Effects of Yerba Mate (Ilex paraguariensis) supplementation on the productive performance of dairy cows during mid-lactation

Yerba Mate (Ilex paraguariensis), a tea known for its high antioxidant content, was fed (250 g/cow.day) to 8 of 16 Holstein cows for 6 weeks to assess its effect on their performance. Cows were weighed and blood samples were taken on Weeks 0, 3 and 6. Blood samples were centrifuged and plasma was analysed for reactive oxygen metabolites, biological antioxidant potential, advanced oxidation protein products and non-esterified fatty acids. Cows were milked two times daily and milk yields were recorded daily for individual cows. On Weeks 0 and 6, individual milk samples were collected from two consecutive milkings, composited, and analysed for somatic cell counts, fat and true protein concentrations. Plasma concentrations of reactive oxygen metabolites, biological antioxidant potential and non-esterified fatty acids were not affected by Yerba Mate supplementation. Similarly, no effect of Yerba Mate supplementation was noted on milk fat and protein content and on somatic cell counts. This study indicates that supplementation of dairy cows’ diet with Yerba Mate during mid lactation seems to improve milk yield when cows are fed with maize silage; however, even if the effect on milk yield was significant it was quite small and needs to be validated with further studies. Cows’ oxidative status was not affected by Yerba Mate supplementation indicating that the effect of Yerba Mate on their productive performances is not mediated by changes in redox status.

We aimed to determine the level of omentin-1 hormone and other in periparturient period of dairy cows. It was also aimed to determine whether there is a correlation between omentin-1, glucose, Non-esterified fatty acids (NEFA), triglycerides (TG) and β-hydroxybutyrate (BHB). Blood samples were taken a month before parturition (PreP30), two weeks before parturition (PreP14), at parturition (P), two weeks after parturition (PostP14) and a month after parturition (PostP30). Concentrations of serum omentin-1 at P, serum glucose at P, PostP14, and PostP30, serum NEFA at P, serum TG PreP30 and PreP14, and serum BHB at P were statistically significantly higher than at other times. A positive correlation was observed between omentin-1 and glucose, NEFA and BHB, glucose and NEFA and BHB, and NEFA and BHB. A negative correlation was found between TG and omentin-1, glucose, NEFA and BHB. In conclusion, findings of the present study shows that omentin-1 may play an important role in the periparturient period. A positive correlation of omentin-1 with energy fuels NEFA, glucose, and BHB suggests that omentin-1 plays a role in energy metabolism like other adipokines. The fact that omentin-1 levels increase during delivery, when the fetus needs energy most, supports this hypothesis.

Background: Lameness in dairy cows is classified as a gait change caused by multifactorial process. The phase of the peripartum causes intense physiological changes for the adaptation of late gestation and onset of lactation. The aim of this study was to characterize the changes in the biochemical profile and productive performance in dairy cows with lameness during postpartum period.Materials, Methods & Results: This study was conducted at the University of São Paulo farm, in Pirassununga, São Paulo State, Brazil, from January to March 2017. A total, of 48 multiparous (2 to 3 lactations and 3-4 years old) dairy cows, that had the milk production of 9,200 kg/ dairy cow in a period of 305 days in the previous lactation, were included in the study. All cows were managed under the same conditions and nutritional regimen. Evaluation of body condition score was performed by a single person on -18, -12, -8, -5, and -2 days before parturition, at parturition, and on days 1, 7, 14, 21, 30, 45 and 60 after parturition. Milk production was recorded on days 7, 14, 21, 30, 45 and 60 after parturition and saved in the software program. Blood samples were performed on -18, -12, -8, -5, and -2 days before parturition, at parturition, and on days 1, 7, 14, 21, 30, 45 and 60 after parturition. Blood samples were assayed for albumin, calcium, cholesterol, triglycerides, non-esterified fatty acids, β-hydroxybutyrate, urea, creatinine, gamma-glutamil-transferase and total protein concentrations. Dairy cows were divided into a lame group (11) and normal group (37) based on locomotion score from parturition to seven days postpartum. Lame cows was classified if their score was > 2, and normal cows was classified if their score was ≤ 2 and free of any disease. Dairy cows that suffer by any health disorder other than lameness were excluded from this study. Cow diagnosed with lameness outside the diagnostic period were excluded from this study. The averages of the milk production, body condition score and biochemical profile were compared with the Tukey’s test. Lame cows showed lower (P < 0.05) concentrations of albumin (on days -18 -12, -8, -5, -2 relative to parturition, at parturition, and on days 7, 14, 21, 45 and 60 after parturition) calcium (on days -18, -12, -8, -5 and -2 before parturition, and on days 7, 14, 21, 45 and 60 after parturition), cholesterol (on days -12, -8, -5 before parturition and on days 7, 14, 21, 30, 45 and 60 after parturition) than normal cows. Cow with lameness showed higher (P < 0.05) concentration of triglyceride (on days 7, 14, 21, 30, 45 and 60 after parturition), non-esterified fatty acids (on days 1, 7, 14, 21, 30, 45 and 60 after parturition) and β-hydroxybutyrate (on days -12, -5 and -2 before parturition, at parturition, and on days 1, 14, 21 and 30 after parturition) than normal group. Cows with lameness presented higher (P < 0.05) values of body condition score on days -18, -12 and -8 before parturition, and lower (P < 0.05) values on days 7, 14, 21, 30, 45 and 60 after parturition than normal cows. Milk production was lower (P < 0.05) for cows with lameness (on days 7, 14, 21, 30, 45 and 60 after parturition) than normal cows.Discussion: The overall prevalence of lameness in the evaluated period was 22.92% (11/48). Among lame cows, 07 presented laminitis, 02 had interdigital hyperplasia and 02 had sole ulcer. Our findings prove that the biochemical profile and productivity of dairy cows during the peripartum and postpartum period were affected by lameness at the early lactation.

The aim of this study was to monitor the metabolic profile and oxidative status in goats during the peripartum period. A blood sample was taken from 10 Red Syrian goats on days −21, −3, +1, +14, and +28 from delivery. Samples were assayed for glutathione peroxidase and superoxide dismutase activities, and reactive oxygen metabolites, insulin, insulin-like growth factor-I, free triiodothyronine (fT3), free thyroxine (fT4), leptin, urea, non-esterified fatty acids, triglycerides, cholesterol, total protein, albumin, globulin, calcium and inorganic phosphorus concentrations. Blood glutathione peroxidase activity decreased during the postpartum period and its values were significantly (P < 0.05) lower on days 14 and 28 postpartum. Albumin levels were significantly (P < 0.05) lower on days −3, 1 and 28 from delivery compared with day −21. Plasma urea levels significantly (P < 0.001) decreased starting from day −3 from delivery. No effect of time from delivery was noted on reactive oxygen metabolites, non-esterified fatty acids, triglycerides, cholesterol, total protein, calcium and inorganic phosphorus, insulin and leptin concentrations and superoxide dismutase activity. Plasma concentrations of fT3 were significantly (P < 0.01) higher on days 14 and 28 compared with days −21, −3 and 1 from delivery. Plasma levels of fT4 were significantly (P < 0.01) lower on days −3 and 1 from delivery. Plasma insulin-like growth factor-I values slowly decreased during the postpartum period and its values were significantly (P < 0.05) lower on day 28. This study indicates that goats experienced moderate oxidative stress during the peripartum period.

SUMMARYThe blood composition of 45 cows with more than 20% fat in the liver (moderate fatty liver) was compared with that of 83 cows with less than 20% fat in the liver (mild fatty liver). The cows were sampled at 1 week after calving and were from three Friesian and one Guernsey herds. Blood concentrations of non-esterified fatty acids, bilirubin and aspartate amino-transferase were significantly elevated in cows with more than 20% in the liver whereas concentrations of glucose, cholesterol, albumin and magnesium were all significantly reduced. Blood samples from cows in two of the herds were analysed for ammonia, insulin and D(-)3-hydroxybutyrate. Cows with moderate fatty liver in these two herds had significantly lower insulin concentrations and higher D(-)3-hydroxybutyrate concentrations than those with mild fatty liver. Using an equation based on blood concentrations of non-esterified fatty acids, glucose and aspartate aminotransferase it was possible to assign correctly three out of four cows to either the mild or moderate fatty liver groups.

The relationships between changes in anti-Müllerian hormone (AMH) concentration and various traits, including milk somatic cell counts (SCC), were evaluated. Blood samples were collected from 43 Holstein cows 14 days before (D-14) and 10 (D10) and 28 days after (D28) parturition, and vaginal discharge score (VDS) and polymorphonuclear leukocyte (PMNL) percentages were assessed in endometrial samples at D28. Cows were separated into four quartiles (Q1–Q4) based on changes in AMH concentration during the peripartum period (AMH ratio: D28/D-14). Correlations between AMH ratio and each parameter were evaluated and classified into high-AMH (Q4, 1.83 ± 0.12, n = 11) and low-AMH (Q1, 0.83 ± 0.05, n = 11) groups. The AMH ratio was positively correlated with magnesium and non-esterified fatty acids levels, and the albumin/globulin ratio at D10 and D28, but negatively correlated with serum amyloid A (SAA) at D10. SAA and γ-globulin levels were significantly higher in the low-AMH group at D28. There was no significant difference in VDS, PMNL percentage, and milk SCC between the two groups. The decreasing AMH ratio from the prepartum to the postpartum period corresponds to high inflammation biomarker levels. Whether it subsequently affects the reproductive prognosis of postpartum cows needs further investigations.

This study was part of an experiment on the effects of cottonseed meal (CSM) and growth hormone-releasing factor (GRF) on growth in lambs fed lucerne chaff. Forty-eight crossbred lambs were fed lucerne chaff ad libitum, alone or with a cottonseed meal supplement (CSM; 300 g/day). Eight lambs from each group were injected twice daily with recombinant or synthetic GRF (rGRF and sGRF respectively; 30 8g per kg body weight/day) or excipient only for 28 or 30 days. Jugular blood samples were obtained on days 0, 7, 14, 21 and 28. On day 29, blood samples were taken from selected lambs (n = 2/group) at 0, 20, 40, 60, 80, 100, 120, 160 and 240 min after injection. Supplementation of lucerne chaff with CSM generally increased glucose and urea concentrations in plasma, reflecting improved energy and protein status. However, concentrations of growth hormone, insulin-like growth factor-1 and insulin were unaffected by CSM, as were plasma proteins, triacylglycerols and non-esterified fatty acids. Injection of rGRF and sGRF stimulated secretion of growth hormone (GH), insulin-like growth factor-1 (IGF-1) and insulin. Peak GH concentrations (20 min) following injection of GRF appeared to be higher in lambs fed CSM than in those receiving lucerne chaff alone. In contrast, maximal concentrations of IGF-1 were seen after 14 days of treatment. These changes were accompanied by increased plasma glucose, with no changes in triacylglycerols, non-esterified fatty acids and protein. Plasma urea was reduced by GRF, indicating that amino acids were diverted away from catabolism towards protein synthesis. These two sources of GRF were very similar in their endocrine and metabolic effects. This confirms similar observations regarding their effects on growth performance and carcass composition.

Determination of somatic cell count (SCC) is used worldwide in dairy practice to describe the hygienic status of the milk and the udder health of cows. When SCC is tested on a quarter level to detect single quarters with high SCC levels of cows for practical reasons, mostly foremilk samples after prestimulation (i.e. cleaning of the udder) are used. However, SCC is usually different in different milk fractions. Therefore, the goal of this study was the investigation of the use of foremilk samples for the estimation of total quarter SCC. A total of 378 milkings in 19 dairy cows were performed with a special milking device to drain quarter milk separately. Foremilk samples were taken after udder stimulation and before cluster attachment. SCC was measured in foremilk samples and in total quarter milk. Total quarter milk SCC could not be predicted precisely from foremilk SCC measurements. At relatively high foremilk SCC levels (>300×103 cells/ml) foremilk SCC were higher than total quarter milk. At around (50–300)×103 cells/ml foremilk and total quarter SCC did not differ considerably. Most interestingly, if foremilk SCC was lower than 50×103 cells/ml the total quarter SCC was higher than foremilk SCC. In addition, individual cows showed dramatic variations in foremilk SCC that were not very well related to total quarter milk SCC. In conclusion, foremilk samples are useful to detect high quarter milk SCC to recognize possibly infected quarters, only if precise cell counts are not required. However, foremilk samples can be deceptive if very low cell numbers are to be detected.
