Oestrus synchronisation and fixed time artificial insemination in beef heifers

To compare two oestrus synchronisation protocols in beef heifers presynchronised with two prostaglandin F2α (PG) treatments, 36 h after the second PG treatment one group (hCG-OB group; n = 112) received 250 IU of hCG and 0.5 mg of oestradiol benzoate (OB) followed by fixed time artificial insemination (FTAI) 30 h later. Additional oestrus detection (OD) and artificial insemination (AI) was done in this group for 3 days after FTAI; heifers rebred during this period were considered open to the FTAI. The Control group (n = 112) received only OD and AI for 6 days after the second PG treatment. Follicular size was measured at the time of AI in a subsample of heifers (59 in the hCG-OB group and 34 in the Control group). Results were evaluated separately either considering heifers pregnant at FTAI or those pregnant at FTAI + 3-day OD + AI. The FTAI conception rate was 45.5 vs 76.5% for the Control group (P < 0.0001) and pregnancy rates were 45.5 and 67.0% (P = 0.0014); FTAI + OD + AI conception rates were 63.4 and 76.2% (P = 0.0404) and pregnancy rates 63.4 and 62.8% (P = 0.5719) (hCG-OB and Control groups, respectively). There was a significant correlation (r = 0.713, P < 0.01) between follicle size at FTAI and pregnancy in the hCG-OB group, but not in the Control group (r = 0.3465, P > 0.1). In conclusion, acceptable synchronised conception rate and pregnancy rate were obtained by using this protocol, and the results were improved by complementing it with OD + AI for 3 days after FTAI.

Aim: This study aimed to evaluate the pregnancy rates in hair ewes using an Ovsynch synchronization protocol under a breeding system that combines fixed-time insemination plus a 10-day mating period as an alternative. Materials and Methods: Through an experimental study (n=27), ewes were randomly located into one of three treatments: (1) Pre-synch (n=9): Prostaglandin F2α (PGF2α)+Gonadotropin-releasing hormone (GnRH)+PGF2α+GnRH; (2) Ovsynch (n=9): GnRH+PGF2α+GnRH; and (3) control: Ewes bred by natural mating (NM) (n=9). Ewes were fixed-time inseminated (fixed-time artificial insemination [FTAI]) with fresh semen, collected just before the insemination time through vaginoscopy at 16 h after the second GnRH (gonadorelin) injection. Each experimental group was placed separately during 15 days and, after this time, fertile rams were allowed back with ewes for a 10-day mating period. Control group ewes remained with the rest of the herd suitable for breeding and were bred under NM. Pregnancy diagnosis was performed by ultrasound at 28-, 56-, and 84-day post-breeding to differentiate between FTAI and NM pregnancies. Total (FTAI±NM) pregnancy rates at 56-day post-breeding were used to compared Pre-synch, Ovsynch, and control. For this purpose, two-tailed proportions comparison z-test was used with a 95% confidence level, for testing as the null hypothesis whether two proportions were equal. Results: Pregnancy rates were higher in control ewes (66.4%) than FTAI (46.6%). When pregnancy rates after a 10-day mating period (40%) were added, the final rate (86.6%) was significantly (p<0.05) higher in Ovsynch-based protocols. The pregnancy rate was significantly lower in FTAI ewes compared to FTAI +10-day mating group (p<0.05). The overall pregnancy rate was 88.0, 85.7, and 67.0 (p>0.05) for Pre-synch, Ovsynch, and control ewes, respectively. Conclusion: These results provide evidence on the benefits of combined FTAI protocols for improving the reproductive efficiency of sheep.

The study was undertaken to find out the estrus induction and conception rate in 18 postpartum anestrus Surti buffaloes divided equally into three groups and treated with Ovsynch protocol alone and in combination with PRID, keeping one group as control. The use of Ovsynch alone and Ovsynch + PRID protocols with fixed-time artificial insemination resulted in 100% estrus induction with 66.66% and 50.00% conception rate at induced estrus, respectively, and the overall conception rate was 83.33% in both the groups. In untreated anestrus control (n=6), only four (66.66%) buffaloes exhibited spontaneous estrus within 120 days of follow-up and conceived giving the first service and overall conception rate as 75.00% and 66.66%, respectively. The mean number of services per conception did not differ significantly among the treatment and control groups. The service periods of buffaloes under both the treatment groups were significantly (p less than 0.05) shorter (57.00±2.81 and 61.20±3.11 days) than the control group (104.00±20.22 days). Thus, the Ovsynch and Ovsynch + PRID protocol can be successfully used to induce fertile estrus in postpartum anestrus buffaloes.

Abstract To determine effects of presynchronization and delayed fixed-time artificial insemination (TAI) on pregnancy rates to TAI (PR/AI) with sex-sorted semen, 2,855 Bos taurus beef heifers were enrolled in a completely randomized design. Within location, heifers were randomly assigned to one of eight treatments: 1 and 2), heifers were exposed to the 7-d CO-Synch+CIDR protocol wherein they received gonadotropin-releasing hormone (GnRH) and a CIDR insert on d 0, prostaglandin F2α (PGF) upon CIDR removal on d 7, and were TAI 54 ± 2 h later with conventional (CTRL54-CNV; n = 359) or sex-sorted semen (CTRL54-SEX; n = 356); 3 and 4), same as CTRL54 but were TAI at 72 ± 2 h with conventional (CTRL72-CNV; n = 366) or sex-sorted semen (CTRL72-SEX; n = 360); 5 and 6), same as CTRL54 but also received PGF 7 d prior to initial injection of GnRH (d -7) and were TAI with conventional (PRE54-CNV; n = 355) or sex-sorted semen (PRE54-SEX; n = 353); 7 and 8), same as PRE54 but had TAI delayed to 72 ± 2 h and were inseminated with conventional (PRE72-CNV; n = 351) or sex-sorted semen (PRE72-SEX; n = 355). All heifers received estrus detection patches on d 7, which were evaluated for activation at TAI. Ultrasonography was performed 30–45 d after TAI to determine PR/AI. Estrus expression was greater (P &lt; 0.01) in the CTRL72 heifers compared to CTRL54, PRE54, and PRE72 heifers (73.0 vs 55.4, 43.4, and 65.7%, respectively). Moreover, estrus expression was greater (P &lt; 0.01) in PRE72 heifers compared to CTRL54 and PRE54 heifers. Within treatment pairs, PR/AI were greater (P ≤0.04) when conventional semen was utilized compared to sex-sorted semen. Furthermore, PR/AI were greater (P = 0.02) in PRE72-SEX heifers than CTRL54-SEX heifers (45.8 vs. 37.5%). No difference (P = 0.20) was determined in PR/AI between CTRL54-CNV and PRE72-SEX heifers (50.5 vs. 45.8%). In conclusion, presynchronization in combination with delayed TAI increased estrus expression and PR/AI with sex-sorted semen in replacement beef heifers.

The present study was conducted to investigate the efficacy of Ovsynch and Ovsynch plus protocol in postpartum (day 60) Sahiwal cows (n=18). Animals were randomly divided into three equal groups, viz., Ovsynch group Ovsynch plus group and Control group. Animals of group I (n = 6) were treated with traditional Ovsynch protocol. The animals (n = 6) of group II were treated with Ovsynch plus protocol which consisted of an initial intramuscular injection of eCG (Folligon) @ 250 IU on day 60 postpartum followed 3 days later by GPG (Ovsynch) protocol. In group-III Control, no treatment was given to animals (n = 6). Treated animals were inseminated at a fixed time between 14 and 20 hrs after second GnRH injection, irrespective of estrus detection. Blood samples were collected from each animal on days 50 and 60 postpartum to determine the status of cyclicity in animals based on serum concentrations of progesterone (P4). A third blood sample was collected on the day of prostaglandin treatment to determine the response of first GnRH injection. Four animals each were cyclic, and two were acyclic in both treatment groups. Four animals each responded to first GnRH treatment in both treatment groups. Similarly, two animals each got conceived giving conception rate of 50% (2/4) in each treatment. In the control group, one out of 6 animals got conceived yielding 16.66 % conception rate (1/6) during the study period. It may be thus concluded that Ovsynch and Ovsynch plus protocol may be used during the early postpartum period to improve the reproductive efficiency in postpartum Sahiwal cows.

AbstractThe objective was to evaluate different oestrous synchronization schemes in beef cattle under range conditions. In experiment 1, 202 heifers averaging 26 months of age were assigned randomly to three treatments: (a) GnRH-PG. (no. = 44) day 0 (D0): injection of GnRH; D7: injection of PGF2α; D0 to D25 oestrous detection (OD) and artificial insemination (AI); (b) OD-PG. (no. = 45) D0 to D4; OD + AI; D5: injection of PGF2α; D5 to D25 OD + AI; and (c)Ovsynch. (no. = 113) D0: injection of GnRH; D7: injection of PGF2α; D9: injection of GnRH and 16 h later AI at fixed time. In experiment 2, 318 non-suckling cows were assigned randomly to three treatments: (a) GnRH-PG. (no. = 106) D0: injection of GnRH; D7: injection of PGF2α; D0 to D25 OD + AI; (b) GnRH-MAP-PG. (no. = 106) D0: injection of a GnRH analogue and insertion of an intravaginal sponge impregnated with 250 mg of medroxyprogesterone acetate; D7: injection of PGF2α and sponge withdrawal; D0 to D25 OD + AI; (c) controls. (no. = 106) OD and AI. The experimental period lasted 25 days and, with exception of the Ovsynch treatment, oestrous detection was carried out twice a day (a.m./p.m.) and inseminations performed 12 h later. The pregnancy rates assessed by ultrasonography 30 days after AI were: experiment 1: (a) 30·0%; (b) 28·6% and (c) 62·3% (P < 0·05); experiment 2: (a) 60·2%; (b) 57·8% and (c) 45·5% (P < 0·05). It is concluded that in heifers and non-suckling cows, oestrous synchronization treatments result in higher pregnancy rates. In non-suckling cows, the addition of a progestagen did not improve the response.

A comparative hormonal study of prostaglandin, gonadotropin releasing hormone and their combination was conducted on 30 randomly selected cross-bred dairy cows of SEMEX project for artificial insemination in Hillat Kuku, Khartoum North, Sudan. The study attempts to determine the response of the animals to hormonal induction and synchronization of estrous as well as fertility following fixed time artificial insemination. Three treatment groups of 10 cows each were undertaken. Groups 1, 2 and 3 involved administration of prostaglandin F2α (PGF2α), gonadotropin releasing hormone (GnRH) and a combination of PGF2α + GnRH, respectively. Descriptive statistical analysis was performed. The results revealed that all the three protocols induced and synchronized estrous were almost synchronous in all the treated groups. However, protocol 3 revealed a significantly (p<0.05) better result of 70% pregnancy rate compared to other two protocols that equally provided 50% pregnancy rates. The study resolves that protocol 3 be highly recommended to change the mindset of farmers about the spread of reproductive technology in Sudan. Protocols 1 and 2 provided acceptable pregnancy rates that can be enhanced with the improvement of management.

Abstract The present study aimed to determine the effects of progesterone-based gonadotropin-releasing hormone (GnRH), prostaglandin F2α (PGF2α), and equine chorionic gonadotropin (eCG) injections on progesterone profiles and pregnancy rates in cows with no estrus symptoms within 60 days after parturition. A total of 80 cows were included in the study. All animals had the progesterone-releasing device PRID®Delta placed intravaginally for nine days with an injection of GnRH. On the eighth day, PGF2α was injected, and PRID®Delta was removed from the vagina on day nine. Artificial insemination was carried out 60 hours after PRID®Delta removal. In half of the animals (n = 40), 600 IU of eCG was injected when PRID®Delta was removed on the ninth day before artificial insemination 60 hours later. Blood samples were taken from the tail vein on days 0 and 8 to determine progesterone levels. The pregnancy rate in the group that received eCG was 37.5%, while it was 27.5% in those that did not (P = 0.4). While the dominant follicle diameter was 15.5 mm in cows injected with eCG during timed artificial insemination, the follicle diameter was 12.4 mm in cows with no eCG injection (P <0.001). There were no differences in serum progesterone values in blood samples taken until the time of artificial insemination. However, progesterone values in the blood taken during artificial insemination were 0.94 ng/ml in the eCG- group and 0.72 ng/ml in the eCG+ group (P <0.05). As a result, it was determined that eCG injections, in addition to progesterone-based GnRH and PGF2α applications, increased the pregnancy rates in cows without symptoms of estrus. The dominant follicle diameter was larger in cows treated with eCG during artificial insemination; however, follicle size did not increase the pregnancy rate.

Ovulatory response to the initial gonadotrophin-releasing hormone (GnRH) of the CO-Synch protocol is affected by circulating progesterone (P4) and follicle size. In addition, heifers that ovulate to the initial GnRH treatment have greater fertility after AI. Thus, this study determined the effect of (1) presynchronization (Presynch) before a 6-day CO-Synch protocol and (2) circulating [RCE1] (P4) on ovulatory response, oestrus expression, and pregnancies per AI (P/AI) in beef heifers. Yearling beef heifers (n=233) at three locations were randomly assigned in a 2×2 factorial design to the following treatments: (1) Presynch+6-day CO-Synch with a new P4 device; (2) Presynch+6-day CO-Synch with a once-used P4 device; (3) 6-day CO-Synch with a new P4 device; and (4) 6-day CO-Synch with a once-used P4 device. Presynch consisted of insertion of a new P4 intravaginal device (1.38g of P4) on Day −17 and removal of P4 device on Day −11 concurrently with 500µg of cloprostenol sodium (PGF). On Day −9, all heifers received either a new (New) or once-used (Used) CIDR and 100µg of gonadorelin acetate (GnRH). Six days later (Day −3), CIDRs were removed, 1000µg of PGF was administered and an oestrous detection patch applied (Estrotect). At 72h after CIDR removal, AI was performed concurrently with administration of 100µg of GnRH. Pregnancy was determined by transrectal ultrasonography 31 days after AI. A subset of heifers (n=155) was examined on Day −9 and Day −3 by ultrasonography to determine ovulation to Day −9 GnRH. Data were analysed using generalized linear mixed models (SAS 9.4; SAS Institute Inc.). Presynch heifers had larger follicle diameter on Day −9 (12.7±0.3 vs. 10.1±0.3 mm; P&lt;0.001), greater ovulatory response to Day −9 GnRH (82.5%; 66/80 vs. 56%; 42/75; P&lt;0.001), greater expression of oestrus (90.6%; 106/117 vs. 78.4%; 91/116; P&lt;0.02), and earlier oestrus (49.8±1 vs. 53.1±1 h; P&lt;0.01) compared with controls. There was a treatment×CIDR interaction on oestrous expression, such that a lesser (P&lt;0.05) percentage of control heifers with new CIDR expressed oestrus compared with all other groups (Table 1). Heifers with a used CIDR during the 6-day CO-Synch tended (P=0.08) to have greater P/AI (52.1%; 61/117) than those with a new CIDR (40.5%; 47/116). In conclusion, presynchronization before initiation of a 6-day CO-Synch increased follicle diameter, enhanced ovulatory response and oestrous expression, but did not affect fertility. The earlier onset of oestrus in presynchronized heifers suggests that the timing of AI may need to be modified. Table 1. SummaryObjective: The aim of this study was to monitor the ovarian response and conception rate following estrous synchronization using CIDR, Ovsynch and double prostaglandin F2α protocols in Egyptian buffalo heifers. Material and methods: A total of 80 cyclic buffalo heifers were divided into four equal groups: CIDR (intravaginal progesterone releasing device, EAZI-BREEDTM CIDR®), Ovsynch (GnRH, PGF2α, GnRH injections), PGF (double PGF2α doses) and control. Timed artificial insemination (TAI) was performed in all heifers. All animals were exa - mined using ultrasound and blood samples were collected for measurement of progesterone. Results: A new follicular wave occurred earlier in the Ovsynch and PGF groups than in the CIDR group (p < 0.05). The mean diameter of the ovulatory follicle was smaller in the CIDR group than in the Ovsynch and PGF groups (p < 0.05). The ovulation rate was 100% in the CIDR group, 75% in the Ovsynch group and 70% in the PGF group. In the control group a lower pregnancy rate (20%) was determined in than in the CIDR (35%), Ovsynch (40%) and PGF (35%) groups. Progesterone concentrations were numerically higher in pregnant heifers of the CIDR group but the difference was non-significantly compared to the Ovsynch, PGF and control groups (p > 0.05). Conclusion and clinical relevance: EAZIBREEDTM CIDR®, Ovsynch-based TAI and PGF protocols were effective in synchronizing oestrus and resulted in nearly similar pregnancy rates in Egyptian buffalo heifers.

Three experiments were conducted to determine the effects of presynchronization and treatment with equine chorionic gonadotropin (eCG) on corpus luteum (CL) and ovarian follicular development, plasma progesterone concentrations, and pregnancy rates in beef heifers subjected to a gonadotropin releasing hormone (GnRH)-based, fixed-timed AI (TAI) protocol. All heifers were given GnRH on day 0, prostaglandin F2α (PGF) on day 7, and a second GnRH on day 9 concurrent with TAI (54 h after PGF). In exp. 1 (N = 148), presynchronization with PGF (days -22 and -11) decreased the percentage of heifers with non-luteal plasma progesterone concentrations on day 0 (5.4 vs 29.7%) and day 7 (0 vs 11.6%; P < 0.05), but not on day 9 (74.3 vs. 66.2%; P > 0.20), and reduced the number of heifers in estrus and bred before TAI (P < 0.05). Although presynchronization reduced preovulatory follicle diameter (12.9 ± 0.3 vs. 14.9 ± 0.3 mm; mean ± SEM; P < 0.01), it did not affect TAI pregnancy rates (36.5 vs. 29.7%; P > 0.20). In exp. 2, heifers (N = 128) were presynchronized with melengestrol acetate (MGA) (days -27 to -12), and received a controlled internal drug release (CIDR) on day 0; on day 7, half were given 300 IU of eCG at CIDR removal. Treatment with eCG tended to increase preovulatory follicle diameter in heifers that did not ovulate to GnRH on day 0 (P = 0.06), but did not affect the percentage of heifers with non-luteal plasma progesterone concentrations on day 9 (57.8 vs. 57.8%) or TAI pregnancy rates (48.4 vs. 53.1%; P > 0.20). Experiment 3 was a 2 × 2 factorial arrangement of presynchronization (PGF concurrent with a CIDR on day -7) and eCG treatments (on day 7) applied to heifers in three herds (A, N = 150, B, N = 260 and C, N = 40). All heifers had a once-used CIDR from days 0 to 7. Presynchronization increased the percentage of heifers (Herd A) with low-luteal plasma progesterone concentrations on day 0 (70.7 vs. 22.7%) and day 7 (90.7 vs. 53.3%; P < 0.01), but did not affect the percentage of heifers with non-luteal concentrations of progesterone on day 9 (97.3 vs. 93.3%; P > 0.20). Combined for all herds, presynchronization reduced the prevalence of a CL on day 0 (23.5 vs. 73.7%; P < 0.01), and increased the prevalence of follicles ≥ 10 mm on day 7 (96.8 vs. 86.7%; P < 0.01); however, TAI pregnancy rates (195/439 = 44.4%) were not improved by either presynchronization or eCG treatment (P > 0.20).Key words: Presynchronization, equine chorionic gonadotropin, GnRH, fixed-time artificial insemination, progesterone
