Two novel ESIPT-based fluorescent probes for Cu2+ detection were developed. Altering the linker in probe molecules reversed their sensing behavior. Both probes exhibited high selectivity and sensitivity to Cu2+, and can be used for cell imaging.

Amide based probes containing phenyl (L1), naphthyl (L2) and anthracenyl (L3) groups were screened towards metal ions. Probes L2 and L3 display sensing for Fe2+ and Fe3+ ions. The L3–Fe3+ system is shown to have potential applications in logic gates and cell imaging.

A novel fluorescent rhodamine-based probe L for selective responding to ClO– has been synthesized and characterized. The spectroscopy showed that probe L can detect ClO– in aqueous solution without interaction with other interfering ions, and the detection is also evident by the colour change from colourless to reddish purple under white light. The remarkable fluorescence enhancement showed the high selectivity and sensitivity of probe L for the detection of ClO–. Furthermore, probe L was applied to intracellular fluorescent imaging of HeLa cells treated with ClO– and MTT assay showed nontoxicity in living cells.

A Zn(II) perchlorate complex has been prepared and characterized by single X-ray crystallography, nuclear magnetic resonance spectroscopy, thermogravimetric analysis, infrared spectroscopy, elemental analysis, and UV-vis spectroscopy. The complex crystallizes in the monoclinic space group P21/c (Z = 4) with a pentacoordinated zinc center. Interestingly, the Zn complex was found be to a potential fluorophore that could sense acetone and other ketones with high selectivity and sensitivity.

A new colorimetric and fluorescence probe NRSH based on Nile-red chromophore for the detection of biothiols has been developed, exhibiting high selectivity towards biothiols over other interfering species. NRSH shows a blue shift in absorption peak upon reacting with biothiols, from 587 nm to 567 nm, which induces an obvious color change from blue to pink and exhibits a 35-fold fluorescence enhancement at 645 nm in red emission range. NRSH displays rapid (<1 min) response for H2S, which is faster than other biothiols (>5 min). The detection limits of probe NRSH towards biothiols are very low (22.05 nM for H2S, 34.04 nM for Cys, 107.28 nM for GSH and 113.65 nM for Hcy). Furthermore, NRSH is low cytotoxic and can be successfully applied as a bioimaging tool for real-time monitoring biothiols in HeLa cells. In addition, fluorescence mechanism of probe NRSH is further understood by theoretical calculations.
