Objective: Vitamin D deficiency may contribute to Systemic Lupus Erythematosus (SLE) development. Vitamin D may involve in pathogen recognition through Toll-like receptor (TLR) 2 in immune cells in saliva. This study aimed to determine the correlation between serum vitamin D/25(OH)D and TLR2 expression of immune cells in the saliva of SLE. Materials and methods: This cross-sectional study conducted at the the SLE patients who met the inclusion and exclusion criteria. Those who had signed informed consent involved to underwent unstimulated saliva collection and blood samples for TLR2 and vitamin D/25(OH)D examination. The correlation between serum vitamin D/25(OH)D concentration and salivary TLR2 expression was analyzed using the correlation test, linear regression with 95% confidence level. Results: Thirty SLE patients had a mean serum vitamin D/25(OH)D concentration of 9.98 ± 4.64 ng/ml. The mean of TLR2 expression of CD11b+ cells in saliva was 26.03 ± 20.92%. There was a significant positive correlation between serum vitamin D/25(OH)D concentration and TLR 2 expression of CD11b+ cells in the saliva. (r = 0.434; P < 0.05). Vitamin D/25(OH)D was the only predictor for TLR 2 expression. Conclusion: Serum vitamin D/25(OH)D concentrations associated with TLR2 expression of CD11b+ cells in the saliva of SLE.

BACKGROUND: Systemic Lupus Erythematosus (SLE) is an autoimmune disorder mediated by inflammatory cytokines. Decreasing vitamin D levels is a common feature in SLE patients. Vitamin D has the capacity in suppressing inflammatory cascade. Seluang fish (Rasbora spp.) contained a high level of vitamin D with the potential as a new therapeutic modality. AIM: This study aimed to assess the efficacy of Seluang fish oil against proinflammatory cytokines, vitamin D levels, and clinical conditions of SLE. METHODS: A randomised, double-blind, clinical trial study design was conducted. The subjects were 16 SLE subjects treated with 500uL Seluang fish oil capsules and 16 SLE subjects with placebo capsules. Measurement of vitamin D, IL-1, IL-6 and IL-17 levels were performed with ELISA. Clinical assessment of SLE was performed with MEX-SLEDAI. Bivariate analysis, T-test, was performed. Data were presented in the form of mean ± SD. RESULTS: The administration of Seluang fish oil was clinically able to show efficacy assessed by the MEX SLEDAI score. Significant results were also shown by increased vitamin D levels and reduced levels of IL-1, IL-6 and IL-17, in Seluang fish oil group. CONCLUSION: Seluang fish oil possessed the efficacy of reducing the inflammatory response in SLE patients by increasing serum vitamin D levels.

Background and aims: : Systemic lupus erythematosus (SLE) is an autoimmune disease which involves various organs. Vitamin D is an essential ingredient in regulating the immune system. This study aimed to investigate the relationship between vitamin D and the severity of lupus activity. Materials and Methods: This case-control study was carried out on 38 patients with lupus on the basis of the American College of Rheumatology (ACR) criteria and 44 healthy subjects with no history of rheumatologic disease. To measure the level of 25-hydroxy vitamin D, venous blood samples (5 cc) were taken from each participant and the activity of the lupus disease was measured by the Systemic Lupus Erythematosus Disease Activity Index (SLEDAI) scale. Finally, the chi-square test, independent sample t test, one-way ANOVA, and multiple linear regression analysis were used to measure multivariate effects. The level of significance was set to be P<0.05. Results: Thirty-five lupus patients and 40 healthy subjects were females (P=0.847). Vitamin D deficiency was observed in the case (42.1%) and control (11.4%) groups. The mean value of serum vitamin D3 level was 35.3 ng/mL in the control group, as well as 24.6 ng/mL and 21.3 ng/mL in patients with mild and severe SLE, respectively (P=0.024). Conclusion: In this study, high levels of 25-hydroxy vitamin D were observed among the healthy subjects compared to patients with SLE. Eventually, the level of vitamin D significantly decreased by increasing the severity of SLE activity.

Abstract Background Recent evidence has suggested that the 1,25(OH)2D3/Vitamin D receptor (VDR) acts to suppress the immune response associated with systemic lupus erythematosus (SLE), a serious multisystem autoimmune disease. Hence, the aim of the current study was to investigate the mechanism by which 1,25-(OH)2D3/VDR influences SLE through regulating the Skp2/p27 signaling pathway. Methods Initially, the levels of 1,25(OH)2D3, VDR, Skp2, and p27 were measured in collected renal tissues and peripheral blood. Meanwhile, the levels of inflammatory factors, biochemical indicators (BUN, Cr, anti-nRNP IgG, anti-dsDNA IgG) and urinary protein levels were assayed in in VDRinsert and VDR-knockout mice in response to 1,25(OH)2D3 supplement. In addition, the distribution of splenic immune cells was observed in these mice. Results Among the SLE patients, the levels of 1,25(OH)2D3, VDR and p27 were reduced, while the levels of Skp2 were elevated. In addition, the levels of anti-nRNP IgG and anti-dsDNA IgG were increased, suggesting induction of inflammatory responses. Notably, 1,25(OH)2D3/VDR mice had lower concentrations of BUN and Cr, urinary protein levels, precipitation intensity of the immune complex and complement, as well as the levels of anti-nRNP IgG and anti-dsDNA IgG in SLE mice. Additionally, 1,25(OH)2D3 or VDR reduced the degree of the inflammatory response while acting to regulate the distribution of splenic immune cells. Conclusion This study indicated that 1,25-(OH)2D3/VDR facilitated the recovery of SLE by downregulating Skp2 and upregulating p27 expression, suggesting the potential of 1,25-(OH)2D3/VDR as a promising target for SLE treatment.

Objectives To investigate possible associations between 25-hydroxyvitamin D3 (25(OH)D3), oestradiol (E2) and IFN-gamma (IFNγ) in female patients with inactive systemic lupus erythematosus (SLE). Methods Female patients with inactive SLE and age-matched healthy controls were recruited into this cross-sectional study. Serum concentrations of 25(OH)D3, E2 and IFNγ were measured by radioimmunoassay with gamma-counters and enzyme-linked immunosorbent assay. Results 36 patients and 37 controls were enrolled. In patients with SLE, the concentration of 25(OH)D3 was lower and E2 was higher compared with controls. In vitamin D deficient (i.e., 25(OH)D3≤20 ng/ml) patients, IFNγ was 150% higher compared with patients with 25(OH)D3>20 ng/ml and controls. The concentration of E2 was higher in all patients compared with controls independently of the vitamin D level. A difference was found between patients and controls in the correlation of 25(OH)D3 with E2 and a positive correlation was found between E2 and IFNγ in all participants. Conclusions Our results suggest that E2 may have a strong modulating effect on vitamin D function which is significant only at low concentration of E2.
