Validity and Factor Structure of the Obsessive Compulsive Thoughts Checklist

The Obsessive Compulsive Thoughts Checklist (OCTC; Bouvard, Mollard, Cottraux, & Guerin 1989) is a 28 item questionnaire. Patients rate their degree of disturbance of the past week on a 5-point scale. The validation study and factor analysis of the OCTC is presented. Three groups were compared: patients suffering from obsessive compulsive disorders (n = 122), patients suffering from panic disorder with agoraphobia (n = 61) and a control group (n = 80). The three groups were comparable in age and sex. Obsessive compulsive patients scored significantly higher than both agoraphobic patients and control subjects. Spearman rank correlations were used to compute convergent validity in a sub-group of obsessive compulsive patients (n = 96). The Obsessive Compulsive Thoughts Checklist correlated positively with the Compulsive Activity Checklist (rho =. 62; p <. 0001). The total score was also positively correlated with the Yale-Brown Obsessive Compulsive Scale (YBOCS total scale: rho =. 42; p <. 0001; YBOCS obsession scale: rho =. 40; p <. 0001; YBOCS compulsion scale: rho. 37; p =. 0002). The factor structure was studied both on the sample which included agoraphobic patients and controls (n = 141) and on the obsessive compulsive patients only (n = 122). In both analyses, three identical factors were found: – factor 1: checking/perfectionism-orderliness – factor 2: responsibility/dread of harming others – factor 3: washing/contamination. Results support the validity and the internal consistency of the Obsessive Compulsive Thoughts Checklist. The factor analysis indicates that the OCTC is a three dimensional scale, reflecting a pathological need to check, a pathological sense of responsibility and a pathological need to wash, probably due to distorted thoughts about perfectionism and orderliness, an excessive need for control and fear of contamination.

Chronic constipation is one of the most common problems in children. It can cause anxiety and psychological problems in patients and parents. The purpose of this study was to investigate the relationship between obsessive-compulsive disorders (OCD) in children with chronic functional constipation. This is a case-control study performed at the gastroenterology clinic of Besat Hospital Sanandaj for one year. The children aged 7-14-year-old with chronic functional constipation were selected as a case group, and the healthy children were recruited as a control group, who referred for growth control. Then, the Maudsley questionnaire for obsessive-compulsive disorders was completed by a trained assistant. With this questionnaire, the presence or absence of OCD and its type can be determined. Forty-three children were selected as the case group and sixty-seven children as the control group. The mean age of the case and control group was 9.3±1.5 and 8.8±1.6 years, respectively. Based on the results, there was a significant difference between the groups in terms of checking, cleaning, slowness, doubting, and total obsessive score (P˂0.05). Therefore, all five types of obsessive-compulsive disorder in the case group were greater than the control group. Obsessive-compulsive disorders were more common in children with functional constipation, and the total obsessive score in these children than the healthy children was a significant difference.

The aim of the study was to determine the psychological characteristics of mothers of children with cerebral palsy. The study included a sample of 30 respondents. The sample consisted of mothers of children with cerebral palsy, chronologically aged 25 to 60. The research was conducted at the Centre for Children with Multiple Disabilities “Koraci Nade”. For the purpose of checking the set research aim, Symptom ChecklistSCL-90-R, consisting of a list of 90 particles (problems), was applied, and the task was to assess the respondent's anxiety about each of the aforementioned problems over the past week (e.g., headaches, tension or anxiety, excessive worrying). The survey data were processed using the parametric statistics method. Central tendency measures, dispersion measures were calculated, and a tabular presentation of the results was done. Based on the results of the research, it can be concluded that the mothers of children with cerebral palsy have the most pronounced problems in areas of somatization, depression, obsessive-compulsive disorders, as well as anxiety.

Autism Spectrum Disorder (ASD) is a pervasive developmental disorder, that is raising at a concerning rate. However, underlying mechanisms are still to be discovered. Obsessions and compulsions are the most debilitating aspect of these disorders (OCD), and they are the treatment priority for patients. SAPAP3 knock out mice present a reliable mouse model for repetitive compulsive behavior and are mechanistically closely related to the ASD mouse model Shank3 on a molecular level and AMPA receptor net effect. The phenotype of SAPAP3 knock out mice is obsessive grooming that leads to self-inflicted lesions by 4 months of age. Recent studies have accumulated evidence, that epigenetic mechanisms are important effectors in psychiatric conditions such as ASD and OCD. Methylation is the most studied mechanism, that recently lead to drug developments for more precise cancer treatments. We injected SAPAP3 mice with an epigenetic demethylation drug RG108 during pregnancy and delayed the onset of the phenotype in the offspring by 4 months. This result gives us clues about possible mechanism involved in OCD and ASD. Additionally, it shows that modulation of methylation mechanisms during development might be explored as a preventative treatment in the cases of high inherited risk of certain mental health conditions.

Behavioral flexibility is the ability of a subject to change its behavior according to contextual cues. In humans, Obsessive Compulsive Disorders (OCD) is characterized by repetitive behavior, performed through rigid rituals. This phenomenological observation has led to explore the idea that OCD patients may have diminished behavioral flexibility. To address this question we developed innovative translational approaches across multiple species, including human patients suffering from obsessive-compulsive disorders, and rodent genetic models of OCD to provide original data in the perspective of enlightening the neurocognitive bases of compulsive behaviors. Behavioral flexibility may be challenged in experimental tasks such as reversal learning paradigms. In these tasks, the subject has to respond to either of two different visual stimuli but only one stimulus is positively rewarded while the other is not. After this first association has been learned, reward contingency are inverted, so that the previously neutral stimulus is now rewarded, while the previously rewarded stimulus is not. Performance in reversal learning is indexed by the number of perseverative errors committed when participants maintain their response towards previously reinforced stimulus in spite of negative reward. Unsurprisingly, this behavioral task has been adapted to mice using various response modalities (T-maze, lever press, nose-poke). Using animal models of compulsive behaviors give much more possibilities to study the deficient functions and their underlying neural basis that could lead to pathological repetitive behaviors. Here we present new behavioral set-ups that we developed in parallel in human (i.e. healthy subjects and OCD patients) and mice (i.e. controls and SAPAP3-KO mice) to study the role of the behavioral flexibility as a possible endophenotype of OCD. We observed that the subjects suffering of compulsive behaviors showed perseverative maladaptive behaviors in these tasks. By comparing the results of a similar task-design in humans and mouse models we will discuss the pertinence of such translational approach to further study the neurocognitive basis of compulsive behaviors.
