The allelopathy of spent substrate extracts including water extract and ethanol extract ofPleurotus ostreatuson the mycelium growth of six edible fungi, i.e.Flammulina velutipes,Ganoderma lucidum Karst,Pleurotus ostreatusandCordyceps (three species), were investigated using Petri dishes approach. The results indicated that the spent substrate extracts have different effects on their mycelium growth. The mycelium ofFlammulina velutipesgrows better than control check with increasing water extract concentration. The mycelia ofGanoderma lucidum Karstgrow first promotion after inhibition with increasing water extract concentration. The mycelia ofFlammulina velutipes, Ganoderma lucidum Karst, Pleurotus ostreatusare promoted by ethanol extracts. The mycelia of Cordyceps (three species) grow first promotion after inhibition with ethanol extracts. The results can provide reference values for rational utilization of the spent mushroom substrate.

Ganoderma lucidum has a long tradition of use in folk medicine of the Far East, which is documented in the oldest Chinese pharmacopoeia, written in the first century B.C, declaring it a superior medicine. The healing properties of G. lucidum reflected on folk names such as: Reishi, Mannentake, Ling Zhi etc., which mean ?herb of spiritual power?, ?mushroom of immortality? or ?10,000-year mushroom?, respectively. It has been known, for thousands of years, that this species extends life span, increases youthful vigour and vitality and it was used in the treatments of hepatitis, kidneys? disease, hypertension, arthritis, asthma, bronchitis, arteriosclerosis, ulcers and various types of cancer. However, Western civilisation did not discover its healing properties until the 20th century. Modern scientific researches and numerous clinical trails, conducted in recent decades, have confirmed the ancient knowledge of Eastern nations and given them a scientific basis. These studies have demonstrated many biological activities of G. lucidum extracts and compounds, including: immunomodulating, antioxidative, cytotoxic, hypoglycaemic, anti-inflammatory, antiallergic, antimicrobial, etc. It has been reported that its extracts play important role in detoxification of the body and protecton of the liver, as well as in reducing cardiovascular problems, stress and anxiety. However, its most important effect is undoubtedly immunostimulating one as it is the basis of many other positive effects. The Japanese government introduced G. lucidum on the official list of auxiliary agents for the treatments of various cancers, Alzheimer?s disease, diabetes and chronic bronchitis. Many chemical components have been isolated from G. lucidum, but polysaccharides and terpenoids are the main carriers of its bioactivities.

Ganoderma lucidum has been extensively studied for its valuable medicinal importance. In this study, the artificial cultivation of G. lucidum strain Philippine in different culture media, including sawdust substrate, was performed and optimized on the Potato Dextrose Agar (PDA) media. Phytochemical, antibacterial, and antioxidant analyses were performed and compared between the ethanol extracts prepared from two different cultures (fruit from synthetic log culture and mycelia from PDA media culture). Both the 200 mg/mL and 100 mg/mL concentrations of extracts inhibited all the tested bacteria, and the results were promising than the corresponding control using antibiotics. The fruit extract showed higher antioxidant potential (150.6 ± 56.92 mg ascorbic acid equivalent/g extract) than mycelial extract (144.28 ± 81.72 mg ascorbic acid equivalent/g extract). The results indicate that fruiting bodies of G. lucidum cultivated in a complex dust medium possess higher antioxidant properties than mycelia culture, which can be further explored for therapeutic applications.

AbstrakPenelitian ini bertujuan untuk menganalisis senyawa fitokimia ekstrak metanol Ganoderma lucidum dengan metode GCMS. Tahapan penelitian ini meliputi  proses ekstraksi serbuk halus G. lucidum menggunakan pelarut metanol dengan metode maserasi selama 3x3 hari dan ekstrak metanol G. lucidum yang didapatkan dianalisis dengan alat GCMS. Senyawa fitokimia yang teridentifikasi oleh GCMS antara lain thiophene, 2-hexyl-; 3-((3-Acetoxythyl)-6-acetoxymethyl-2,4-dimethyl)phenyl)-2-methyl-(E)-2-propenyl acetate; 2,7-Diphenylindole; dan 5-(2-Bromophenyl)-7-chloro-2,3-dihydro-1H-1,4-benzodiazepin-2-one. Dapat disimpulkan bahwa senyawa fitokimia pada ekstrak metanol G. lucidum memiliki efek farmakologis bagi kesehatan manusia dan aktivitas biologis lainnya.  Katakunci: Ganoderma lucidum, senyawa fitokimia, GCMS AbstractThis study aims to analyze the phytochemical compounds of methanol extract of Ganoderma lucidum using the GCMS method. The stages of this study include extraction of fine powder G. lucidum using methanol solvent by maceration method for 3x3 days and methanol extract of G. lucidum obtained were analyzed by GCMS. Phytochemical compounds identified by GCMS is thiophene, 2-hexyl-; 3 - ((3-Acetoxythyl) -6-acetoxymethyl-2,4-dimethyl) phenyl) -2-methyl- (E) -2-propenyl acetate; 2,7-Diphenylindole; and 5- (2-Bromophenyl) -7-chloro-2,3-dihydro-1H-1,4-benzodiazepine-2-one. Polysaccharides from Ganoderma lucidum have been demonstrated to possess diverse biological activities. Despite lots of studies on the biological activities of Ganoderma lucidum polysaccharide (GLP), little is known regarding the medicinal potential of low–molecular weight enzymatically hydrolyzed Ganoderma lucidum polysaccharide (EGLP). EGLP was prepared by enzymatic degradation and its potential effects in U14 cervical tumor–bearing mice were evaluated. Both GLP and EGLP delayed tumor growth of the tumor xenograft. The EGLP was superior to native polysaccharide. Moreover, EGLP treatment could effectively protect the immune organs of U14 cervical carcinoma–bearing mice. In addition, the EGLP treatment ameliorated oxidative stress as compared with cyclophosphamide (CTX). Compared with the MC group, the expression of Bcl-2 and COX-2 was obviously decreased by EGLP treatment, whereas the expression of Bax and cleaved caspase-3 was obviously increased. These results indicated that EGLP showed stronger antitumor activity with lower toxic effects and had the potential to be a novel antitumor agent.
