BACKGROUND: Trichuris trichiura is a soil-transmitted parasitic worm commonly found in humid, tropical to subtropical areas, as well as poor sanitation area. These worms are cosmopolitan, especially in tropical and humid countries like Indonesia. This worm infection is more common in developing countries and more in children than adults due to poorer child self-hygiene. Worm disease is one of the common society diseases with 60% of children in Indonesia which are estimated to be affected. One species of T. trichiura worm that is often used in research on experimental animals is Trichuris muris. The administration of T. muris infective eggs can significantly increase the number of goblet cells in intestinal crypts of colon and cecum. Goblet cell hyperplasia will occur after exposure to high-dose T. muris (acute/200 infective eggs) in resistant mice. Various studies on the development of natural and traditional materials have been developing in the past few years to see the effects of betel nut on the number of goblet cells in the large intestine. AIM: The aim of this research is to determine the effect of ethanol extract and the ethyl acetate fraction of betel nut on the change in the number of goblet cells in male mice given orally T. muris infective eggs. METHODS: This study was an experimental study with a post-test only control group design in male mice (Mus musculus) which were given 200 infective eggs of T. muris. The study sample consisted of 70 mice divided into seven groups. Doses of the ethanol extract of betel nut are 100 mg/kg b.w. and 150 mg/kg b.w., and doses of ethyl acetate fraction are 100 mg/kg b.w. and 150 mg/kg b.w. The independent variable was ethanol extract and ethyl acetate fraction of betel nut. The dependent variable is the number of goblet cells. Mean differences in groups were tested by Mann–Whitney. RESULTS: Statistical analysis showed a significant difference of p < 0.05 in the colonic crypts and cecum. In Group K (+), there was no increase in the number of goblet cells (54.2000 ± 30.54864) compared to Group K (−) (79.0750 ± 11.79221) in the colon. There was an increase in goblet cells in the 150 mg/kg b.w. of ethanol extract group (64.1000 ± 29.50381), the 150 mg/kg b.w. of ethyl acetate fraction group (56.2250 ± 22.11035), and the 1 mg/20 g of albendazole group (68.3750 ± 11.30956) in the colon. Likewise, there was an increase in the number of goblet cells in the cecum. It can be concluded that 150 mg/kg b.w. of the ethanol extract of betel nut can effectively increase the number of goblet cells in the colon and cecum. CONCLUSION: Ethanol extract of betel nut 150 mg / kg of weight can effectively increase the number of goblet cells in the colon and caecum.

Penicillium expansum Link causes an economically important postharvest blue mold disease in a number of fruits and vegetables. In the present study, this fungus was isolated from rotted grapes and identified on morphological basis. Identification of the pathogen was further confirmed on molecular basis by using four different primer pairs namely ITS, β-tubulin, CMD and CF under accession numbers MN752155, MN787831, MN787832 and MN787833, respectively. Leaf extract of Chenopodium murale was assessed for its potential to control in vitro growth of P. expansum. For this purpose, leaves were extracted in methanol and after evaporation of the solvent, the resulting extract was successively partitioned with n-hexane, chloroform, ethyl acetate and n-butanol followed by antifungal bioassays with different concentrations (1.562 to 200 mg mL-1) each organic solvent fraction. Although all the fractions variably controlled the fungal pathogen, however, n-butanol fraction showed the highest antifungal activity causing 45–86% reduction in biomass of the pathogen. Ethyl acetate fraction was also highly antifungal and reduced fungal biomass by 44–81%. Chloroform and n-hexane fractions were comparatively less effective and reduced biomass of P. expansum by 30–72% and 11–44%, respectively. This study concludes that ethyl acetate and n-butanol fractions are highly antifungal in nature against P. expansum.

This study was aimed at identifying secondary metabolites of ethyl acetate fraction of Dendrophtoe falcata (L.f.) Ettingsh mindi plant parasite (Melia azedarach L.). This research was conducted by maceration method using ethanol solvent, partitioning sequentially with n-hexane, chloroform, and ethyl acetate. Ethyl acetate fraction was separated by gravity column chromatography (GCC) in two stages. Phase I GCC used n-hexane : ethyl acetate (9 : 1). Phase II GCC used chloroform : methanol (9 : 1) eluent to obtain one pure compound. Purity identification used thin layer chromatography. Characterization of pure compounds obtained was carried out using UV-Vis and IR. The results show that the isolated compounds maximum wavelengths are at 351.20, 262.60, and 207.20 nm which corresponded to the conjugated synamoyl, benzoyl and chromophore phenol. IR spectrum data shows the presence of O-H, C-H aliphatic, C = O carbonyl, C = C aromatic, and C-O. From these data, the isolated compounds show flavonoid type flavanols.IDENTIFIKASI SENYAWA METABOLIT SEKUNDER DARI FRAKSI ETIL ASETAT BATANG Dendropthoe falcata.Penelitian ini bertujuan untuk mengidentifikasi senyawa metabolit sekunder dari fraksi etil asetat batang Dendrophtoe falcata (L.f.) Ettingsh parasit tumbuhan mindi (Melia azedarach L.). Penelitian ini dilakukan dengan metode maserasi menggunakan pelarut etanol, partisi secara berurutan dengan n-heksana, kloroform, dan etil asetat. Fraksi etil asetat dipisahkan secara kromatografi kolom gravitasi (KKG) dalam dua tahap. KKG tahap I menggunakan eluen n-heksana : etil asetat (9 : 1). KKG tahap II menggunakan eluen kloroform : metanol (9 : 1) sehingga diperoleh satu senyawa murni. Identifikasi kemurnian menggunakan kromatografi lapis tipis. Karakterisasi senyawa murni yang diperoleh dilakukan menggunakan UV-Vis dan IR. Berdasarkan hasil analisis dengan spektrofotometer UV-Vis, senyawa hasil isolasi menunjukkan panjang gelombang maksimum pada 351,20; 262,60; dan 207,20 nm yang sesuai dengan gugus sinamoil, benzoil, dan kromofor fenol terkonjugasi. Data spektrum IR menunjukkan adanya ikatan O-H, C-H alifatik, C=O karbonil, C=C aromatik, dan C-O. Dari data tersebut, senyawa hasil isolasi menunjukkan golongan flavonoid jenis flavonol.

Traditional Filuferru is an ancient spirit from Sardinia, Italy, which is usually obtained from the distillation of wine or grape marc. In this contribution, the results of the first chemical characterization of a wide number of crafts Filuferru samples has been accomplished in terms of an evaluation of the alcoholic strength, qualitative and quantitative gas-chromatography–mass spectrometry (GC-MS) analysis of the volatile composition of the distillate, and its trace element composition by means an inductively coupled plasma–mass spectrometry (ICP-MS) method. Both instrumental methods have been validated and applied on 21 craft samples of Filuferru, whereas one sample of commercial distillate has been analyzed for comparison purposes. Alcoholic strength ranged between 41.0 and 62.4% (v/v). Sixty volatile compounds were identified and ten of them have been quantified. Analogies and differences with Grappa (i.e., the Italian distilled spirit closer to Filuferru) have been highlighted in the qualitative and quantitative profile of this matrix. Often meaningful amounts of acetaldehyde, ethyl acetate, dietyl acetal, and acetic acid were measured. Elemental analysis, performed on toxic, non-toxic elements, and oligoelements, 18 in total, revealed a wide variability of concentrations in both analytes and samples. High concentrations of Cu are sometimes evidenced, which are likely caused by losses from the distillation apparatus. The principal components analysis (PCA) allowed the differentiation of the ten volatile compounds quantified in two groups: the former, as described mainly by PC1, constituted by acetic acid, ethyl acetate, dietyl acetal, and acetaldehyde, and the second, described by PC2, constituted by 1-propanol, 2-methyl-1-propanol, the two coeluiting isomers 2-methyl-1-butanol and 3-methyl-1-butanol,1-hexanol, 2-phenylethanol, and 2,3-butanediol. Data obtained may be useful in order to establish a regulation for the production of high-quality traditional Filuferru from Sardinia.

Actinidia callosa var. ephippioides (ACE) has been widely used to treat anti-pyretic, antinociceptive, anti-inflammation, abdominal pain and fever in Taiwan. This study aims to determine the mechanism of anti-inflammatory activities of ethyl acetate fraction of ACE (EA-ACE) using a model of λ-carrageenan (Carr)-induced paw edema in mouse model. In HPLC analysis, chemical characterization of EA-ACE was established. In order to investigate the anti-inflammatory mechanism of EA-ACE, we have detected the activities of catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx) and the levels of malondialdehyde (MDA) in the paw edema. Serum NO, tumor necrosis factor α (TNF-α), and interleukin-1β (IL-1β) were evaluated. Chemical characterization from HPLC indicated that EA-ACE contains betulinic acid, ursolic acid and oleanolic acid. In the anti-inflammatory test, EA-ACE decreased the paw edema after Carr administration, increased the activities of CAT, SOD, and GPx and decreased the MDA level in the edema paw at the 5th hr after Carr injection. EA-ACE affects the serum NO, TNF-α, and IL-1β levels at the 5th hr after Carr injection. EA-ACE decreased Carr-induced inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expressions by Western blotting. Actinidia callosa var. ephippioides have the potential to provide a therapeutic approach to inflammation-associated disorders.

Isolation and characterization of terpenoid compounds from ethyl acetate fraction of Macaranga beccariana Merr. have been done. The separation process at ethyl acetate fraction using gravity column chromatography with gradient elution method. The obtained isolates were tested for purity using thin layer chromatography with various eluents namely n-hexane, chloroform, methanol, acetone and ethyl acetate with Rf values 0; 0.75; 0.80; 0.87 and 0.97 respectively. The terpenoids which were successfully isolated were white crystals with a mass of 0.0119 grams. UV analysis produced 1 peak at l 207.03 nm. Analysis of compounds with FT-IR indicated the functional groups O-H, aliphatic C-H, aliphatic C=C, C-O and C=O. Based on the results of the UV and FT-IR spectrum, the isolate compounds were thought to contain terpenoids.
