Abstract Introduction: TRALI is acute lung injury occurring during or within hours of a blood transfusion. The etiology is thought to be infusion of leukocyte antibodies or neutrophil priming and activation caused by biologically active lipids in blood components. We report a TRALI reaction associated with fresh frozen plasma (FFP) and activation of complement in both the unit of FFP and the patient at the time of the reaction. Case History: A 59 year old male with factor XI was admitted to the hospital with hematochezia and given 3 units of FFP. During infusion of the third unit, he developed dyspnea and cyanosis requiring ventilator and O2 support. A chest x-ray showed bilateral diffuse pulmonary infiltrates, CVP was 3 mm Hg, and an echocardiogram was normal. The symptoms resolved in 3 days. Methods: Samples from donors and/or units were screened for the presence of HLA antibodies by ELISA and lymphocytotoxicity and antibodies detected were typed for HLA specificity and antibody class. Reactivity was determined by flow crossmatch. Serologic and molecular HLA typing was completed on donor and patient samples. Priming activity of the implicated FFP, fresh plasma from donor and recipient, and plasma from controls was completed against freshly isolated neutrophils from the three sources. Significant activity was defined as >1.5 times the fMLP stimulated superoxide anion (O2−) production. C3aLE, C4aLE, SC5b-9, and Bb were determined by standard techniques. Results: HLA antibodies were only detected in the third unit of FFP. Samples from this unit and the donor exhibited HLA Class I and II reactivity by ELISA but not lymphocytotoxicity. Flow crossmatch cells demonstrated Class II, IgG reactivity of donor serum against recipient DR11, 13. No autologous reactivity was demonstrated. The FFP unit primed the fMLP response in donor, recipient and control neutrophils 2.6, 3.1, and 3.4 fold above baseline. Testing of donor, recipient and control plasma obtained 3 months after the reaction showed no priming against the same battery of cells (priming ratio 0.8–1.3). C4aLE (105%, control range 24–176%); C3aLE (476%, control range 21–180%); and Bb (351%, control range 31–169%) were elevated in recipient samples obtained during the TRALI reaction and SC5b-9 was at the high end of normal (164%, control 0–200%). These returned to normal after the reaction. Strikingly, evidence of complement activation was seen in the FFP unit (C4aLE 214%, C3aLE 402%, C5b-9 213%) but not in subsequent samples from the donor. Conclusion: These studies document a TRALI reaction with symptoms expressed during the administration of FFP. One unit exhibited HLA Class I and II antibodies, the latter of which bound to the recipient’s cells. Priming activity was seen with plasma from the implicated unit, not in subsequent samples from the donor. Laboratory studies document activation of complement in the FFP infused but not donor samples. Plasma from the recipient at the time of the reaction also exhibit activation of complement which became normal after the TRALI resolved. Infusion of the FFP with activated complement capable of priming neutrophils may have induced pulmonary leukostasis and TRALI quite distinct from any subsequent effect of antibodies. Although the cause of FFP complement activation is not defined, these results suggest alternative mechanisms involving complement may be responsible for HLA antibody-associated TRALI.

Abstract. The aim of this study was to investigate the effects of age on pecking behaviour and plumage quality of free-range laying hens during first access to range. Six hundred 16-week-old layer pullets obtained from a commercial company were housed in a experimental free-range house. The experimental house consisted of a slat-and-litter floor with range area and was divided into three similar pens. The pullets were randomly assigned to a control and two experimental groups according to age at first access to range area. The birds were allowed into the range area for the first time at 18, 20 and 22 weeks of age in the groups of treatment I, treatment II and control, respectively. Pecking behaviour and plumage quality of the birds were measured for 8-week intervals from 24 to 48 weeks of age. Total feather damage significantly increased with age in all groups (P < 0.040, P < 0.006, P < 0.001). Compared to the control group, birds allowed into the range area earlier had less feather damage. The feather score of the tail body region in all groups was significantly greater (more damage) than in the others (P < 0.001). There were no significant differences for the total pecking behaviour of the birds in the groups at 24, 32, 40 and 48 weeks of age. Incidents of gentle feather pecking in every period were more frequent, while aggressive pecking was less frequent (P < 0.026, P < 0.007, P < 0.001). It can be said that access to range area as early as possible at the beginning of the laying period may be useful to reduce the negative effect of feather pecking in free-range layer chickens.

An observational study on range enrichment was conducted on three commercial fixed-range, free-range layer farms comprising four flocks in South Australia (flock sizes ranged from 3000 to 11700 hens). Two strategies were compared; a ‘standard’ Control treatment, typical of the type of range used in the commercial industry and a highly ‘enriched’ treatment. Both treatments were implemented on the same range with no subdivisional fences. The highly enriched side of the range incorporated shade shelters, alongside a continuum of additional enrichment structures (e.g. dust baths, peck objects and hay bales). Range treatments were investigated to determine the effects on motivating hens to use the range more effectively, and their potential to lead to a decrease in abnormal behaviours, such as injurious feather pecking and cannibalism. Additionally, two farms with high levels of tree coverage were also observed. Farms were visited monthly for 6 months. At each visit several measures were recorded live: the numbers of hens out on the range, location of hens on the range and plumage score of birds outdoors and indoors. Enrichment structure utilisation on the range was measured via video recordings and continuous counts of the number of hen visits to enrichment structures within 1 h. It was found that more hens used the ‘enriched’ side of the range, in comparison with the ‘standard’ Control range treatment (P ≤ 0.01). Overhead cover provided by constructed shelters and dust bathing pits were instrumental in enticing birds out onto the range (306 ± 27 s.e.m. and 366 ± 24 s.e.m., average number hen visits). Hay bales were also highly utilised (116 ± 14 s.e.m., average number hen visits), even though the hen numbers attracted to the hay bales were not as high as shelters and dust baths. It was also observed on one property that natural cover provided by trees attracted the greatest number of hens (1652 ± 358 s.e.m., average number of hen visits). Prevailing weather conditions and the age of the flock strongly influenced the number of hens utilising the range outdoors. Furthermore, hens outside on the range had better plumage scores with lower feather loss and decreased areas of bare skin compared with birds scored in the shed.

AbstractTuberculosis like lesions (TBL) in free-range pigs are characterised by presenting a marked heterogeneity in pathology and microbiology features, with a notorious role of Mycobacterium tuberculosis complex (MTC), Trueperella pyogenes and different Streptococcus species. However, the capacity of these microorganism to spread to different organic cavities leading to a generalised disease is unknown. Therefore, this study evaluated the organic distribution of these agents in free-range pig carcasses whole condemned due to generalised TBL.A total of 37 totally condemned animals were analysed, and samples of lymph nodes and organs were obtained (n = 262) and subjected to histopathological and microbiological examination. In addition, T. pyogenes and streptococci species were further characterised by PFGE analysis. Two different patterns were evidenced with lack or occasional lesions in superficial inguinal (SILN) and popliteal (PLN) lymph nodes and advanced lesions in submandibular (SLN) (35/36) and gastrohepatic (GHLN) (33/35) lymph nodes (stages III and IV). Early stage granulomas (stage I and II) prevailed in lungs (16/20), liver (14/31) and spleen (7/18). The microbiological analysis revealed that MTC, detected by qPCR, was present in 31 out of 37 animals and 90 (90/262) samples. In 26 out of the 31 pigs, MTC was detected from two or more organs. SLN (24/31) and GHLN (19/31) were the MTC+ organs most frequently detected, with 29 out of 31 MTC+ pigs detected as positive in one or both samples, which points out that both lymph nodes must be included in the sampling of surveillance programs. Other pathogens, such as T. pyogenes and Streptococcus spp., were also involved in generalised lymphadenitis, being frequently isolated from SLN and other organs, such as liver (T. pyogenes), tonsils or lung (Streptococcus spp.). A wide genetic diversity among streptococci was observed, showing the ubiquitous character of these pathogens, however, the isolation of a single clone of T. pyogenes from different organic locations from animals with generalised TBL was a common finding of this study, highlighting that the role of this pathogen in porcine lymphadenitis may be underestimated. These results should be considered in future studies on the pathogenesis and control of porcine lymphadenitis.

THE SOUTH EAST WALES INFECTIOUS DISEASE LIAISON GROUP

Domestic kitchen food handling risk factors for sporadic salmonella food poisoning are largely unknown. We compared food consumption and food handling practices, opportunities for cross contamination and refrigerator temperature control, in 99 households in South East Wales in 1997/8 with a case of salmonella food poisoning, and control households matched for electoral ward. On univariate analyses, cases were significantly more likely than control respondents to have purchased free-range eggs in the preceding week, and more likely than control households to have handled frozen whole chicken in the previous week, and to handle raw chicken portions at least weekly. In multivariate analysis, only consumption of raw eggs and handling free-range eggs were significant risk factors, independent of the age structure of the family and of the season.

With the purpose to search upon the cytokinins origin, a study was conducted on the effects of prohexadione-ca (P-Ca) on cytokinins in xylem sap and on stomatal resistance in apple. Golden Delicious seedlings were sprayed with 100 ppm P-Ca. Stomatal leaf resistance was measured at intervals between 1 and 4 hours of spraying and 15 days after the treatment. Golden Delicious trees were sprayed with 200 ppm P-Ca two weeks after full bloom. At intervals between 1 and two weeks after P-Ca treatment, the main de-shooted stem was removed from root system and xylem sap was collected for cytokinins analysis using the suction technique. Prohexadioneca significantly increased stomatal resistance (range 2.74-4.35 s/cm−1) when compared to control (range 2.12-3.44 s/cm−1); however, P-Ca did not modify the level of cytokinins in xylem sap since range values were between 0.10 to 0.20 μg kinetin equivalents/20 ml xylem sap for both, cytokinin and control.
