-hexane fraction had inhibition of 8 mm against Escherichia coli bacteria, 7 mm against Staphylococcus aureus while Candida albicans had very good activity in n-hexane fractions of n-hexane. 9.3 mm, so it can be recommended as an antimicrobial. Keywords : Antimicrobial, Candida albicans, Escherichia coli, Soft Coral (Dendronephtya sp.), Staphylococcus aureus. ABSTRAKKarang lunak Dendronephtya sp., hidup di perairan terumbu karang yang agak dalam, pada kedalaman di bawah 10 meter, tumbuh melekat di dasar yang keras, terlindung di bawah bongkahan karang hidup atau karang mati. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antimikroba dari karang lunak Dendronephtya sp., yang dikoleksi dari perairan Desa Tumbak Kecamatan Pusomaen Kabupaten Minahasa Tenggara terhadap Escherichia coli, Staphylococcus aureus dan Candida albicans. Dendronephtya sp., diekstraksi dengan metode maserasi menggunakan pelarut etanol 96%, fraksinasi menggunakan metode partisi cair-cair dengan pelarut n-heksan, kloroform dan metanol, dan pengujian aktivitas antimikroba menggunakan metode difusi agar Kirby Bauer. Hasil penelitian menunjukkan bahwa ekstrak etanol karang lunak (Dendronephtya sp.) Fraksi karang n-heksan memiliki daya hambat sebesar 8 mm  terhadap bakteri Escherichia coli, 7 mm pada Staphylococcus aureus  sedangkan  Candida albicans aktivitas  yang sangat baik terjadi pada fraksi dan ekstrak n-heksan sebesar 9,3 mm, sehingga dapat direkomendasikan sebagai antimikroba. Kata Kunci : Antimikroba, Candida albicans, Escherichia coli,  Karang Lunak (Dendronephtya sp.), dan Staphylococcus aureus

ABSTRACTTunicate is an invertebrate that lives in a coral reef ecosystem and produces many compounds such as, antibacterial, antitumor and anticancer. This study aimed to determine the antimicrobial activity of extracts and fraction of tunicate (Polycarpa aurata) collected in the Lembeh Strait, Bitung against Escherichia coli, Staphylococcus aureus and Candida albicans. Tunicate (Polycarpa aurata) was extracted by maceration method using 96% ethanol solvent, fractination using liquid-liquid partition method with n-hexane, chloroform and methanol solvent, and antimicrobial testing using Kirby Bauer’s disk diffusion method. The results showed that ethanol extract of tunicate (Polycarpa aurata) had antimicrobial activity againts Escherichia coli with an inhibition of 15.12 mm, and againts Candida albicans with an inhibition of 15 mm. Hasil penelitian menunjukkan bahwa ekstrak etanol tunikata (Polycarpa aurata) memiliki aktivitas antimikroba kategori kuat pada fraksi metanol dengan daya hambat sebesar 16, 17 mm terhadap Escherichia coli, pada ekstrak etanol dengan daya hambat sebesar 15, 12 mm terhadap Staphylococcus aureus sedangkan pada Candida albicans aktivitas yang sangat baik terjadi pada ektraksi etanol sebesar 15 mm. Kata Kunci: Tunikata (Polycarpa aurata), Ekstraksi dan Fraksinasi, Antimikroba

ABSTRACT Sponges are marine animals, which contain active compounds known to have potential in the pharmaceutical field, including the antimicrobials. The purpose of this study was to determine the activity of microorganism growth inhibitors from the sponge Liosina paradoxa against microbes Staphylococcus aureus, Escherichia coli, and Candida albicans. Samples were extracted by maceration method with ethanol solvent and fractionation using methanol, chloroform, and n-hexane solvents. Antimicrobial activity was carried out by agar diffusion method (Kirby and Bauer). Methanol fractions of sponge Liosina paradoxa showed the greatest antimicrobial activity against Escherichia coli with an average value of 11.33 mm, and categorize as strong, chloroform fraction was 7.33 mm and categorize as medium, and n-hexane fraction was 7.16 mm and categorize as medium, ethanol extracts was 7.83 mm and categorize as moderate antimicrobial activity. Keywords: Liosina paradoxa, Antimicrobial, Staphylococcus aureus, Escherichia coli, Candida      albicans. ABSTRAKSpons merupakan merupakan hewan laut yang mengandung senyawa aktif yang diketahui berpotensi dalam bidang farmasi, diantaranya sebagai antimikroba. Tujuan dari penelitian ini yaitu untuk mengetahui aktivitas penghambat pertumbuhan mikroorganisme dari spons Liosina paradoxa terhadap mikroba Staphylococcus aureus, Escherichia coli, dan Candida albicans.Sampel diekstraksi dengan metode maserasi dengan pelarut etanol dan fraksinasi menggunakan pelarut metanol, kloroform, dan n-heksan. Aktivitas antimikroba dilakukan dengan  metode difusi agar (Kirby and Bauer). Fraksi metanol spons Liosina paradoxa menunjukkan aktivitas antimikroba paling besar terhadap Escherichia coli dengan nilai rata-rata 11,33 mm, dengan kategori kuat, fraksi kloroform 7,33 mm kategori sedang, fraksi n-hexan 7,16 mm kategori sedang, ekstrak etanol 7,83 mm kategori aktivitas antimikroba sedang. Kata kunci: Liosina paradoxa, antimikroba, Staphylococcus aureus ,Escherichia coli, Candida       albicans.

ABSTRACTSponges are multicellular metazoa animals belonging to the Porifera phylum, which has a different structure from other metazoans. The purpose of this study was to determine whether ethanol extracts from the marine organism sponge Ianthella basta have antimicrobial activity against several pathogenic microbes Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans. The extraction method used is maceration with 96% ethanol solvent. The method used is the Diffusion Method (Disc Diffusion Kirby and Bauer). The antimicrobial activity test uses a 6 mm paper disc with 50 µL absorption per disc. The results of crude ethanol extract of Ianthella basta sponge from all test microbes, namely Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans, were seen to provide the greatest inhibitory activity against Staphylococcus aureus bacteria with an average of inhibitory zone of 7.00 mm categorize as intermediate The results obtained showed that the crude extract of the sponge Ianthella basta has antimicrobial activity because it can inhibit the growth of Staphylococcus aureus, Escherichia coli and Candida albicans microbes even though the inhibition zone is categorized as intermediate. Keywords: Ianthella basta, antimicrobial, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans. ABSTRAKSpons adalah hewan metazoa multiseluler tergolong ke dalam filum Porifera, yang memiliki perbedaan struktur dengan metazoan lainnya. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol 96%. Metode yang digunakan yaitu Metode Difusi (Disc Diffusion Kirby and Bauer).  Pengujian aktivitas antimikroba ini menggunakan kertas cakram (paper disc) berukuran 6 mm dengan daya serap 50 µL tiap cakram. Hasil ekstrak kasar etanol Spons Ianthella basta dari semua mikroba uji yaitu Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa dan Candida albicans, terlihat yang memberikan daya hambat paling besar terdapat pada bakteri Staphylococcus aureus dengan jumlah rata – rata zona hambat yaitu 7,00 mm dengan kategori sedang. Hasil penelitian yang diperoleh menunjukkan bahwa ekstrak kasar dari Spons Ianthella basta memiliki aktivitas antimikroba karena mampu menghambat pertumbuhan mikroba uji Staphylococcus aureus, Escherichia coli dan Candida albicans walaupun dengan zona hambat yang  dikategorikan sedang. Kata Kunci : Ianthella basta, antimikroba, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans.
