Abstract Dietary manipulation with respect to crude protein (CP) content has been suggested as part of the overall strategy for the nutritional management of weanling pigs to improve intestinal health. This has focused on the use of low CP diets that are appropriately fortified with crystalline amino acids (AA). Use of low CP diets minimizes the amount of undigested dietary protein entering the large intestine and being subjected to bacterial fermentation. This is important because protein fermentation leads to the production of toxic metabolites and encourages the proliferation of pathogenic bacteria, thus causing enteric problems such as post-weaning diarrhea. There have been considerable efforts to elucidate the mechanisms underlying the potential benefits of feeding low CP diets to piglets. In addition to impacting the intestinal microbiome and its associated activities, it is clear that feeding a low CP diets interferes with the attachment of enterotoxigenic E. coli to the intestinal mucosa, thus minimizing its ability to cause disease. Another area of interest has been how use low CP diets in combination with other dietary manipulations to further enhance intestinal health in piglets. In this regards, existing evidence suggests that a low CP diet may be used in combination with other dietary interventions, such as probiotics and dietary fiber, to further enhance gut health outcomes in piglets. Also, addressing the potential reduction in piglet performance when feeding low CP diets by looking more into diet formulation to avoid deficiencies of essential AA or even some of non-essential AA, is critical for successful use low CP diets. Based on the available information, a reduction of dietary protein by four percentage units coupled with appropriate AA supplementation can be a useful dietary strategy to improve intestinal health.

Abstract Qing Hao Gan Cao (QHGC), a Chinese medicinal formula containing Artemisia annua and Glycyrrhizae Radix et Rhizoma, has been used to treat sunstroke and as an antiviral agent for more than 800 years. It has not previously been subject to a toxicological safety evaluation in acute and subacute (28 days) studies. Therefore, the acute and subacute toxicity of an aqueous extract of QHGC were evaluated in vivo. For the QHGC preparation, the botanical raw materials were crushed into pieces and mixed in the ratio of 10:1 in distilled water for 12 h, then boiling three times for 2 h each time. The three decoctions were mixed and filtered, then spray-dried with hot air at 160°C for 30 min, and stored at room temperature. For the acute toxicity test, 72.0 g/kg of QHGC extract was administered by gavage to male and female mice. Body weight, general observations, and autopsy results were recorded. No mortality or toxicity signs were observed during the studies. For the subacute toxicity test, 4.0, 8.0, or 16.0 g/kg/day of QHGC extract was administered to rats for 28 days. General observations and mortality, body weight, biochemical and hematological parameters, organ weight, and pathological morphology were analyzed. The acute and subacute toxicity studies did not show significant changes in body weight, general observations, hematology and biochemical parameters, organ weight, and liver, spleen, stomach, duodenum, testis, ovary, lung, heart, and kidney histopathological analyses. The consumption of QHGC aqueous extract can be considered safe within the conditions of this study.

The Beta-glucan Effects on Lipid profile, glycemia and inTestinal health (BELT) Study investigated the effect of 3 g/day oat beta-glucans on plasma lipids, fasting glucose and self-perceived intestinal well-being. The Study was an 8-week, double-blind, placebo-controlled, cross-over randomized clinical trial, enrolling a sample of 83 Italian free-living subjects, adherent to Mediterranean diet, with a moderate hypercholesterolemia and a low cardiovascular risk profile. Beta-glucans reduced mean LDL-Cholesterol (LDL-C) levels from baseline by 12.2% (95%CI: −15.4 to −3.8) after 4 weeks of supplementation and by 15.1% (95%CI: −17.8 to −5.9) after 8 weeks of supplementation (p < 0.01 for both comparison and versus placebo). Between baseline and 4 weeks Total Cholesterol (TC) levels showed an average reduction of 6.5% (95%CI: −10.9 to −1.9) in the beta-glucan sequence; while non-HDL-C plasma concentrations decreased by 11.8% (95%CI: −14.6 to −4.5). Moreover, after 8 weeks of beta-glucan supplementation TC was reduced by 8.9% (95%CI: −12.6 to −2.3) and non-HDL-C levels by 12.1% (95%CI: −15.6 to −5.3). Decreses in TC and non HDL-C were significant also versus placebo (respectively p < 0.05 and p < 0.01 to both follow-up visits). Fasting plasma glucose and self-perceived intestinal well-being were not affected by both beta-glucan and placebo supplementation.

Background Mannan oligosaccharides (MOS) are a promising feed additive in animal husbandry due to mainly improving animal health status. The purpose of this study was to investigate the effects of MOS on growth performance, nutrient digestibility, ruminal fermentation, and twelve hematological parameters in sheep. Methods Ninety-six healthy Hu rams with similar body weights were chosen and divided into four treatment groups (twenty-four rams in each group), in which four different doses of MOS were tested: 0%, 0.8%, 1.6% and 2.4% of the basal diet (on an as-fed basis). Results The results showed that supplementation dietary MOS did not affect feed intake, body weight, average daily weight gain, or ruminal short-chain fatty acids (SCFAs) concentration; the ratio of individual fatty acids to total SCFAs, the C2/C3 ratio, and the hematological parameters in the sheep were also unaltered (P > 0.05). Conversely, supplementation dietary MOS increased the dry matter, organic matter, crude protein, neutral detergent fiber, acid detergent fiber, and ash apparent digestibility (P < 0.05), and decreased the ruminal ammonia concentration in the sheep (P < 0.05), especially at a dose of 1.6%. Conclusions This indicates that supplementation dietary MOS improved nutrient utilization by the sheep and nitrogen metabolism in the rumen; however, the effects are too slight to interfere with the basal metabolism in the sheep.
