Abstract Background Cholestasis is the major cause of bile acid accumulation leading to liver damage. Chronic infection of worms can modulate the immune response towards T helper (Th)2-related cytokines. The present study aims to evaluate the protective impact of an ascarid nematode Toxocara vitulorum extract (TvE) against alpha-naphthylisothiocyanate (ANIT)-induced cholangitis male wistar rat model compared to ursodeoxycholic acid (UDCA) as a standard drug. Results Pretreatment with TvE and/or UDCA induced a marked reduction in the levels of liver function tests and malondialdehyde, while antioxidant markers were increased compared to cholestatic rats. Pretreatment with either TvE or combination before cholangitis induction attenuated the predominant Th1-related cytokines (IFN-γ and TNF-α) to Th2 (IL-13 and IL-10). TvE administration promoted higher expression levels of Bcl-2 protein and lower levels of caspase-3 compared to cholestatic rats. Conclusions Treatment with TvE has improved the liver functions and elevated the levels of oxidative stress markers. The upregulation of Th2-related cytokines and suppression of apoptosis through caspase-3 might be considered as a potential mechanism of TvE. Thereby, this natural extract revealed an opportunity for use in treatment of cholangitis disease.

Background and Objective: Lithospermum officinale is a famous medicinal herb in the traditional medicine of India. However, the medicinal use of its root extract is limited due to the presence of pyrrolizidine alkaloids (PzAl). It was recently shown that PzAl are not accumulated in the cell culture of L. officinale while the biosynthetic pathway of phenolic acids remains active so that rosmarinic acid (RsA) is the main product in the proliferated callus. Considering the existing literature on the anti-inflammatory effects of caffeic acid (CfA) and its derivatives, this research was devoted to the evaluation of the anti-inflammatory capacity of methanolic extracts of L. officinale callus (LoE) on the rat microglial cells as the immune cells of the Central Nervous System, which play an essential role in the responses to neuroinflammation. Methods: primary microglia were obtained from Wistar rat, then they were subjected to various amounts of CfA and methanolic extracts of 17 and 31-day L. officinale callus prior to stimulation by LPS. In addition to HPLC analysis of the extracts, viability, nitric oxide production, evaluation of the pro-inflammatory genes and cytokines in the inflamed microglia were investigated. Results: Methanolic extract of the 17-day old callus of L. officinale exhibited anti-inflammatory effects on the LPS- stimulated microglial cells much higher than that was observed for CfA. The data was further supported by the decreased expression of NOS2, TNF-α, and Cox-2 mRNA and the suppression of TNF-α and IL-1β release in the activated microglial cells pretreated with the effective dose of LoE (0.8 mg mL-1). Conclusion: It was assumed that better anti-neuroinflammatory performance of LoE than CfA in LPS-activated primary microglia could be a result of synergism of the components of the extract and the lipophilic nature of RsA as the main phenolic acid of LoE. Considering the fact that LoE shows high antioxidant capacity and lacks PzAl, it is anticipated that LoE is considered as a reliable substitute to the extract of the natural root of L. officinale and plays a key role in the preparation of neuroprotective pharmaceutical formula.

Changes in consumption behavior to instant food cause various health problems, such as obesity, dislipidemia, and atherosclerosis. A study was conducted to investigate Moringa oleifera extract as an anti-inflammation product that decreases the levels of biochemical markers IL-1 and TNF-a. This experiment was done with randomized pre- and posttest control-group design, employing 40 Wistar rats separated into five groups: control group 0% M. oleifera leaf extract (P0), treatment group 1 with 10% M. oleifera leaf extract (P1), treatment group 2 with 15% M. oleifera leaf extract (P2), treatment group 3 with 20% M. oleifera leaf extract (P3), and treatment group 4 with 25% M. oleifera leaf extract (P4). This research observed that intake of 20% M. oleifera leaf extract results in the highest significant decrease of 15.42% of IL-1 level (134.64 ± 1.98 to 113.87 ± 4.30 pg/mL) and decrease of 45.63% of TNF-α level (28.62 ± 1.25 to 15.56 ± 7.20 pg/mL). Therefore, it can be concluded that intake of M. oleifera leaf extract by Wistar rat has anti-inflammatory effects on chronic dyslipidemia through decrease of IL-1 and TNF-α levels and histopathology profile. Further research is required to determine whether the application of M. oleifera leaf extract (daun kelor) in humans will have similar anti-inflammation effects.

Abstract Background: Osteoarthritis (OA) is the main cause of older pain and disability, its dysfunction due to discomfort as well as the quality of life of patients having adverse effects. Medical ozone (O3) has been found to have antioxidative and anti-inflammatory effects in the treatment of osteoarthritis. However, it is unclear whether O3 can induces autophagy through the PPARγ/mTOR autophagy pathway in chondrocytes treated with IL-1β. Methods: Primary chondrocytes were isolated from wistar rat cartilage within 3days. The OA chondrocyte model was induced via treatment with IL-1β to chondrocytes for 24 hours. Then the cells were treated with O3 and GW9662, the inhibitor of PPARγ. Cell viability was assessed by CCK-8. Further, the cells subjected to western blot analysis, qRT-PCR and immunofluorescence assay. The numbers of autophagosomes were observed via transmission electron microscopy. Results: 30μg/ml O3 improved the viability of chondrocytes. O3 significantly increased the level of autophagy proteins and the numbers of autophagosomes in chondrocytes treated with IL-1β via treated with O3. qRT-PCR results showed that O3 decreased the levels of IL-6,TNF-α and MMP-3,MMP-13 in chondrocytes treated with IL-1β. Conclusions: 30μg/ml O3 improved autophagy via activating PPARγ/mTOR signaling and suppressing inflammation in chondrocytes treated with IL-1β.

Background: Although acetaminophen (APAP) is considered safe at therapeutic doses, intake of high amounts of this drug can cause liver failure. In the present experiment, we examined the hepatoprotective effects of resveratrol (RES) in HepG2 cells and rat liver. Objectives: This study aimed to evaluate the influence of RES on liver function in rat model of necrosis and HepG2 cells. Materials and Methods: In this study, rats were randomly assigned into 4 groups (7 rats in each group) as follows; group 1: control rats (received normal saline), group 2: hepatotoxic control (control rats that received 640 mg/kg/d APAP), group 3: positive control (received 150 mg/kg N-acetylcysteine), group 4: RES (received 30 mg/kg RES). The animals were treated for 7 days. Afterwards, the levels of liver enzymes, protein carbonyl content, glutathione (GSH) level, and Tumor necrosis factor (TNF-α) level were determined. Results: In the in vitro experiment, APAP-induced HepG2 cells were treated with RES at different concentrations and various factors such as cell viability, liver enzymes, GSH and TNF-α levels were measured. Conclusions: Our results indicated that RES could normalize all these factors in vitro and in vivo (P<0.05). In fact, RES had potential hepatoprotective effect against APAP -induced hepatotoxicity in HepG2 cells and animal models mainly via dual change of oxidative stress and cytokine levels.

Abstract Objective: To investigate the effect and mechanisms of Andira anthelmia lectin in rat models of acute inflammation.Material: AAL anti-inflammatory activity was evaluated in Wistar rat models of paw edema, peritonitis and hyperalgesia.Methods: AAL (0.01-1 mg/kg i.v.) was injected 30 min before stimulation with carrageenan and with initial and late phase inflammatory mediators into the animals paw and peritoneum for evaluation of: cell migration (optical and intravital microscopy), paw edema (plethysmometry and histopathology); hyperalgesia (analgesimetry).Results: AAL inhibited leukocyte migration induced by carrageenan, mainly neutrophils to the peritoneal fluid, decreasing leukocyte adhesion. In the peritoneal fluid, AAL reduced the gene expression of TNF-α and cyclooxygenase, as well the levels of PGE2. ALL also inhibited the paw edema induced by carrageenan, mainly the late phase, but also the edema induced by serotonin, histamine, TNF-α, PLA2 and PGE2, but not by L-arginine and bradykinin. In this model, AAL inhibited mechanical hypernociception induced by TNF-α, PGE2, db-cAMP and capsaicin, and the activity of myeloperoxidase in the paw tissues.Conclusion: AAL presents anti-inflammatory effect in acute models of rat inflammation involving the participation of prostaglandins, TNF-α and lectin domain.

AbstractObjectiveThis study aims to observe the curative effect of Huaiqihuang Granules adjuvant therapy on primary nephrotic syndrome (PNS).MethodsA total of 112 children with PNS were randomly divided into three groups, and changes in serum inflammatory cytokines, interleukin, lymphocyte subsets and immunoglobulin were observed.ResultsBefore treatment, IL-18, TNF-α, CD8+ increased, while IL-10, CD4+, NK cells, IgA, IgG and Foxp3+Treg cells decreased. After Huaiqihuang Granules treatment, IL-18, TNF-α, CD8+ decreased, while IL-10, CD4+, NK cells, IgA, IgG and Foxp3+Treg cells increased.ConclusionFunctions of cell immunity and humoral immunity in PNS patients before treatment were suppressed and disordered. Huaiqihuang granules can play a role in immunoregulation, with slight side reactions.

                                                                                                  Objective: This study was designed to explore the benefit of curcumin as adjuvant therapy to the standard H pylori eradication triple therapy in both duodenal and gastric ulcers patients.                       Methods: The present study enrolled 40 patients newly diagnosed endoscopically with peptic ulcer disease to be allocated into group1 treated with standard H. pylori eradication triple therapy, and group2 patients treated with curcumin (500mg) capsules three times daily for 14 day as adjuvant to standard triple therapy. Stool antigen test, immunoglobulin M serology test, tumor necrosis factor-alpha (TNF-α), interleukin 1 beta (IL1β), and total antioxidant capacity (T-AOC) are measured at the baseline and after 6 weeks of treatment.Results: The result showed that the  use of curcumin as adjuvant therapy produced highly significant improvement in healing efficacy which was significantly distinguished  in duodenal ulcer patients compared to gastric ulcer groups 2 patients (p <0.05), along with highly significant reduction in pro inflammatory IL1β level in group 2 patients(p<0.01). After 6 weeks of treatment there was highly significant elevation in level of TNF- α in groups 1 (p value <0.01), though, group 2 patients presented with non significant elevation in TNF-α level. Moreover, the total antioxidant capacity was improved with curcumin adjuvant therapy, though non- significant, compared to group 1patients who showed reduction in total antioxidant capacity.Conclusion: This study revealed that addition of curcumin as adjuvant therapy produced improvement in ulcer healing efficacy, and controlled the inflammatory and oxidative stress process induced by H pylori infection.Keywords: Peptic ulcer disease, Curcumin, H. pylori infection, inflammation and oxidation markers.

The present study investigated the effect of tumor necrosis factor (TNF)-α on myocardial energy metabolism as estimated by myocardial oxygen consumption (MV˙o 2). MV˙o 2 of electrically stimulated isolated trabeculae of right ventricular Wistar rat myocardium was analyzed using a Clark-type oxygen probe. After the initial data collection in the absence of TNF-α, measurements were repeated after TNF-α stimulation. In separate experiments, pretreatment with the nitric oxide (NO) synthase inhibitor N G-nitro-l-arginine methyl ester (l-NAME) or the ceramidase inhibitor n-oleoylethanolamine (NOE) was done to investigate NO/sphingosine-related effects. TNF-α impaired myocardial economy at increasing stimulation frequencies without altering baseline MV˙o 2. Incubation with TNF-α in the presence of l-NAME further impaired myocardial economy. NOE preincubation abrogated the TNF-α effect on myocardial economy. Moreover, the negative inotropic effect of TNF-α was observed in NOE-pretreated but not l-NAME-pretreated muscle fibers. Exogenous sphingosine mimicked the TNF-α effect on mechanics and energetics. We conclude that TNF-α impairs the economy of chemomechanical energy transduction primarily through a sphingosine-mediated pathway.
