Phytochemical investigation of the stem bark of F. racemosa JACK ex WALL (Loganiaceae) from East Java, Indonesia, has resulted in the isolation of a new alkaloid fagraeoside along with the iridoid glycoside secologanoside. Fagraeoside may be derived from the condensation of secologanin with L-asparagine, and represents a rare example of a terpene alkaloid in which the amino acid component is non-aromatic. Investigation of three additional species of Fagraea provided known lignans, iridoid or secoiridoid glycosides, and flavanol-6-C-glucosides, thus it is likely that iridoid and secoiridoid glucosides are chemotaxonomic markers for the Fagraea genus. Fagraeoside inhibited the production of prostaglandin E2 in 3T3 murine fibroblasts (IC50 ~5.1 µM), and was not cytotoxic to this cell line or to a P388 murine leukaemia cell line. Selected isolated compounds, including fagraeoside, showed low to moderate activity in anti-acetylcholinesterase screening.

Phytochemical investigation of the methanolic extract of the stem bark of Casimiroa edulis afforded four coumarins. Various spectroscopic experiments were used to characterize the isolated coumarins. The structures were identified as auraptene (K-1), suberosin (K-2), 5-geranyloxypsoralen (bergamottin) (K-3), and 8-geranyloxypsoralen (K-4), based on the chemical and spectral analysis. Among these compounds, suberosin (K-2) and 5-geranyloxypsoralen (bergamottin) (K-3) were isolated for the first time from this genus, and auraptene (K-1) was isolated from this plant for the first time. Cytotoxicity of pure compound K-4 and sub-fraction MD-3 was evaluated against HeLa and T47D cell lines and moderate activity was found with an IC50 value in the range 17.4 to 72.33 μg/mL.

AbstractPhytochemical investigation of the stem bark of Diospyros longiflora yielded longiflorol (1), a new bergenin α-d-apioside, together with bergenin (2) and five known compounds: lupeol (S1), betulin (S2), betulinic acid (S3), stigmasterol (S4) and stigmasterol glucoside (S5). Their structures were determined by one-dimensional (1D) and 2D nuclear magnetic resonance experiments along with electrospray ionization high-resolution mass spectrometry and extended density-functional theory calculations of chiroptical properties. Longiflorol (1) and bergenin (2) were evaluated for their DPPH (2,2-diphenyl-1-picrylhydrazyl) antioxidant activity, with the crude extract for comparison and ascorbic acid as standard. The results showed that the extract and 2 had good antioxidant activity, whereas 1 showed only moderate activity at high concentration (>2 mg mL−1).

Phytochemical study on the stem bark of P. arborescens has isolated six secondary metabolites from the crude extracts by using column chromatography, while identification was performed by using Gas Chromatography - Mass Spectrometry (GC-MS), Nuclear Magnetic Resonance (NMR) and Fourier Transform Infrared (FTIR). The isolated metabolites were identified as caryophyllene oxide (1), α-bisabolol (2), benzamide 2-(methylamino) (3), 2-ethylpiperidine (4), piperine (5) and methyl eugenol (6). Toxicity test on the four crude extracts of P. arborescens shows a high cytotoxicity against Artemia salina brine shrimp with LC50 values ranging from 13.12 to 58.70 µg/mL. Greater cytotoxicity of the crude extracts of P. arborescens indicated the presence of potent cytotoxic components in this Piper spp. Antioxidant assay of P. arborescens against 2-diphenyl-1-picrylhydrazyl (DPPH) indicated moderate antioxidant activities of methanol, dichloromethane, chloroform and hexane crude extracts with EC50 values of 21.68, 23.82, 32.88 and 36.88 µg/mL, respectively.

We describe the cloning of a cDNA from the rat basophilic leukaemia cell line (RBL.2H3) encoding a novel member of the Rab family of small GTP binding proteins. The novel clone, which we call Rab8b, is most highly related to the Rab8 family with substantial divergence in the variable C-terminal domain. Northern blot analysis reveals highest levels of expression of Rab8b in the spleen, testis and brain, which is in marked contrast to the tissue distribution of Rab8. The Rab8b cDNA was modified to introduce a c-myc epitope tag at the extreme N terminus of the protein, and transient transfection studies were performed to analyse the intracellular localization of Rab8b by confocal microscopy. Transient expression of the c-myc/Rab8b fusion protein in both PC12 and RBL.2H3 cells shows staining of both the plasma membrane and ill-defined vesicular structures, and in the case of RBL.2H3 cells appears to induce striking outgrowths of the plasma membrane.
