The lipid and fatty acid profile of muscles in beaked redfish, caught and fixed in the wild versus specimens from food supermarkets (“commercial”), were evaluated, as well as the health implications of this popular food for its consumers based on the calculation of nutritional quality indexes. The contents of the total lipids (TLs), total phospholipids (PLs), monoacylglycerols (MAGs), diacylglycerols (DAGs), triacylglycerols (TAGs), cholesterol (Chol), Chol esters, non-esterified fatty acids (NEFAs), and wax esters were determined by HPTLC; the phosphatidylserine (PS), phosphatidylethanolamine (PE), phosphatidylinositol (PI), phosphatidylcholine (PC), and lysophosphatidylcholine (LysoPC) were determined by HPLC; and fatty acids of total lipids were determined using GC. The TL content was higher in commercial products due to DA and NEFAs, among PL fractions the content of LysoPC was also higher. The results indicated multidirectional processes of slight degradation of lipids in commercial products in comparison to wild. The flesh lipid quality index was lower due to EPA and DHA in commercial specimens while the index of thrombogenicity was significantly higher. The differences in the quantities of lipid classes between muscle biopsy regions in fish apparently corroborate the morphology and physiology of deep-water fish.

Background: Men and women with valvular heart disease have different risk profiles for clinical endpoints. Non-esterified fatty acids (NEFA) are possibly involved in cardio-metabolic disease. However, it is unclear whether NEFA concentrations are associated with physical performance in patients undergoing transcatheter aortic valve implantation (TAVI) and whether there are sex-specific effects. Methods: To test the hypothesis that NEFA concentration is associated with sex-specific physical performance, we prospectively analysed data from one hundred adult patients undergoing TAVI. NEFA concentrations, physical performance and anthropometric parameters were measured before and 6 and 12 months after TAVI. Physical performance was determined by a six-minute walking test (6-MWT) and self-reported weekly bicycle riding time. Results: Before TAVI, NEFA concentrations were higher in patients (44 women, 56 men) compared to the normal population. Median NEFA concentrations at 6 and 12 months after TAVI were within the reference range reported in the normal population in men but not women. Men but not women presented with an increased performance in the 6-MWT over time (p = 0.026, p = 0.142, respectively). Additionally, men showed an increased ability to ride a bicycle after TAVI compared to before TAVI (p = 0.034). NEFA concentrations before TAVI correlated with the 6-MWT before TAVI in women (Spearman’s rho −0.552; p = 0.001) but not in men (Spearman’s rho −0.007; p = 0.964). No association was found between NEFA concentrations and physical performance 6 and 12 months after TAVI. Conclusions: NEFA concentrations improved into the reference range in men but not women after TAVI. Men but not women have an increased physical performance after TAVI. No association between NEFA and physical performance was observed in men and women after TAVI.

Introduction. Vibration is one of the leading unfavourable physical factors of the working environment and leads to vibration disease (VD) development. The study of metabolic profiles of markers of fatty acids (FA) metabolism is one of the directions in the early diagnosis of vascular diseases and metabolic syndrome (MS), caused, among other things, by exposure to vibration. The aim of this work is to study the spectrum of esterified fatty acids of the omega-3 and omega-6 families in the blood of patients with vibration disease (VD) and combination with metabolic syndrome (MS). Materials and methods. The study involved 66 people, divided into two groups: Group I - 34 patients with an established diagnosis of VD; Group II - 32 patients with an established diagnosis of VD in combination with metabolic syndrome (MS). The levels of esterified forms (EF) of polyunsaturated fatty acids (PUFA) of the ω-3 and ω-6 families in blood plasma samples were determined using the method of gas chromatography-mass spectrometry on an Agilent 7890A gas chromatograph with an Agilent 5975C mass selective detector. Results. The distribution of quantitative indices of PUFA of the ω-3 and ω-6 families in the examined groups was studied. In VD patients with MS, compared with VD patients, there was a statistically significant increase in the level of eicosapentaenoic, α-linolenic acids of the ω-3 family, the sum of ω-3 PUFAs, γ-linolenic and linoleic acids of the ω-6 family. Conclusion. The study of the quantitative indices of EF of the families ω-3 and ω-6 in the blood plasma of VD patients with MS showed the levels of ω-6 to be 4.5-5.4 times statistically higher than the levels of ω-3 fatty acids.

The introduction of hyperprolific sows has resulted in more low birth weight (LBW) piglets, accompanied by higher mortality. A possible strategy to enhance the resilience and survival of LBW piglets is oral supplementation (drenching) of bioactive substances. This study evaluated the supplementation of bovine colostrum, short-chain fructo-oligosaccharides (scFOS) or quercetin that were dissolved separately in a milk replacer. The study was divided into two sub-experiments. First, the milk replacer was compared with a sham drenched group. Secondly, each dissolved compound was compared with the milk replacer. The LBW piglets, defined as weighing between (mean litter birth weight −1*SD) and (mean litter birth weight −2.5*SD), were randomly allocated to the different treatments and drenched once a day for seven days. On day 1, 3, 9, 24 and 38, piglets were weighed and scored for skin lesions. Blood samples were collected on day 9 and 38 and analyzed to determine glucose, non-esterified fatty acids, urea, immunoglobulin G, insulin-like growth factor 1, and a standard blood panel test. There was no difference between sham drenched piglets and piglets that were drenched with milk replacer regarding any of the parameters. No effect was observed between the milk replacer group and any of the bioactive compounds either, except a higher mortality within the scFOS group. In conclusion, this study showed that drenching the evaluated bioactive compounds, in the used dosages, did not improve LBW piglets’ resilience or survival and more research is required to determine the effect of scFOS on small piglets.

Abstract Metabolic syndrome increases risk of complicating co-morbidities. Current clinical indicators reflect established metabolic impairment, preventing earlier intervention strategies. Here we show that circulating sphingolipids are altered in the very early stages of insulin resistance development. The study involved 16 overweight but healthy, euglycemic monkeys, one-half of which spontaneously developed metabolic syndrome over the course of 2 years while the other half remained healthy. As previously reported, adiposity and glycemic status were equivalent across the cohort and did not explain divergent health outcomes. Using mass spectrometry, we detected and quantified circulating ceramides and other sphingolipids in healthy and impaired animals at both time points. Several ceramides were significantly different between healthy and impaired at time of diagnosis. Correlation analysis revealed differences in the interactions among ceramides in impaired animals at diagnosis and even pre-diagnosis, when animals were clinically indistinguishable from healthy controls. Importantly, correlations between ceramides and diacylglycerols and non-esterified fatty acids were distinct for healthy and impaired states, indicative of coordinated changes in lipid handling. Impaired animals displayed extensive differences in correlations among sphingolipids even in advance of loss of insulin sensitivity. These data suggest that circulating ceramides are clinically relevant in identifying disease risk and in devising preventative strategies.

The diet of dairy cows in Ireland traditionally changes abruptly from predominantly pasture silage before calving to grazed perennial ryegrass immediately after calving. This potentially leads to problems with adaptation of microbes in the rumen with consequences of reduced intake and ultimately lower milk production. This experiment aimed to determine if introducing first-lactation dairy cows to perennial ryegrass herbage in the final weeks of pregnancy, thus eliminating a major dietary change at calving, could improve the adaptation process, potentially increasing dry matter intake (DMI) and milk production in early lactation. Three weeks prior to their expected calving date, 14 spring calving dairy cows were assigned to one of two treatments (n = 7): pasture silage pre-partum and perennial ryegrass herbage post-partum, or perennial ryegrass herbage both pre- and post-partum. Treatment diets were fed for 11 (±7) d pre-partum and for 14 (±0) d post-partum. For both treatments, DMI increased post-partum, but there was no difference between treatments, pre- or post-partum (5.9 and 8.8 kg DM/cow per day, respectively). There were no differences in milk yield or composition between the treatments. Body condition score declined following parturition but there were no differences between treatments. Plasma non-esterified fatty acids, glucose and β-hydroxybutyrate were also unaffected by treatment but did indicate a state of negative energy balance in early lactation. The results of this experiment suggest that pre-partum adaptation to perennial ryegrass herbage would not benefit milk production in first-lactation dairy cows in early lactation in Irish dairy farms employing this system.

Abstract Background Non-esterified fatty acids (NEFAs) are one of the main lipid components of follicular fluid at concentrations that depend on circulating levels. Elevated levels of NEFAs impair oocyte quality, development potential, and may subsequently influence the metabolism and reproductive fitness of offspring. Granulosa cells (GCs) are the follicular cells that are closely communicating with the oocyte. However, the responses of GCs exposed to high levels of NEFAs when cocultured with cumulus-oocyte complexes (COCs), and how they attenuate the negative effects of NEFAs on oocytes, are unclear. Results To better understand this protective effect, monolayers of porcine GCs were cocultured with COCs during in vitro maturation (IVM) in the presence of elevated levels of NEFAs. Genomic expression analysis was conducted to explore the responses of the GCs to the elevated levels of NEFAs. After limma algorithm analysis, 1,013 genes were differentially expressed between GCs cultured with and without elevated NEFAs. Among them, 438 genes were upregulated and 575 were downregulated. The differentially expressed genes were enriched in pathways related to metabolism, inflammation, and epithelial-mesenchymal transition. Conclusions The pathways and upstream regulators suggested that the cocultured GCs responded to the elevated NEFAs with (1) inhibition of the transition from granulosa to luteal cell, (2) interactions of metabolism change, anti-inflammation, mitochondrial function, and cell transition, (3) intercommunication with cocultured COCs of anti-inflammatory factors.

Cows are affected by environmental factors associated with warm weather conditions; however, little is known about the effect of shade access especially during the prepartum period of dairy cows in temperate regions. This study assessed the effect of shade on the behavior (lying, rumination, feeding, and drinking), body fat mobilization, and health status of outdoor-housed dairy cows during the prepartum period under temperate summer conditions. During the 3 weeks prior to calving, 24 multiparous Holstein cows were grouped (4 cows/group) and assigned to either an open corral without shade or with access to shade until calving. We daily measured shade use, lying, rumination, feeding, and drinking behavior. Weekly, prepartum non-esterified fatty acids (NEFA) and postpartum b-hydroxybutyrate (BHB) concentrations were measured. Clinical examination was periodically performed individually until 21 postpartum days. Shade use averaged 45.6, 46.0, and 19.8% during the hottest hours of the day (11–18 h) in weeks 3, 2, and 1 prior to calving, respectively. Shaded cows had higher values for rumination time and feeding time during the morning but spent less time drinking during the warmest hours than unshaded cows. NEFA and BHB concentrations and clinical diseases were similar between both treatments. These findings suggest that under temperate summer conditions the access to an artificial shade is an important resource, observing beneficial effects mainly on behavioral variables.
