To control the spread of bovine spongiform encephalopathy, several DNA methods have been described for the detection of the species origin of meat and bone meal. Most of these methods are based on the amplification of a mitochondrial DNA segment. We have developed a semiquantitative method based on real-time PCR for detection of ruminant DNA, targeting an 88-bp segment of the ruminant short interspersed nuclear element Bov-A2. This method is specific for ruminants and is able to detect as little as 10 fg of bovine DNA. Autoclaving decreased the amount of detectable DNA, but positive signals were observed in feeding stuff containing 10% bovine material if this had not been rendered in accordance with the regulations, i.e., heated at 134°C for 3 instead of 20 min.

The pancreatic microcirculation in noninbred ob/ob mice and normal controls was visualized in a fluorescence microscope and examined for responses to intravenous infusions of epinephrine or norepinephrine. Evaluations of coded video recordings revealed a smooth dose-response relationship and validated a semiquantitative method of analysis. In ob/ob mice the islet microcirculation was markedly and reversibly inhibited by 0.14–4 micrograms of epinephrine X min-1 X kg body wt-1; the flow was almost totally stopped at the highest infusion rates. Capillary flow in the exocrine pancreas appeared unaffected, except for some inhibition at 4.0 micrograms X min-1 X kg-1. Norepinephrine was less potent an inhibitor of islet blood flow. Normal lean mice exhibited minor responses to 1–11.3 micrograms of epinephrine X min-1 X kg-1; in most cases there was no visible effect. It is concluded that there is a selective regulation of blood flow through the endocrine portion of the pancreas and that the islet microcirculation is hypersensitive to catecholamines in noninbred ob/ob mice. A defective inhibitory influence from the brain may play a role in the development of excessive hyperinsulinemia in ob/ob mice.

Objective.Ankylosing spondylitis (AS) is a chronic inflammatory disease mainly affecting the axial skeleton and characterized by ossification of the spinal disc, joints, and ligaments leading to progressive ankylosis. Vertebral osteoporosis is a recognized feature of AS. Studies have confirmed a moderate to high prevalence of vertebral fractures with extremely varying ranges in patients with AS. Our objective was to estimate the prevalence of vertebral fractures in a representative Spanish population of patients with AS using a validated semiquantitative method, MorphoXpress®.Methods.Patients were randomly selected from the 10 initial participating centers of the Spanish National Registry of Spondyloarthropathies (REGISPONSER) by consecutive sampling. All patients fulfilled the New York modified criteria for AS and had a baseline thoracolumbar radiograph. A prevalent vertebral fracture was defined according to the Genant classification criteria.Results.The estimated prevalence of vertebral fractures was 32.4% (95% CI 25.5%–39.3%). The majority of fractures were localized in the thoracic segment (n = 100; 82.%) and were mild (n = 79; 64.8%). In logistic regression analysis, age (odds ratio per year 1.05, 95% CI 1.03–1.08, p < 0.001), disease duration (OR per year 1.03, 95% CI 1.01–1.06, p = 0.011), Bath Ankylosing Spondylitis Functional Index score (OR per score 1.16, 95% CI 1.03–1.30, p = 0.015), Bath Ankylosing Spondylitis Radiographic Index-TS (OR per score 1.25, 95% CI 1.12–1.39, p < 0.001), and wall-occiput distance (OR per cm 1.15, 95% CI 1.08–1.23, p < 0.001) were all associated with prevalent fracture.Conclusion.Semiquantitative methods are needed to improve the diagnosis of vertebral fractures in AS in order to start early treatment and to avoid complications arising from osteoporosis.

Frequency domain analysis of heart rate variability (HRV) has been proposed as a semiquantitative method for assessing activities in the autonomic nervous system. We examined whether absolute powers, normalized powers, and the low frequency-to-high frequency ratio (LF/HF) derived from the HRV power spectrum could detect shifts in autonomic balance in a setting with low sympathetic nervous tone. Healthy subjects were examined for 3 h in the supine position during 1) control conditions ( n = 12), 2) acute β-blockade ( n = 11), and 3) chronic β-blockade ( n = 10). Heart rate fell during the first 40 min of the control session (72 ± 2 to 64 ± 2 beats/min; P < 0.005) and was even lower during acute and chronic β-blockade (56 ± 2 beats/min; P < 0.005). The powers of all spectral areas rose during the first 60 min in all three settings, more so with β-blockade ( P < 0.05). LF/HF was found to contain the same information as powers expressed in normalized units. LF/HF detected the shift in autonomic balance induced by β-blockade but not the change induced by supine position. In conclusion, none of the investigated measures derived from power spectral analysis comprehensively and consistently described the changes in autonomic balance.

When acetic acid is handled in aluminum, it is important, for reasons of corrosion resistance, that the acid be not completely anhydrous, a condition that results if traces of acetic anhydride are present. A quick colorimetric test depending on the reaction between acetic anhydride with aniline has been developed by which it is possible to detect 0.05% acetic anhydride in acetic acid.

Abstract We evaluated a new rapid semi-quantitative immunometric assay of C-reactive protein (CRP) as a screening test for sepsis by comparison with an automated nephelometric method. Plasma samples (n = 101) from preterm infants during the first week of life were saved for CRP analyses. We measured CRP by the Nycocard semiquantitative method and compared the results with those obtained with a Behring Nephelometer. A CRP value less than 10 mg/L was considered to be negative for infection. All CRP results read as less than 10 mg/L (negative) by the Nycocard method were also less than 10 mg/L by the comparison method, and all CRP values found to be greater than 20 mg/L (positive) by the Nycocard method were also positive by the comparison method. Results in the 10-20 mg/L range were considered equivocal. We conclude that the Nycocard CRP semi-quantitative method is a rapid and useful screening test for sepsis in preterm infants.
