Calibration curves giving Mv for stars of luminosity classes III, II, Ib, Iab and 0 are derived and shown graphically in the HR diagram. There are serious gaps in which the calibration needs to be improved.

This paper gives the first results of a work in progress, in collaboration with G. Michaud and G. Vauclair. It is a first attempt to compute the effects of meridional circulation and turbulence on diffusion processes in stellar envelopes. Computations have been made for a 2 Mʘstar, which lies in the Am - δ Scuti region of the HR diagram.Let us recall that in Am stars diffusion cannot occur between the two outer convection zones, contrary to what was assumed by Watson (1970, 1971) and Smith (1971), since they are linked by overshooting (Latour, 1972; Toomre et al., 1975). But diffusion may occur at the bottom of the second convection zone. According to Vauclair et al. (1974), the second convection zone, due to He II ionization, disappears after a time equal to the helium diffusion time, and then diffusion may happen at the bottom of the first convection zone, so that the arguments by Watson and Smith are preserved.

BACKGROUND Perihilar cholangiocarcinoma (pCCA) is a highly aggressive malignancy with poor prognosis. Accurate prediction is of great significance for patients’ survival outcome. OBJECTIVE The present study aimed to propose a prognostic nomogram for predicting the overall survival (OS) for patients with pCCA. METHODS We conducted a retrospective analysis in a total of 940 patients enrolled from the Surveillance, Epidemiology, and End Results (SEER) program and developed a nomogram based on the prognostic factors identified from the cox regression analysis. Concordance index (C-index), risk group stratification and calibration curves were adopted to test the discrimination and calibration ability of the nomogram with bootstrap method. Decision curves were also plotted to evaluate net benefits in clinical use against TNM staging system. RESULTS On the basis of multivariate analysis, five independent prognostic factors including age, summary stage, surgery, chemotherapy, together with radiation were selected and entered into the nomogram model. The C-index of the model was significantly higher than TNM system in the training set (0.703 vs 0.572, P<0.001), which was also proved in the validation set (0.718 vs 0.588, P<0.001). The calibration curves for 1-, 2-, and 3-year OS probabilities exhibited good agreements between the nomogram-predicted and the actual observation. Decision curves displayed that the nomogram obtained more net benefits than TNM staging system in clinical context. The OS curves of two distinct risk groups stratified by nomogram-predicted survival outcome illustrated statistical difference. CONCLUSIONS We established and validated an easy-to-use prognostic nomogram, which can provide more accurate individualized prediction and assistance in decision making for pCCA patients.

The peculiar outburst of the star baptized Sakurai’s Object (SO) is a conceivable example of a late He shell flash in a post-AGB object. The new source of nuclear energy forces such objects toward high luminosities and eventually low effective temperatures; they cross the HR diagram in a comparable fashion as FG Sge did in the past - i.e., they move noticeably on the HR diagram on human timescales. From monitoring campaigns of SO during the last year, first estimates of its cooling rate were derived and in particular cyclic light variability was established. We present first results from attempts to model stellar envelopes appropriate for SO. As we hypothesize the light variability to be attributable to stellar pulsations, we aim at constraining the basic stellar parameters based on stability analyses of our envelope models. Radial, nonadiabatic stability computations provided predictions of the modal content which should be observable as SO evolves. The particular components in such mode spectra of SO as they are to appear in the coming years should indeed help to constrain basic stellar parameters such as mass and luminosity.

The Hipparcos catalogue provides an accurate and extensive sampling of the solar neighbourhood HR diagram. The morphology of this diagram depends on selection criteria of the catalogue such as the limiting magnitude, angular separation and on the characteristics of the stellar populations near the sun (space density, metallicity, star formation rate, etc). Since the Hipparcos data are so accurate, one needs to model precisely the different selection bias and, at the same time, parametrize models of the galactic stellar populations with sufficient flexibility that as much information as possible can be grasped from the catalogue. Comparisons between our model and the Hipparcos catalogue will be presented elsewhere. Since the quantity of information contained in the Hipparcoscatalogue is so important, models ought to be complex, and external contraints, obtained prior to any general comparison with the model, are welcome. A major factor that influences the distribution of the stars in the HR diagram is the metallicity. For the late type stars, the metallicity distribution can be best studied by re-analysing a volume-limited sample of stars from the catalogue.

AbstractBackgroundInvasive fungal disease is a life-threatening condition that can be challenging to treat due to pathogen resistance, drug toxicity, and therapeutic failure secondary to suboptimal drug concentrations. Frequent therapeutic drug monitoring (TDM) is required for some anti-fungal agents to overcome these issues. Unfortunately, TDM at the institutional level is difficult, and samples are often sent to a commercial reference laboratory for analysis. To address this gap, the first paper spray-mass spectrometry assay for the simultaneous quantitation of five triazoles was developed.MethodsCalibration curves for fluconazole, posaconazole, itraconazole, hydroxyitraconazole, and voriconazole were created utilizing plasma-based calibrants and four stable isotopic internal standards. No sample preparation was needed. Plasma samples were spotted on a paper substrate in pre-manufactured plastic cartridges, and the dried plasma spots were analyzed directly utilizing paper spray-mass spectrometry (paper spray MS/MS). All experiments were performed on a Thermo Scientific TSQ Vantage triple quadrupole mass spectrometer.ResultsThe calibration curves for the five anti-fungal agents showed good linearity (R2 = 0.98–1.00). The measured assay ranges (lower limit of quantification [LLOQ]–upper limit of quantitation [ULOQ]) for fluconazole, posaconazole, itraconazole, hydroxyitraconazole, and voriconazole were 0.5–50 μg/mL, 0.1–10 μg/mL, 0.1–10 μg/mL, 0.1–10 μg/mL, and 0.1–10 μg/mL, respectively. The inter- and intra-day accuracy and precision were less than 25% over the respective ranges.ConclusionsWe developed the first rapid paper spray-MS/MS assay for simultaneous quantitation of five triazole anti-fungal agents in plasma. The method may be a powerful tool for near-point-of-care TDM aimed at improving patient care by reducing the turnaround time and for use in clinical research.

Comparative analysis of the sensitivity of two surface plasmon resonance (SPR) biosensors was conducted on a single-metallic Au sensor and bimetallic Ag–Au sensor, using a cathepsin S sensor as an example. Numerically modeled resonance curves of Au and Ag–Au layers, with parameters verified by the results of experimental reflectance measurement of real-life systems, were used for the analysis of these sensors. Mutual relationships were determined between ∂Y/∂n components of sensitivity of the Y signal in the SPR measurement to change the refractive index n of the near-surface sensing layer and ∂n/∂c sensitivity of refractive index n to change the analyte’s concentration, c, for both types of sensors. Obtained results were related to experimentally determined calibration curves of both sensors. A characteristic feature arising from the comparison of calibration curves is the similar level of Au and Ag–Au biosensors’ sensitivity in the linear range, where the signal of the AgAu sensor is at a level several times greater. It was shown that the influence of sensing surface morphology on the ∂n/∂c sensitivity component had to be incorporated to explain the features of calibration curves of sensors. The shape of the sensory surface relief was proposed to increase the sensor sensitivity at low analyte concentrations.

We use the Hipparcos survey to derive an improved model of the local galactic structure. The availability of parallaxes for all the stars permits direct determination of stellar distributions, eliminating the basic indeterminacy of classical methods based on star counts. Hipparcos gives for the first time a truly three-dimensional view of the solar vicinity, and a complete, homogeneous and highly accurate set of magnitudes and colours. This means that new techniques can be applied in the treatment of the data which place strong constraints on a model that tries to describe the local Galactic structure. Here we investigate how well a static model of low complexitycan describe the Hipparcos observations. The interpretation of the Hipparcos data is complicated by various observational errors and selection effects that are hard to treat correctly. We do not try to correct the data, but instead use a model and subject this model to the same observational errors and selection effects. A model catalogue is created that can be compared with the observed catalogue directly in the observational domain, thereby eliminating the effects from various biases. Many features in the HR diagram are for the first time seen in field stars thanks to Hipparcos, such as the slanted red giant clump, previously seen in rich old open clusters such as Berkeley 18. This and other features ofthe observed HR diagram are well reproduced by the model thanks to the rather detailed modelling of the joint Mv/B — V distribution. Actually, separate distributions were derived for the three different components, disk, thick disk and halo, using the kinematic characteristics of the components to discriminate between them.

ABSTRACTAdvances in accelerator mass spectrometry have resulted in an unprecedented amount of new high-precision radiocarbon (14C) -dates, some of which will redefine the international 14C calibration curves (IntCal and SHCal). Often these datasets are unaccompanied by detailed quality insurances in place at the laboratory, questioning whether the 14C structure is real, a result of a laboratory variation or measurement-scatter. A handful of intercomparison studies attempt to elucidate laboratory offsets but may fail to identify measurement-scatter and are often financially constrained. Here we introduce a protocol, called Quality Dating, implemented at ETH-Zürich to ensure reproducible and accurate high-precision 14C-dates. The protocol highlights the importance of the continuous measurements and evaluation of blanks, standards, references and replicates. This protocol is tested on an absolutely dated German Late Glacial tree-ring chronology, part of which is intercompared with the Curt Engelhorn-Center for Archaeometry, Mannheim, Germany (CEZA). The combined dataset contains 170 highly resolved, highly precise 14C-dates that supplement three decadal dates spanning 280 cal. years in IntCal, and provides detailed 14C structure for this interval.

AbstractPoly(ethylene adipate) (PEA) and poly(ethylene dodecanedioate) (PED) with dicarboxy-, dihydroxy-, dimethyl ester- and hydroxy-carboxy end-groups were synthesized and characterized by 13C NMR and MALDI-TOF MS. Size-exclusion chromatography (SEC) fractions of these polyesters were analyzed by MALDI-TOF MS and used as standards to obtain SEC calibration curves leading to the absolute molar masses of the unfractionated polyesters. PEA and PED calibration curves were different from each other, but the same calibration curve was obtained for each type of polyester, irrespective of the nature of end-groups, showing the predominant role of the nature of repeating units on the hydrodynamic volume of oligoesters. The study also shows that the conventional SEC calibration method using polystyrene standards leads to a significant overestimation of polyester average molar masses.
